Purification, Characterization And Distribution Of A Calmodulin-Dependent Protein Kinase From Rat Brain Cytosol, James Richard Goldenring

Tubulin is a major endogenous substrate for Ca('2+)- calmodulin-dependent kinase activity in rat brain cytosol. This dissertation details the purification and characterization of a calmodulin-dependent tubulin kinase from rat brain cytosol. The kinase, purified from chelated brain cytosol by sequential chromatography on phosphocellulose, calmodulin-affinity resin, and Fractogel TSK-55F, contains two calmodulin-binding, autophosphorylating subunits of 52,000 and 63,000 daltons, designated rho and sigma, respectively. The kinase holoenzyme complex has a native molecular weight of 600,000 daltons. Both kinase subunits demonstrate isoelectric points near neutrality. Solid neoplasms are known to contain deficient or poorly functional vascular beds and as a result populations of cells in these solid tumors may contain hypoxic or poorly oxygenated tumor cells. These hypoxic tumor cells may form a therapeutically resistant cell population within the tumor difficult to eradicate by ionizing radiation and most existing chemotherapeutic agents. As a consequence, this dissertation has investigated the mechanism of bioactivation and cytotoxicity of mitomycin C and porfiromycin, structurally similar antibiotics which are selectively cytotoxic to hypoxic cells.Mitomycin C was preferentially cytotoxic to hypoxic EMT6 and V79 cells in culture, but was not selectively …

Chemiosmotic Properties Of Isolated Secretory Granules From Parotid Glands Of Normal And Isoproterenol Treated Rats, Peter Raymond Arvan

Secretory granule fractions were prepared isoosmotically from the parotid glands of normal rats and from rats that had received repeated injections of isoproterenol. These granules, purified extensively, are stable after isolation, making it possible to study biophysical parameters which may relate to their role in intracellular transport and secretion.Normal parotid granules have an internal pH of (TURN)6.8 and a low ionic permeability with respect to internal buffering capacity. The studies described in this dissertation demonstrate that basement membrane may be required for maintenance of organized epithelial tissue architecture. The structural organization of normal rat pancreatic acinar epithelium is fully characterized in order to analyze its neoplastic disorganization within a pancreatic acinar cell tumor. Transmission electron microscopy and indirect immunofluorescence microscopy of frozen semithin tissue sections are utilized to localize different components of the plasmalemma, cytoskeleton, basement membrane and connective tissue. Normal acinar cells sit on a continuous basement membrane and display a polarized distribution of intracellular organelles, cytoskeletal elements, and distinct membrane domains while these organized cell relations …

In the late 1960's, a new method for advanced airway management in the pre-hospital setting, the esophageal obturator airway (EOA), was introduced. Ever since, it has been the subject of much discussion over whether it was an effective replacement or alternative to the device traditionally used for airway protection, the endotracheal (ET) tube. Much of the comment has been in the form of personal opinion and anecdotal reports. The clinical potential of specific passive mucosal immunity is discussed and it is proposed that bulk quantities of hybridoma produced specific dimeric IgAs could be clinically useful as made to order super colostrums. SIgAs function on mucosal surfaces to prevent the adherence of pathogens. They are poor opsonins and do not activate complement except by the alternate pathway. Angiographic Patterns Of Arteriosclerosis Obliterans In The Legs Of Diabetics And Nondiabetics : Review Of The Literature And The Recent Experience At Yale-New Haven Hospital, Aron Wahrman

The literature regarding angiographic patterns of arteriosclerosis obliterans (ASO) in the legs of diabetics and nondiabetics is reviewed. The medical records of 28 adult-onset diabetics and 35 nondiabetics with chronic ASO of the lower extremities who underwent angiography for the first time were analyzed for age, presenting symptoms, smoking history, history of hypertension and therapeutic fate of the affected limbs. The arteriograms were analyzed for basic disease patterns and degree of disease at specific segments of the arterial tree, based on the radiologist's findings. Diabetics have more popliteal and trifurcation disease than nondiabetics, but less than has been found in …
