The efficacy of using 0.01% chlorophacinone on steam-rolled oat (SRO) groats applied in CA alfalfa by spot-baiting/hand baiting around burrow entrances (~11.5 g) to control free-ranging Belding's ground squirrels (Spermophilus beldingi) were compared in 6 randomly assigned square treatment units (TUs). Four TUs were given the rodenticide and 2 treated with placebo bait. Each TU was a 0.4 ha square surrounded by a similarly treated 5.5 ha square buffer zone. Baits were applied on May 13 and re-applied, on May 20 and May 22, after 7 days of un-forecasted cool wet weather greatly reduced their above ground activity. Pesticide (EPA …

Organic farming includes growing food and fiber—animals, agronomic crops, horticultural fruits and vegetables, related products—as one dynamic and rapidly evolving component of our complex U.S. food system. Even as more farmers are moving toward organic certification and participation in an environmentally sound and economically lucrative market, questions arise about the long-term social impacts and sustainability of a set of practices that has gone from a movement to an industry. Sera from white-tailed deer (Odocoileus virginianus) were collected in Iowa during the winter months (1999-2003), 2 years before and after West Nile virus (WNV) was first reported in Iowa (2001), and were analyzed for antibodies to WNV. Samples from 1999 to 2001 were antibody negalive by blocking enzyme-linikedI immunosorbent assay (bELISA) and plaque reduction neutralization test (PRNT90). Prevalence derived from bELISA (2002, 12.7%; 2003. 11.2%) and WNV PRNT90 (2002,7.9%; 2003, 8.5%) assays were similar. A molecular paleoparasitological diagnostic approach was developed for Enterobius vermicularis. Ancient DNA was extracted from 27 coprolites from archaeological sites in Chile and USA. Enzymatic amplification of human mtDNA sequences confirmed the human origin. We designed primers specific to the E. vermicularis 5S ribosomal RNA spacer region and they allowed reproducible polymerase chain reaction identification of ancient material. We suggested that the paleoparasitological microscopic identification could accompany molecular diagnosis, which also opens the possibility of sequence analysis to understand parasite-host evolution.

Scientists face many challenges in harnessing web-based resources. Information overload, misinformation, fees, poorly designed navigation, and loss of browsability all hamper the scientist searcher. In addition, many scientists rely on only one or two databases and often miss unique information that is available through other sources. Librarians can team up with scientists to develop strategies to overcome the challenges of web-based information. For example, a librarian can teach effective information seeking techniques, including how to use controlled vocabularies, how to evaluate information on the web, and how to complement web-based resources with print resources. In order to determine the prevalence and distribution of the human pathogen, Escherichia coli O157:H7, in free-ranging deer, hunters were asked to collect and submit fecal samples from deer harvested during a regular firearm season (14–22 November 1998). Prior to the season, 47% of the hunters with permits in the southeastern Nebraska (USA) study area indicated a willingness to participate in the study. Approximately 25% of successful hunters in the area submitted deer fecal samples. Escherichia coli O157:H7 was cultured from four (0.25%) of 1,608 total samples submitted. Affinity chromatography is a type of liquid chromatography that makes use of biological-like interactions for the separation and specific analysis of sample components. This review describes the basic principles of affinity chromatography and examines its use in the testing of clinical samples, with an emphasis on HPLCbased methods. Some traditional applications of this approach include the use of boronate, lectin, protein A or protein G, and immunoaffinity supports for the direct quantification of solutes. Newer techniques that use antibody-based columns for on- or off-line sample extraction are examined in detail, as are methods that use affinity chromatography in combination with …

Sections 307 and 309 of the 1988 Indoor Radon Abatement Act (IRAA) direct EPA to identify areas of the United States that have the potential to produce elevated levels of radon. EPA, the U.S. Geological Survey (USGS), and the Association of, American State Geologists (AASG) have worked closely over the past several years to produce a series of maps and documents which address these directives. Reversed-phase high-performance liquid chromatography (RP-HPLC) is an important tool in the purification of radiolabeled peptides and proteins for immunoassay. However, for some proteins and peptides it is difficult to achieve reproducible behavior in RP-HPLC because of the low recovery of these compounds. Factors that can be varied to improve recovery include the strength or pH of the mobile phase, the chain length and spacing of groups on the reversed-phase support, and the flow rate or steepness of the elution gradient (1-5). ... The performance and clinical utility of an automated assay of intact parathyroid hormone (parathyrin, PTH) are evaluated. The method is based on the extraction of PTH from plasma by an HPLC column containing immobilized anti-(44-68 PTH) antibodies. The PTH retained is detected with a postcolumn reactor and use of anti-(1--34 PTH) chemiluminescent-labeled antibodies. The total cycle time of the assay is 6.5 mm per injection after a 1-h incubation.The lower limit of detection for PTH in a 66-pL plasma sample was 0.5 pmol/L based on peak heights and 0.2 pmol/L based on peak areas. The compounds used as rodenticides are tremendously varied in their chemical structure and mechanism of action. With a few exceptions, these agents are generally poisonous to all animals, including man, and a great deal of study has been directed to their toxicity in animals other than rodents. However, the development of new compounds as Norbormide and certain antifertility drugs which are highly selective in their action may justify the hope that the ideal rodenticide free of secondary toxic hazards will soon be available.
