Orphan Drug Development: Incentives Under The Orphan Drug Act, Sara H D Smith

After describing the intellectual property and regulatory environment for orphan drugs in the United States, this thesis compares the investment decisions in the orphan drug market with the larger pharmaceutical industry. A series of case studies trace the development paths of different orphan products using information collected through the Food and Drug Administration (FDA) Approved Drug Product and Orphan Drug Product Designation Databases. In addition to this analysis, difference-in-differences estimates calculated using annual revenues compare the relative success of different orphan products under the current incentive system. This study finds that partial orphan drugs are associated with larger revenue growth. …

Autism spectrum disorder (ASD) is a neurological disorder characterized by decreased sociability, deficits in communication, and restricted and repetitive behaviors. The ketogenic diet (KD), a high-fat, low-carbohydrate, and moderate-protein diet has been shown to improve these three behavioral symptoms in the BTBR mouse model of autism. However, further research is required to strengthen the body of knowledge surrounding the potential of KD as diet therapy for autism. Epidemiological observations have shown that maternal immune activation (MIA) during pregnancy increases the risk of autism in offspring. Acquired Brain Injury (ABI) can result from internal factors (e.g. tumor) or external causes (e.g. trauma). Three case studies of Korsakoff’s syndrome, meningioma, and traumatic brain injury (TBI), will be presented to illustrate the variety of cognitive deficits across different individuals with ABI. This study uses cognitive rehabilitation therapies to target individual cognitive symptoms. Results and discussion place emphasis on the use of prospective memory (PM) training for treating Korsakoff’s syndrome. The Memory for Intentions Screening Test (MIST) served as the assessment for PM analysis pre and post rehabilitation. The data from this study will be used as a model …

Society today puts extreme pressure on individuals to take on a magnitude of daily responsibilities in order to be successful. We are pushed beyond our limits both mentally and physically, which results in heightened state and trait anxiety. Adopting the theory of mindfulness teaches us to live in the present moment and not criticize our thoughts (Bormann et. al, 2013). The current study tests the effects of daily mindfulness meditation on state and trait anxiety as measured by the State-Trait Anxiety Inventory. National statistics reveal a startling trend concerning ecstasy use among high school students, with over 5% of 10th graders and 8% of 12th graders reporting lifetime use (Dennis & Ballard, 2002). Ecstasy use among college students is even higher, with some studies reporting rates up to 10% (Boyd et al., 2003). In a recent survey of 18-35 year olds, 15% reported using ecstasy (Businelle et al., 2009) and many emerging adults viewed it as “safer” than other illicit drugs, with limited negative consequences (Bahora et al., 2009). Although numerous quantitative studies have explored the topic of ecstasy use in college students, there is limited qualitative research, most of which is limited to users. Thus, in the current study, we used a focus group methodology to better understand users’ and nonusers’ knowledge, expectations, and perceived risks of ecstasy use, for the purpose of informing prevention efforts on college campuses. The study of β-sheet mimetics represents an important step in understanding the structural and function of β-sheets. The purpose of this report was to develop a β-sheet mimetic through a diphenylacetylene backbone, coordinate the complex to tungsten and determine the structural and conformational characteristics of the complex. In order to characterize the different compounds ESI-MS, 1HNMR, COSY NMR, NOESY NMR, and ROESY NMR were employed. The diphenylacetylene backbone contained two phenyl substituents with a peptide chain of two amino acids. This work investigated the effects of the ketogenic diet on catecholamine concentrations in the brains of mice. The ketogenic diet has been show to modulate the catecholamine concentrations in the cerebrospinal fluid of children as well as altering hippocampal norepinephrine levels in mice. Tissue homogenates of the motor cortex, somatosensory cortex, nucleus accumbens, anterior caudate, posterior caudate and midbrain regions were analyzed using high pressure liquid chromatography to quantitate norepinephrine, dopamine, 3, 4-dihydroxyphenylacetic acid (DOPAC), 5-hydroxyindoleacetic acid (5HIAA), homovanillic acid (HVA) and serotonin (5-HT). The human body is colonized with commensal microbes that attach to the skin and mucosal membranes and function to protect the host as part of the innate immune response. These indigenous microbiota are able to prevent infections by blocking attachment sites and outcompeting invading pathogens for necessary nutrients. Previous research within the Curran group has demonstrated that ferrocene may be used as an organometallic constraint to induce the formation of α-helices in short peptides which traditionally possess undefined conformations. Through strategic placement of lysine residues at the i and i+3 positions within the peptide, such a constraint was accomplished via the crosslinking of the lysine side chains to ferrocene dicarboxylic acid chloride in solution phase synthesis.
