This double blinded placebo controlled pilot study examined the use of combined inhaled medication for patients with moderate-to-severe COPD. The inhalation of formoterol versus placebo was studied when combined with the routine use of tiotropium. Maximizing bronchodilitation through the use of combination inhaler therapy is felt to be superior to the use of a single inhaled medication, yet few studies are found in the literature. Currently the Global Initiative for Obstructive Lung Disease (GOLD) recommends the use of combination inhaled therapy in patients with moderate-to-severe COPD. This study of 18 patients with moderate-to-severe COPD measured the effects on the functional …
Exploration Of The Cultural Beliefs In Mexican American Young Adult Childhood Cancer Survivors And Their Decisions To Engage In Cancer Screening Behaviors, Lisa Bashore
Nursing Dissertations - Archive
Gaps in the literature exist exploring the cultural beliefs and the cancer screening practices in Mexican American (MA) young adult childhood cancer survivors (YACCS). Lack of representation of MA in past survivor research is one reason for the gaps in knowledge about the beliefs in MA survivors, and the current study is the beginning of the examination of the cultural beliefs in MA YACCS and how these beliefs influence their decisions about cancer screening. Kleinman's (1978) cultural explanatory model (CEM) approach served as the conceptual framework in which this study was conducted. Survivors were recruited from the investigators institution where …

Identifying Influential Observations Through The Intraclass Correlation Coefficient, Angel De Jesus Davalos
Open Access Theses & Dissertations
In this thesis, we analyze the performance of adapting the DFBETA statistic for identifying influential observations on the intraclass correlation coefficient under the assumptions of the one-way random effects model. Additionally, we introduce an approach for transforming negative intraclass correlation coefficient estimation values using the method of moments estimator. We apply this method on a data set of repeated blood pressure measurements, after which we will investigate implications of identifying influential observations.
The Role Of Toll-Like Receptors 2 And 4 In Helicobacter Felis Infections And Asthma, Carolyn Durham
The Role Of Toll-Like Receptors 2 And 4 In Helicobacter Felis Infections And Asthma, Carolyn Durham

The inverse correlation between the industrialization and disease prevalence is termed the "hygiene hypothesis." Supporting this, immunological studies show Th1 cytokines modulate Th2 immune responses. Because the mucosa is essential in antigenic exposure, understanding bacterial sensing here is critical to addressing three foundations of this hypothesis: the role of bacterial exposure, relevance of noninvasive bacterial exposure, and influence of these exposures on immune responses. Key components of antigen sensing in the mucosa are toll-like receptors (TLRs) 2 and 4, which have been implicated in disease. The hypothesis of this thesis is, by globally knocking out one or both of these …

All ETDs from UAB
Porphyromonas gingivalis is a main causative agent for adult chronic periodontitis and immunization with its virulence factor Hemagglutinin B (HagB) provides protection against infection. Toll-like receptors (TLRs) recognize various microbial products and are crucial in eliciting and regulating the innate and adaptive immune responses to infections. The objective of this dissertation was to investigate the cellular mechanisms that influence the innate and adaptive immune response to HagB and P. gingivalis, focusing on the role of TLR signaling in the response. We started with investigating the ability of HagB to activate dendritic cells (DC), the most efficient antigen-presenting cell in priming …

Reproductive tract infections include sexually transmitted infections, endogenous infections caused by overgrowth of organisms that can be present in the genital tract of a healthy woman, and iatrogenic infections. In Jamaica, women in the reproductive age group of 15-44 years old accounts for 95% of all RTIs. RTIs among Jamaican women of reproductive age can seriously impact the health of the women and their children. The advent of HIV/AIDS has had a significant impact on increasing awareness of the effect of STIs. Health care organizations have reacted with great commit-ment to STI prevention and treatment as a significant factor in …
Regulation Of Redox Signaling By Lipid Electrophiles In Breast Cancer, Anne R. Diers
Regulation Of Redox Signaling By Lipid Electrophiles In Breast Cancer, Anne R. Diers
All ETDs from UAB
A number of steps in breast cancer progression and metastasis are regulated by redox signaling pathways. Electrophilic lipids such as 15-deoxy-delta12,14-Prostaglandin J2 (15d-PGJ2) are mediators of redox signaling pathways because of their ability to modify critical cysteine residues (thiols) in redox-sensitive proteins. In this thesis, we examine the effect of lipid electrophiles such as 15d-PGJ2 and others on redox signaling pathways in breast cancer. Furthermore, we develop new strategies to regulate cancer cell behavior in response to lipid electrophiles using three strategies: 1) through organelle-specific targeting of electrophiles 2) by exploiting the concentration-dependence of effects of electrophiles, and 3) utilizing …

