Since its discovery in 1922, insulin has been the life-saving treatment for type 1 diabetes mellitus. As the disease is caused by the loss of insulin-producing pancreatic islets, transplantation of donor islets has the potential to not only supplement insulin replacement therapy but to cure type 1 diabetes. However, long-term insulin independence (> 2 years post-transplant) remains a challenge partly due to low islet blood flow immediately following transplantation leading to hypoxic stress on islets. The persistence of Porphyromonas gingivalis in the inflammatory environment of the periodontal pocket requires an ability to overcome oxidative stress. DNA damage is a major consequence of oxidative stress. Unlike other organisms, a non-base excision repair mechanism for the removal of 8-oxo-7,8-dihydroguanine (8-oxoG) in P. gingivalis was suggested. Because the uvrB gene is known to be important in nucleotide excision repair, the role of this gene in the repair of oxidative stress-induced DNA damage was investigated. A 3.1 kb fragment containing the uvrB gene was PCR-amplified from the chromosomal DNA of P. gingivalis W83. This gene was insertionally inactivated using …

Osteoporosis is a disease characterized by decreased bone mass and microarchitectural deterioration of bone. Bone loss occurs when the increased bone resorption that occurs with age is not compensated by a corresponding increase in bone formation. Therefore, understanding the mechanisms which regulate bone formation and bone resorption is important for developing diagnostic and treatment strategies for osteoporosis. In my dissertation, we investigated the role of Glutaredoxin 5 (Grx5), Diabetes and Obesity Related protein (DOR), and Claudin 18 (Cldn-18), novel molecules involved in regulating bone formation and bone resorption. Adipose tissue secretes the adipokine, adiponectin (ADPN), which is insulinsensitizing, anti-inflammatory, and anti-atherogenic. Exercise training improves insulin sensitivity and lowers the risk of cardiovascular complications. As some of the metabolic effects of exercise training and ADPN overlap, exercise training has been proposed to increase ADPN. However, most single bout exercise, or short-term (ï‚£3 months) and constant-effort (fixed session duration, fixed number of sessions/week, and fixed intensity) exercise protocols do not produce increases in ADPN in untrained and trained cohorts. Type 1 diabetes mellitus is a chronic inflammatory disease in which the insulin and glutamic acid decarboxylase producing β-cells of the pancreatic islets are killed by autoreactive cells of the immune system in response to a loss of tolerance. Early experiments showed that self-tolerance could be partially restored by oral inoculation with small amounts of insulin. Later experiments showed that insulin autoantigen induced immune tolerance could be greatly enhanced by linkage of β-cell autoantigens (Auto Ag) to adjuvant molecules such as the non-toxic cholera enterotoxin B subunit (CTB). The purpose of this study was to establish a foundation for developing an evidence-based assessment guideline to be used by nursing and medical personnel when evaluating a firefighter's ability to return to work after a lower extremity injury. Two on-line survey instruments were used to record the opinions and beliefs of healthcare providers and firefighters. The final samples included 63 California healthcare providers (with and without professional work experience with firefighters) and 312 California firefighters. Introduction: The purpose of this study is to explore what happens within families in which a child participates in a pediatric weight management program for the treatment of overweight or obesity and identify strategies and processes for successful weight management.

Method: A qualitative approach was used to collect data from seven families. Individual interviews were conducted with parents, target children, and siblings. Data was analyzed within families and compared between families.

Results: Grounded theory analysis revealed three major essential themes: Shared Burden, Family Congruence, and Collaboration. Descriptive analysis was used to identify recommendations for improving family based weight management programs. …

Chronic pain is the most common cause of disability in the United States with depression, anxiety, and stress often occurring co-morbidly. The purpose of the study was to explore how religion may modulate the frequency of back pain, influence inflammatory markers, influence the use of medications, and impact functional ability.

Participants were from the Biological Manifestations of Religion Study (BioMRS; N= 508) and the Psychological Manifestations of Religion Study (PsyMRS; N = 10,988). In the United States, obesity rates have climbed steadily over the years. Many different diets for weight loss have been proposed and studied, yet no one diet seems to clearly be more beneficial. Physical inactivity is a major public health concern in the United States and is no less of a concern among college students. According to the 2000 National College Health Assessment, as cited by Kilpatrick, Herbert, and Bartholomew (2005), only 38% of college students participate in regular vigorous activity, and only 20% participate in regular moderate activity. Background and Purpose. Breast cancer is the second leading cause of cancer deaths among Hispanic/Latina women who are one of the fastest growing ethnic groups in the United States. Although Hispanic women have a lower incidence of breast cancer when compared to non-Hispanic White women, they are more likely to develop breast cancer at an advanced stage. In California, Hispanic women have the lowest rate of insurance coverage compared to other ethnic groups.

