Understanding The Relationship Between [Atp], Growth Rate, And Inoculum Effect In Β-Lactamase Producing Bacteria, Ariane R. Kalifa
All HCAS Student Capstones, Theses, and Dissertations
2.8 million Americans are infected with antibiotic-resistant bacteria on an annual basis, leading to increased mortality and morbidity. With the incidence of infection due to antibiotic-resistant bacteria on the rise, it is critical to understand the mechanisms bacteria use to resist antibiotics. The inoculum effect (IE) is a phenomenon that has been observed for many antibiotics and bacterial species. In IE, the density of the bacterial population determines the minimum inhibitory concentration of the antibiotic (MIC) of an antibiotic; as the density of the bacterial population increases, the MIC increases. IE is highly prevalent in bacteria that express β-lactamases where …

Fine-Tuning Gene Expression: A Novel Therapeutic Approach For Rett Syndrome, Yewon Rhee
Fine-Tuning Gene Expression: A Novel Therapeutic Approach For Rett Syndrome, Yewon Rhee
Master's Theses
Rett Syndrome (RTT) is a neurodevelopmental disorder that is characterized by a myriad of debilitating symptoms impacting nearly every aspect of life, including motor and gait impairment, social and cognitive deficits, as well as seizures and breathing abnormalities. RTT is caused by X-linked de novo loss-of-function mutations in the methyl-CpG binding protein 2 gene (MECP2), which is an essential epigenetic modulator. Any cure for RTT must directly target MECP2, a highly dosage-sensitive gene. This thesis developed a novel MECP2-targeted therapeutic approach using site-blocking antisense oligonucleotides to prevent microRNA-induced repression of MECP2, allowing fine-tuned control of gene expression. Here we report …

Building Trust: The Role Of Place-Based Consciousness In Vaccine Hesitancy, Ramsey W. Ash
Quantitative Social Science Undergraduate Senior Theses
Vaccine hesitancy poses a threat to public health initiatives of the 21st century. Following a stark divide in the rural/urban COVID-19 vaccine uptake and harsh disparities between these areas in COVID-19 moralities and morbidities, further understanding of the drivers of vaccine hesitancy are needed. This study makes use of the framework of place-based consciousness (PBC), pioneered by scholars of rural political science, such as Katherine Cramer, to explore this phenomenon as a mechanism by which to understand vaccine hesitancy. To examine this, an experimental survey was distributed to 300 Americans (150 rural and 150 urban) that inquired about their PBC, …

Transport Of The Fluorescent Substrate Asp+ By Organic Cation Transporter-3 In Human Corneal Epithelial Cells, Jeremy Laws
Theses & Dissertations
ABSTRACT
Organic cation transporter-3 (OCT3) is expressed abundantly throughout the body, but little is known about the presence of these transporters within the eye and, more specifically, the cornea epithelium. An extensive library of compounds interacts with OCT3, including the fluorescent molecule ASP+ and metformin. Metformin is an OCT3 substrate and antidiabetic medication that can lower the risk of eye-related diseases like keratopathy and retinopathy linked to hyperglycemia. Utilizing high throughput microplate assays in a human cornea epithelial cell line (HCE-S), the time-dependent saturation of ASP+ uptake and the competition of ASP+ uptake by corticosterone (CORT), a …
