Within the past several years, a great deal of attention has been focused on the phenomena of drug-drug interactions and their importance in the therapeutic regimen of patients for whom multiple therapeutic agents might be indicated. The rather sudden concern for this aspect of medical and pharmaceutical practice is evidenced by the proliferation of literature devoted to the topic for both professions. While iatrogenic disease has long been recognized as one of the hazards inherent in prescribing practices, only isolated reports of specific interactions were found until recently. This study was conducted in an attempt to define the anti inflammatory efficacy of cryogenine, the major alkaloid of Heimia salicifolia Link and Otto. The anti inflammatory effects produced by cryogenine, phenylbutazone, indomethacin, mefenamic acid, paramethasone, hydrocortisone and 6-mercaptopurine were compared considering the sequential changes in various motameters during the course of adjuvant-induced polyarthritis in rats. Polyarthritis was induced by the subplantar injection of a suspension of heat-killed Mycobacterium butyricus in mineral oil into the right hind paw of Sprague-Dawley rats. The manifestations of the diseases were monitored in terms of hind paw swelling, subjective appearance of secondary lesions, impairment …

An in vitro method was tested whereby the processes of dissolution and distribution of active ingredient into a lipoid solvent were combined in one model and evaluated.

Until the past few years, cellulose derivatives, (e.g., cellophane, collodion, and parchment) and animal membranes (e.g., goldbeater’s skin) have been the only dialysis membranes employed commercially. Cellophane has been used as the dialysis membrane in the artificial kidney since the machine’s inception in 1914. It continues to serve in this capacity, even though, in the last few years, attempts have been made to develop better films.

It would seem appropriate that studies be conducted on the development of an oral inhalation dosage form antinauseant. The present project will consist of developing and testing a series of aerosol formulations to determine their feasibility in antinauseant therapy. In vitro pressure, valve release, spray pattern, particle size, and aging studies will be performed. The present work will also include an in vivo evaluation of promising formulations.

Intravenous administration of tetracycline has been known to result in fatty degeneration of the liver. This study was undertaken in order to elucidate some of the actions of tetracycline on mammalian cells in culture and to attempt to relate these findings to the clinical condition of fatty degeneration of the liver mentioned above.

It is suggested that lipid accumulation occurring after tetracycline administration is possible the result of a decrease in B-lipoprotein synthesis and enzyme inhibition. These factors could result from the high blood levels of tetracycline seen after large and rapid intravenous injections of the drugs into normal patients …

The possibility of drug incompatibilities is a matter of serious concern when administering two or more therapeutic agents. This is especially true in the case of parenteral administration when one must be aware not only of therapeutic interactions but also chemical and physical reactions that might occur.

In order to avoid incompatibilities, drug manufacturers have suggested that parenteral solutions be used immediately after reconstitution and that admixtures of parenteral products not be administered wherever possible (1,2). Intravenous admixtures consisting of one or more drug formulations, diluted in a large volume of intravenous solution, are used extensively in current medical practice. Development of reliable methods to predict the compatibility of the admixture components is essential as a guide for their selection.

Because the majority of the reports published, to date, have used a visible change as the criterion for judging compatibility, it was the purpose of this study to demonstrate the use of spectrophotometric analysis to detect the occurrence of chemical interactions between two components of an intravenous admixture.

Tetracyclines, originally isolated in the late 1940's and early 1950's from the mycelium of Streptomyces, have achieved major importance as therapeutic and prophylactic agents against a wide range of infections in human and veterinary medicine. They have also achieved great importance in agriculture where they are widely used to promote weight gain in livestock. Commercial production, during 1958 in the United States alone was 120 tons with a value of approximately $1 million per ton.

The objective of the present work was to study the release of 5-FU from various ointment bases and to attempt to determine the type of base which would release the compound most satisfactorily.

Fluidization operations, although relatively unexplored in pharmacy, have been firmly established on a broad scale in other industries. Fluidization may be defined as the suspension and agitation of a bed of particulate solids by a vertically rising stream of gas. Each suspended particle is surrounded by the gas. Research into the chemical and pharmacological properties of aza­steroids has become of interest because of their potential value as pharmacodynamic and chemotherapeutic agents. A nitrogen atom, for example, would be able to interact, unlike carbon, with a variety of sites in enzymes by means of ionic-ionic, ionic-dipole, dipole-dipole and hydrogen.bonds. Azasteroids having a nitrogen, atom might be adsorbed more strongly or in a different orientation to a given enzyme, and also may produce.a different biologic effect with a new spectrum of biological activities. The substance choline, one of the quaternary nitrogen bases, has been the subject of a number of reviews and investigations. It has been described as a cholinergic agent, an inotropic agent, a vitamin, and perhaps in other ways. In spite of the interest shown in its action and uses, there is as yet very little information available as to the mechanism by which it is absorbed by the intestinal mucosa.

Many attempts have been made to prepare aryl boronic acids and their esters which show a preferential affinity for tumor tissues for use in radiation therapy of neoplatic disorders (1,2). The data of Soloway, as extended and refined by Hansch and his coworkers, successfully correlates the low lipid solubility and electron releasing substituents of phyenl boronic acid derivatives with increased tumor concentration (3,4,5). However, most of these organoboron compounds do not produce a sufficiently high tumor-to-normal tissue boron concentration ratio for practical use in human cancer chemotherapy by the neutron capture technique. The organic base choline is of considerable interest to the physiologist because of its role in normal metabolic function, and to the pharmacologist because of its possible application in the treatment of several human diseases.

The overall function of choline in the biologic system must be seen as an equilibrium between its lipotropic activity as a whole molecule and its function as a source of methyl groups in biochemical reactions.

The roots and rhizomes of Anemopsis Californica (Nutt.) Hook and Arn. (family Saururaceae) have been used by the early Spanish Americans and Indians to treat a variety of ailments. The plant is commonly known as Yerba del Mansa, Yerba Mansa, or Manza.

We decided to investigate the essential oil of Anemopsis Californica in greater detail. The results of our investigation to date are included and described in this thesis.

Quantitative Calculation Of Gas Chromatographic Peaks In Pesticide Residue Analyses, Jean A. Gaul

Comparison of five methods for calculating gas chromatographic peaks (disc integration, triangulation, peak height X width at half height, Rt X peak height, and peak height) shows no significant difference in the calculated results when aldrin, heptachlor epoxide, and dieldrin are injected into a gas chromatograph equipped with electron capture detector.

Problems are associated with the calculation of toxaphene, chlordane, DDT, and BHC. The author makes suggestions for calculating these four residues when present individually and in combinations with other pesticides. Use of the last four toxaphene peaks compares favorably with results obtained by using the entire toxaphene curve. …