Utilizing grounded theory and the semi-structured interviews of25 participants, this study sought to understand how an insecure attachment affects one's relationship with God. Participants' stories suggested that: I) one's struggle to connect with God was a part of their struggle to connect with others; 2) that experiencing others as safe and responsive contributed to building a more secure attachment with God; and 3) that attachment is less a fixed style and more of an ongoing process: defined as gradual changes made by the participants towards the set-goal of feeling safe and secure. Using the lenses of social constructionism and critical feminist theory, this dissertation examines how transnational Mexican couples in the agricultural town of Mecca, Southern California, respond to the crisis created by their adolescent children using illegal substances.

As part of the Contemporary Couples’ Study of Loma Linda University, sixteen

families were interviewed about how this problem affected their lives. The rich stories

collected were analyzed using the Strauss and Corbin (1998) guidelines for qualitative

According to the U.S. Department of Health and Human Services, 41% of the children in this age group experience decay in their primary teeth. Prostate cancer is stimulated to grow in response to testosterone. Androgen deprivation therapy (ADT) leads to chemical castration and suppression of prostate cancer cell production. Testosterone levels less then 300ng/ml decreases bone mineral density and could result in osteoporosis. Studies have shown that during the first year of ADT, fracture risk, mainly in hips and spine increases about 50%. In men, 40% of hip fractures result in death. Exercise may reduce the risk of osteoporosis and thus contribute to the prevention of hip and other fractures. Background: Over half of the known stillbirths occur in four highly populated countries—India among them. While acknowledged as a significant public health issue in western societies, little is known about maternal coping with stillbirth in developing countries. The purpose of this mixed methods study is to explore how issues of gender and power, social support, coping efforts, and religious beliefs influence perinatal grief outcomes among poor women in rural Chhattisgarh, India.

Background: Evidence suggests that prolonged and exclusive breastfeeding is associated with health benefits to children and nursing mothers beyond the period of breastfeeding. Studies that support the role of breastfeeding in postpartum weight change primarily focused on short-term weight change (within 12 to 24 months postpartum) (Binns et al. 2003; Lederman 2004; Johnson et al. 2006), while few focused on the association between breastfeeding and long-term weight change (5 years or more postpartum) (Rooney, Schauberger et al. 2005).

In breast cancer (BC), IGF2 is initially stimulated by estrogen, progesterone and prolactin to regulate proliferation and cancer progression. These actions are mediated by the IGF-1R and insulin receptor A (IR-A) both members of the tyrosine- kinase receptors family. The activation of Estrogen Receptor (ER) is also very important in BC growth and progression. Background: The National Cholesterol Education Program and American Heart Association have recommended the use of functional or cholesterol-reducing foods, some categories of which include viscous or soluble fibers, soy protein, plant sterols, and nuts, as aids to reduce serum cholesterol concentrations. Figs are a rich source of viscous fiber and antioxidants. Fig consumption has not been studied in regard to effects in reducing serum lipid concentrations. Previous studies demonstrated enhanced fetal adrenal cortisol biosynthesis following exposure to long-term hypoxia (LTH). The studies presented here were designed to determine the mechanism(s) regulating this important adaptive endocrine response. Ewes were maintained at high altitude (3,820 m) from approximately day 40 to 138-141 of gestation. Fetal adrenal glands were then collected from LTH and age matched normoxic controls. Dispersed fetal adrenal cortical cells (FACs) were untreated, treated with ACTH, treated in combination with ACTH precursors (POMC and 22-kDa pro-ACTH), or pre-treated with H-89 and or UO126 followed by ACTH treatment. Background. In the US women are the fastest growing group for sexually transmitted infections (STIs), including HIV and AIDS. In addition, the estimated AIDS cases among female adults and adolescents, aged 13-19, increased from 7% in 1985 to approximately 26% in 2002. Most infections occur by heterosexual transmission with 53% occurring through contact with a high-risk sexual partner. While overall HIV/AIDS rates in the Asian Pacific Islander (API) community remain low, they are rising and HIV testing rates, one of the major prevention strategies for HIV, are lower than that of other populations. Background

Obesity is a major concern in the United States and Black Americans are among those with the highest rates of overweight, obesity, and related co- morbidities. While weight loss among Black women has been studied within the context of a faith-based setting, there is little information about the relationship between spirituality, or relationship with the transcendent, and self-efficacy in the achievement and maintenance of a healthy weight.