Characterization Of The Oligosaccharides Of B. Anthracis Exosporium, Shengli Dong
All ETDs from UAB
Fatal systemic anthrax is caused by exposure to spores of Bacillus anthracis. The outermost layer of the B. anthracis spore is called the exosporium. It consists of a paracrystalline basal layer and an external hair-like nap. The filaments of the hair-like nap are primarily composed of the glycoprotein BclA. Our previous studies showed that a 715-Da tetrasaccharide and a 324-Da disaccharide are attached to BclA through GalNAc. We named the novel nonreducing terminal sugar of the 715-Da tetrasaccharide anthrose. We subsequently proposed a plausible anthrose biosynthetic pathway and identified a gene cluster of four continuous genes that appeared to encode …

All ETDs from UAB
The cataract is a common ailment affecting the aged population. It appears over time and affects the quality of one’s life by the eventual loss of vision. Currently, there is no effective strategy to prevent or treatment to reverse the development of a cataract. As well, there is no consensus on one particular mechanism or sequence of events that contributes to its formation. In order to better understand the possible etiology of the cataract, it is necessary to define the biochemical changes that occur to lens proteins as one ages, as these are the key players in the prevention of …
The Role Of Fc Receptor-Like 2 In B Cell Signaling, Tanisha Anne Jackson
The Role Of Fc Receptor-Like 2 In B Cell Signaling, Tanisha Anne Jackson
All ETDs from UAB
Fc receptor-like 2 (FCRL2) is a transmembrane protein with immunomodulatory potential that is preferentially expressed by memory B cells in humans. It has two consensus immunoreceptor tyrosine-based inhibitory motifs (ITIM) in addition to a putative immunoreceptor tyrosine-based activation motif (ITAM) sequence in its cytoplasmic domain. We have confirmed the cellular distribution of FCRL2 and ana-lyzed its functional potential to show that coligation with the B cell receptor (BCR) leads to tyrosine phosphorylation of its ITIM motifs and subsequent SHP-1 recruitment to facilitate inhibition of BCR signaling. Mutational analysis indicates that the tyrosine residues in both inhibitory motifs of FCRL2 are …
Epigenetics: Blurring The Line Between Nature And Nurture, Elizabeth H. Rose
Epigenetics: Blurring The Line Between Nature And Nurture, Elizabeth H. Rose
CMC Senior Theses
This long-standing nature versus nurture debate is cited in behavioral and physical expressions of disease dysfunctions, resiliencies, and recovery. Their purposes are noted both in scientific pursuits as well as literature. This discourse has been particularly intense in the fields of psychology, psychiatry, and biology where there is a long history of scientists’ attempts to disprove or discredit others’ intellectual and professional measures. Interestingly, recent advances in the neurosciences and genetic technologies have brought these fields closer together with a new focus – the interactional relationship between nature and nurture – epigenetics.
The Radiosensitization Effect Of Parthenolide In Prostate Cancer: Implications For Selective Cancer Killing By Modulation Of Intracellular Redox State, Yulan Sun
University of Kentucky Doctoral Dissertations
Parthenolide (PN), a major active component of the traditional herbal medicine feverfew, has been shown to have anti-inflammatory and anti-tumor properties. More remarkably, the cytotoxicity of PN seems selective to tumor cells but not their normal cell counterparts. In the present study, we investigate whether and how PN selectively enhances tumor sensitivity to radiation therapy by using prostate cancer cells LNCaP, DU145 and PC3, as well as normal prostate epithelial cells PrEC.
Our study demonstrates that inhibition of NF-κB pathway and suppression of its downstream target MnSOD are common mechanisms for the radiosensitization effect of PN in prostate cancer cells. …

Modulation Of The Alpha-7 Nicotinic Acetylcholine Receptor Following Experimental Rat Brain Injury Improves Cellular And Behavioral Outcomes, Thomas Matt Woodcock
University of Kentucky Doctoral Dissertations
Traumatic brain injury (TBI) is a leading cause of death and long-term disability worldwide, and survivors are often left with cognitive deficits and significant problems with day to day tasks. To date, therapeutic pharmacological treatments of TBI remain elusive despite numerous clinical trials. An improved understanding of the molecular and cellular response to injury may help guide future treatment strategies. One promising marker for brain injury is the translocator protein (TSPO), which is normally expressed at a low level, but is highly expressed following brain damage and is associated with neuroinflammation. The isoquinoline carboxamide PK11195 binds selectively to the TSPO …
