The fibrinogen receptor on human platelets is a prototypic member of the integrin family and is composed of subunit glycoproteins IIb (gpIIb) and IIIa (gpIIIa) in a 1:1 stoichiometric ratio. We have isolated cDNA clones for gpIIb and gpIIIa and localized both genes to chromosome 17. In the current study, several approaches were used to localize and map the genes for gpIIb and gpIIIa. A preliminary evaluation of subchromosomal localization was performed by using a panel of mouse-human somatic cell hybrids that contain different amounts of the long arm of human chromosome 17. Patterns Of Polyadenylation Site Selection In Gene Constructs Containing Multiple Polyadenylation Signals., Roger Denome, Charles Cole

We have constructed a series of plasmids containing multiple polyadenylation signals downstream of the herpes simplex virus type 1 (HSV) thymidine kinase (tk)-coding region. The signals used were from the simian virus 40 (SV40) late gene, the HSV tk gene, and an AATAAA-containing segment of the SV40 early region. This last fragment signals polyadenylation poorly in our constructs and not at all during SV40 infection. All plasmids contained the SV40 origin of replication. Plasmids were transfected into Cos-1 cells; after 48 h, cytoplasmic RNA was isolated and the quantity and 3'-end structure of tk mRNAs was analyzed by using S1 …

The interpretation of data on genetic variation with regard to the relative roles of different evolutionary factors that produce and maintain genetic variation depends critically on our assumptions concerning effective population size and the level of migration between neighboring populations. In humans, recent population growth and movements of specific ethnic groups across wide geographic areas mean that any theory based on assumptions of constant population size and absence of substructure is generally untenable. We examine the effects of population subdivision on the pattern of protein genetic variation in a total sample drawn from an artificial agglomerate of 12 tribal populations …

Control of growth and differentiation during mammalian embryogenesis may be regulated by growth factors from embryonic or maternal sources. With the use of single-cell messenger RNA phenotyping, the simultaneous expression of growth factor transcripts in single or small numbers of preimplantation mouse embryos was examined. Transcripts for platelet-derived growth factor A chain (PDGF-A), trans- forming growth factor (TGF)-a, and TGF- 1, but not for four other growth factors, were found in whole blastocysts. TGF-cx, TGF-(31, and PDGF antigens were detected in blastocysts by immunocytochemistry. Both PDGF-A and TGF-a were detected as maternal transcripts in the unferilized ovulated oocyte, and again …

Adolescent Smoking Status And Socioeconomic Dependence On The Tobacco Crop In Southeastern Kentucky, Gregory Colwell

The purpose of the study was to quantify the relationship between adolescent smoking and economic dependence on the tobacco crop. A survey of 1322 students from a random sample of all secondary schools in four counties in southeastern Kentucky was performed. The survey gathered information concerning smoking behavior and social variables. Chi-square analysis was performed to assess the relationships between these variables in students from families who worked with tobacco and families who did not.
Chi square analysis of female adolescent smoking behavior revealed a significant association between the instances of adolescent smoking and age, friend smoking status and sibling …

Previous characterization of GLI, a gene found to be amplified and expressed in a subset of human brain tumors, revealed the presence of five tandem zinc fingers related to those of Krüppel (Kr), a Drosophila segmentation gene of the gap class. We have used the GLI cDNA as a molecular probe to isolate related sequences from the human genome. Partial characterization of six related loci, including sequence determination, expression studies, and chromosome localization, revealed that each locus could encode a separate finger protein. The mechanism underlying the ability of insulin to acutely activate acetyl-CoA carboxylase [acetyl-CoA: carbon-dioxide ligase (ADP-forming), EC 6.4.1.2; AcCoA-Case] has been examined in Fao Reuber hepatoma cells. Insulin promotes the rapid activation of AcCoACase, as measured in cell lysates, and this stimulation persists to the same degree after isolation of AcCoACase by avidin-Sepharose chromatography. The insulin-stimulated enzyme, as compared with control enzyme, exhibits an increase in both citrate-independent and -dependent activity and a decrease in the Ka for citrate. Direct examination of the phosphorylation state of isolated 32P-labeled AcCoACase after insulin exposure reveals a marked decrease in total enzyme phosphorylation …

T lymphocytes are stimulated to proliferate in an autocrine/paracrine manner by the lymphokine interleukin 2 (IL-2). In seeking further insight into the mechanisms by which IL-2 induces progression of T cells through the G1 phase of the cell cycle, studies were performed with agents that increase cellular adenosine 3',5'-cyclic monophosphate (cAMP), a well-known inhibitor of lymphocyte growth. The addition of dibutyryl-cAMP, cholera toxin, forskolin, or 3-isobutyl-1-methylxanthine to an IL-2-dependent murine T-cell line evoked a dose-related suppression of S-phase transition without affecting cellular viability. Moreover, elevation of cAMP levels led to an accumulation of uniformly small cells, suggesting an arrest in …

Pharmacology and Nutritional Sciences Faculty Patents
Compounds including phloretin and/or phlorizin derivatives coupled to an undigestible and non-absorbable matrix are provided. The compounds have pharmacological efficacies. They inhibit sugar uptake from the intestine and may be used in composition form in methods for treating diabetes and for promoting weight loss. A method for the manufacture of these compounds and a novel intermediate phlorizin derivative for making these compounds are also disclosed.

The diets of juvenile (mm) Syngnathus scovelli and of Hippocampus zosterae, abundant members of a resident fish community in a Thalassia testudnum seagrass bed in Tampa Bay, Florida, were examined from Apnl to October 1984. Harpacticoid copepods comprised most of the diet, both in terms of percent number and percent biomass, for the smaller size classes of S. scovelli and for H. zosterae, and harpacticoids generally had the highest index of relative importance (IRI) for both syngnathids. S. scovelli displayed ontogenetic switching to larger food items, such as amphipods, shrimp and crustacean eggs. Harpacticus sp. 1 was the …

A variety of magazines are directed at American women. Some emphasize fashion, while others cover child rearing, homemaking, or gardening; still others stress career, coping with multiple women's roles, or feminism. Almost without exception, women's health concerns are periodically addressed by these magazines. Because women are the target audience, I have always assumed that these publications have women's best interests at heart. Is this a realistic assumption? I'm not sure. I also do not know what influence women's magazines have on women's health behavior, but I think it is substantial. If this is so, then shouldn't we hold these publications …
