Balloon angioplasty, the most prevalent non-surgical treatment for Atherosclerosis, damages the endothelial layer of the artery, baring an underlying collagenous layer, which causes platelet adhesion and activation and eventual thrombosis and intimal hyperplasia. Previous work in our lab has used a collagen-binding peptidoglycan, dermatan-sulfate-SILY (DS-SILY), that has been shown to bind to type I collagen and prevent platelet adhesion and activation. Our goal is to fabricate nanoparticle-SILY by cross-linking SILY to a poly(N-isopropylacrylamide) (NIPAm) nanoparticle instead of a DS backbone, while retaining the SILY’s high collagen binding affinity and platelet inhibition capacity observed in DS-SILY. Adenylyl cyclases (AC) is a critical family of enzymes which modulates the dynamic cellular level of cAMP, cyclic adenosine monophosphate. The study of cAMP showed that it is indispensable for the signal transduction cascades during many physiological processes, such as immune responses and metabolism which highly relate to cancers. Previous studies of AC inhibitors have been limited due to a lack of isoform-selective small molecule modulators. In vivo toxicology assays are expensive, low-throughput, and often not predictive of a human response. Three-dimensional in vitro human cell-based tissue systems incorporating cell-cell and cell-matrix interactions have promise to provide high-throughput, physiologically-relevant information on the mechanism of the toxin and a more accurate assessment of the toxicity of a chemical before progression to human trials. Quantification of the disruption of vasculogenesis, the de novo formation of blood vessels from endothelial progenitor cells, can serve as an appropriate indicator of developmental toxicity since vasculogenesis is critical to the early development of the circulatory system. Abdominal aortic aneurysms (AAAs), a localized dilation of the vessel wall of 50% or more above normal, claims approximately 14,000 U.S. lives yearly due to aortic rupture. This commonly asymptomatic disease can only be treated by endovascular stent grafts or invasive surgery, usually after the AAA diameter reaches 5 cm. Because these treatment methods carry serious risk, stem cell therapy is being explored in order to provide a low risk option for managing smaller AAAs. To determine if stem cell therapy, once administered, could stabilize or reduce AAA growth, baseline 3D ultrasound measurements in a control group were first needed. …

The ability to make predictions about an epidemic could enable scientists to evaluate inoculation/ isolation plans, control the mortality rate, and prevent the future course of an outbreak. Accurate predictions, however, are extremely hard to make and requires interdisciplinary solutions from epidemiology, sociology, statistics, graph theory, and Computer Science. Protein nanoparticles are ideal carriers for bioactive compounds such as nutraceuticals and drugs because they are biodegradable, less immunogenic and non-toxic and can be nanoparticulated. This study focuses on the desolvation method to form lactalbumin protein nanoparticles. Lactalbumin is soluble in water and insoluble in many organic solvents. Different solvent/non-solvent ratios are evaluated in this research project for the effect they have on the size, PDI and stability of protein nanoparticles. Different methods including sonication and centrifugation were used and compared in terms of their effectiveness to produce small nanoparticles during fabrication of the nanoparticles. Twin screw wet granulation has been considered as an efficient and effective technique of manufacturing granules (agglomerates made up of small particles) and has been widely applied in pharmaceutical industry. However, narrow granule size distributions are difficult to achieve. This study aims to elucidate the effect of process parameters such as liquid feed rate and screw configuration on the granule attributes through granule characterization. The methods used are sieve analysis (for size distribution), liquid distribution (determining the dye concentration in the granules) and density and porosity (percentage gas volume in the granule) analyses. The goal of this experiment is to validate the relationship between hemoglobin saturation (SaO2) and partial pressure of dissolved oxygen (pO2) in breast tumors in mice using photoacoustic computed tomographic (PCT) imaging and OxyLite probe, respectively. In its simplest form, the relationship is described by the dissociation curve, or Hill’s equation, for hemoglobin, and is modeled as a sigmoidal curve that is a function of two parameters – the Hill coefficient, n, and the net association constant of HbO2, K (or pO2 at 50% SaO2). In the field of pharmaceuticals, the crystallization process significantly impacts the purity, morphology and polymorphism of active pharmaceutical ingredients (API), all of which are important in drug development. Polymorphism is the existence of more than one solid-state form of the same chemical entity. Tolfenamic acid (TFA), a Non-Steroid Anti-inflammatory drug (NSAID), has been shown to exhibit a unique physical phenomenon called concomitant polymorphism, whereby two polymorphs crystallize from the solution simultaneously. Systemic toxicity and poor solubility of existing chemotherapeutic drugs piqued an interest in the use of nanocrystals for chemotherapy. To increase cytotoxicity, surface coating of nanocrystals is of interest to enhance tumor targeting and reduce treatment toxicity. As such, we tested in this project various coated paclitaxel nanocrystals on cancer cells for determining the efficacy of surface coating. An IC50 assay was chosen to determine the cytotoxicity of surface-coated paclitaxel nanocrystals; the lower the IC50 value, the higher the efficacy of the drug. The authors consider trends in mobile device usage for the Internet as a whole, for EBSCO Discovery Service across all client libraries, and at two specific libraries: Preston Medical Library, serving the University of Tennessee (UT) Graduate School of Medicine and UT Medical Center, and the Joyner Library at East Carolina University, serving students and faculty on the main campus. Librarians at Preston Medical Library conducted a survey to determine which mobile devices, platforms, and apps were used by their patrons in 2012.
