IMPORTANCE: The most common inherited blood disorder in the world, sickle cell disease (SCD) is a known global health concern. While survival of children into adulthood has improved significantly over the last few decades in high-resource countries, substantial gaps in care standards and limited practice harmonization persist.
OBJECTIVE: To provide up-to-date recommendations for preventive care and treatment of all people living with SCD and its related comorbidities.

BACKGROUND: Comprehensive, multidisciplinary care is crucial for managing early-stage non-small cell lung cancer (NSCLC), typically treated via lung resection or stereotactic body radiation therapy (SBRT). We evaluated how variable access to on-site SBRT may be associated with clinical outcomes for patients with stage I NSCLC receiving surgical resection within an integrated health-care system.
METHODS: We performed a retrospective cohort study of patients with stage I NSCLC treated with lung resection at Veterans Health Administration (VHA) facilities between 2006 and 2016. Cantu syndrome (CS) is a rare disease caused by gain-of-function (GOF) mutations of Kir6.1 or SUR2 subunits of ATP-sensitive potassium (KATP) channels. CS patients with SUR2 and Kir6.1 variants display a similar constellation of symptoms, including muscle weakness and fatigue. The effects of CS mutations on skeletal muscle KATP channels, and any consequent direct effects on contractility, are currently unclear. Here, we used two knock-in mouse models of CS, respectively, carrying GOF mutations Kir6.1[V65M] or SUR2[A478V], to assess KATP channel properties and contractility in isolated fast-twitch extensor digitorum longus (EDL) and slow-twitch soleus (SOL) muscles. Inwardly rectifying potassium (Kir) channel activity is important in the control of membrane potentials in both excitable and non-excitable cells and is regulated through various ligands, including specific membrane lipids. Phosphatidyl-4,5-bisphosphate (PIP2) is required for activity of all Kir channels, binding to the cytoplasmic domain in a compact conformation tightly tethered to the transmembrane domain. Most Kir2 channel structures determined in complex with PIP2 molecules are nevertheless in a closed state, requiring additional conformational changes for channel opening. Mutations that initiate acute myeloid leukemia (AML) can cause clonal expansion without transformation (clonal hematopoiesis). Cooperating mutations, usually in signaling genes, are needed to cause overt disease, but these may require a specific fitness state to be tolerated. Here, we show that nearly all AMLs arising in a mouse model expressing 2 common AML-initiating mutations (Dnmt3aR878H and Npm1cA) acquired a single copy amplification of chromosome 7 (chr7), followed by activating mutations in signaling genes. We show that overexpression of a single gene on chr7 (Gab2, which coordinates signaling pathways) was tolerated in the presence of the Npm1cA mutation, could accelerate …

Extracranial metastasis of IDH-wildtype glioblastoma is very rare and poorly understood at the molecular level. We report a case of FGFR3::TACC3 fusion IDH-wildtype glioblastoma in a 61-year-old male, whose preoperative blood sample showed highly aberrant cfDNA fragmentation patterns, which could be suggestive of early systemic dissemination, undetected by standard-of-care imaging of his body. Eleven months post-resection and adjuvant therapy, he developed widespread extracranial metastases. Comprehensive molecular profiling of matched primary and metastatic tumors revealed broadly conserved genomic, transcriptomic, and copy number landscapes, with the metastasis harboring an additional ERCC6 deletion and enriched expression of receptor tyrosine kinase signaling genes. BACKGROUND: Nigeria is experiencing a growing burden of diet-related non-communicable diseases, particularly hypertension and cardiovascular disease, largely driven by high sodium intake and poor diet quality. Unhealthy dietary patterns characterized by excessive sodium, sugars, and unhealthy fats, and low intake of essential nutrients contribute to approximately 31% of global deaths annually. However, there is limited data on the sodium content and nutritional quality of foods in Nigerian markets. BACKGROUND: In Nigeria, cardiovascular diseases, especially hypertension, are on the rise. This increase in hypertension correlates with more strokes, significantly impacting mortality. Since hypertension often persists into adulthood, early interventions are crucial to prevent its complications. Non-invasive methods, such as music and creative activities, can effectively improve blood pressure and reduce stroke risk. This study aims to improve intergenerational awareness of hypertension and promote sustainable preventive practices by involving youth and caregivers within families and communities.

Background: Falls are the leading cause of injury-related death in adults over age 65 (CDC, 2024). Traditional exercise-based interventions may have an impact on fall prevention (US Preventive Services Task Force et al., 2024). Providing these programs is resource-intensive, however mHealth applications may expand comprehensive fall prevention. Background: Peer influence is central to adolescent smoking initiation, yet its impact varies depending on individual and contextual factors. Understanding which moderators (personality, contextual, cultural, and environmental traits) shape these processes can inform more effective prevention strategies. We investigated hypothesized moderators of peer influence for adolescent smoking/vaping norms and other smoking-related outcomes in high and low-middle-income countries (LMICs): Northern Ireland and Bogotá.

Animal venom, known for its complex biochemical composition, presents a valuable source of therapeutic molecules, particularly for antiviral applications. Despite this potential, the industrial use of venom remains limited, with fewer than a dozen venom-derived compounds reaching commercial markets. This study underscores the significance of exploring venom’s natural diversity as a reservoir for novel bioactive compounds that could drive innovative drug development. We investigated the venom of the Moroccan black scorpion Androctonus mauritanicus (Am), applying solid-phase extraction (SPE) and high-performance reversed-phase liquid chromatography (RP-HPLC) to fractionate the venom into 80 distinct samples. KRAS mutations are implicated in approximately 23% of all human malignancies, with particularly high prevalence in pancreatic ductal adenocarcinoma (PDAC) (~92%), colorectal cancer (CRC) (~49%), and non-small cell lung cancer (NSCLC) (~35%). The recent approval of the KRASG12C-specific inhibitors for NSCLC represents a pivotal advancement in KRAS-targeted therapy. Nevertheless, the emergence of intrinsic and acquired resistance to KRAS-targeted therapies poses a significant clinical obstacle to targeting KRAS, which necessitates a deeper understanding of the resistance mechanisms. Insecticide Resistance Profile In Nashville Culex Spp. Mosquito Populations, Ellie Sebaugh, Zachary Bement, Abelardo Moncayo, Darlene Panvini

Mosquitoes may develop resistance to insecticides used by control programs, potentially increasing population density and the risk of mosquito-borne diseases. Culex spp. mosquitos are the primary vector of West Nile Virus (WNV), the leading cause of mosquito-borne disease in the United States and a reportable disease in Tennessee. Resistance profiles were constructed for two insecticides, permethrin and malathion, in a Culex spp. population from Nashville, Tennessee. Adult female mosquitoes were live-captured using gravid traps and tested using the Centers of Disease Control and Prevention (CDC) bioassay protocol to assess resistance or susceptibility. Many musculoskeletal imaging modalities are considered the cornerstone of sports medicine, though the diagnostic yield is different across each modality. Radiographs often detect abnormalities in fewer than half of cases, with the MURA dataset reporting only 39% of abnormal extremity films (Rajpurkar et al., 2018). In contrast, musculoskeletal ultrasound has been shown to identify pathology in up to 83% of examinations (Chiavaras et al., 2008). Point-of-care ultrasound (POCUS) further enhances rapid bedside diagnosis, with emergency department studies showing pathology in approximately 79% of cases (Situ-LaCasse et al., 2018). This systematic review synthesized evidence on the cost-effectiveness and economic favorability of interventions for the detection, treatment, and prevention of perinatal mood and anxiety disorders (PMADs).Methods
This protocol is registered on PROSPERO (CRD42024528777). PubMed, EMBASE, Web of Science, EconLit, Scopus, PsycINFO, and the National Health Service economic evaluation databases were searched from inception through April 2025. Intervention studies reporting quality-adjusted life years, incremental cost-effectiveness ratios, or equivalent cost analyses during pregnancy or up to one year postpartum were included. Colorectal cancer progression is driven in part by dysregulated signaling pathways that alter cell growth and migration. One key pathway is the Wnt signaling pathway, a key regulator in colorectal tumorigenesis, which promotes MYC overexpression, leading to uncontrolled proliferation. Interestingly, in the absence of p53, MYC has been found to induce expression of the tumor suppressor gene EGR1, shifting the cellular response toward apoptosis rather than growth. This study investigates the MYC–EGR1 pathway in HCT116 colorectal cancer cells with either wild-type (p53+/+) or knockout of the TP53 gene (p53−/−). Gut microbiota have been implicated in severe malaria in murine models, but their contribution to the pathogenesis of severe malaria in children is unknown. Here we show through analysis of gut bacteria in stool samples from two separate African studies enrolling children with severe malaria, and children from local communities, that children with severe malaria have gut bacteria dysbiosis. Among children with severe malaria, there is increased abundance of Enterobacteriaceae that associates with multiple clinical complications of severe malaria. Background: Implementing small and sick newborn care (SSNC) requires skilled health workers; however, there is a shortage, adversely impacting patient outcomes and health worker wellbeing. In this case, a 67-year-old male experienced a progression of his Parkinson’s disease symptoms following an inpatient hospitalization for pneumonia. He presents to the neurology clinic for follow-up, where the neurology ambulatory care pharmacist was consulted to assist with his case. The pharmacist identified a medication reconciliation discrepancy that resulted in an unintentional change in the dosing frequency of his carbidopa/levodopa. This, combined with the initiation of metoclopramide, a dopamine-receptor antagonist, led to a worsening of his Parkinson’s symptoms. His carbidopa/levodopa dose was titrated back to the previous regimen, metoclopramide was discontinued, and his symptoms significantly improved. This case highlights …

BACKGROUND: Of patients who receive standard concomitant chemoradiation (CRT; intensity-modulated radiation therapy [IMRT] plus cisplatin) for locally advanced head and neck cancer (HNC), approximately two-thirds will develop severe oral mucositis (SOM), limiting their ability to eat solids (WHO grade 3) or drink liquids (WHO grade 4). In a randomized, double-blind phase 2b trial, avasopasem manganese substantially reduced duration and incidence of SOM versus placebo. This phase 3 trial further assessed avasopasem's reduction of SOM due to CRT.

Rationale
Opioid-related overdose deaths involving benzodiazepines have increased in recent years, and prior studies have reported that clinically used benzodiazepines can enhance the respiratory-depressant effects of mu opioid receptor (MOR) agonists. TPA023B is an α1-sparing GABAA positive allosteric modulator developed as a potential anxiolytic with fewer benzodiazepine-typical side effects. However, it is unknown if and to what degree TPA023B can affect MOR-agonist induced respiratory depression.
Objectives
The current study compared the effects of either TPA023B or midazolam, alone and combined with fentanyl, on respiratory depression in rats, using whole-body plethysmography.
