Different Roles Of Endothelial Cell-Derived Fibronectin And Plasma Fibronectin In Endothelial Dysfunction, Xiaoxin Luo, Weixiong Jian

MDABackground/ aim: Atherosclerosis is significantly influenced by endothelial cell activation and dysfunction. Studies have demonstrated the substantial presence of fibronectin (Fn) within atherosclerotic plaques, promoting endothelial inflammation and activation. However, cellular Fn (cFn) secreted by various cell types, including endothelial cells and smooth muscle cells, and plasma Fn (pFn) produced by hepatocytes. They are distinct forms of Fn that differ in both structure and function. The specific contribution of different types of Fn in promoting endothelial cell activation and dysfunction remain uncertain. Therefore, this study aimed to investigate the respective roles of pFn and endothelial cell-derived Fn (FnEC) in promoting …

Aims: Drug resistance is a major obstacle for a successful cancer therapy. Cell adhesion mediated drug resistance (CAM-DR) is a novel type of drug resistance and generated via interaction of cancer cells with the microenvironment. In this study, CAM-DR phenotype was analyzed in HL60 acute myeloid leukemia (AML) cells. Materials and Methods: Fibronectin (FN) adherence of HL60 cells was tested by a colorimetric adhesion assay. Flow cytometry analyses were performed to evaluate doxorubicin-induced apoptosis and to determine cell cycle status. Proliferation rate was evaluated by [^3H]-thymidine incorporation assay. Western blot and RT-PCR were used for analysis of the factors involved …

Distribution Of Different Fibronectin Isoforms In The Extracellular Matrices Of Human Term Placenta, Ayşe Yasemi̇n Demi̇r Weusten

Fibronectins are considered to be molecules that play important roles in cell migration and cell differentiation. Fibronectin molecules are thought to influence the differentiation of trophoblasts and the adhesion of these cells to maternal tissues. The aim of the present study was to analyze the distribution of fibronectin isoforms and to evaluate the differences between matrix-type fibrinoids, and other types of matrices produced by both maternal and fetal tissues. Tissue samples were collected from different sites of human term placentas. By using immunohistochemistry, the expression of fibronectin isoforms was determined and they were demonstrated by SDS-PAGE and Western blotting methods. …

The involvement of both the peripheral nervous system (PNS) and central nervous system (CNS) in multiple sclerosis is a seldom encountered combination in neurology clinics. In this report, immunohistochemical and ultrastructural techniques were used to analyse the disease course of a patient exhibiting involvement of both systems. In 1980, a 44-year-old man was admitted to our clinic with progressive weakness and decreased sensation of the right lower extremity, and sensory abnormality exhibiting stocking distribution. In electromyolography (EMG), demyelinating sensory motor polyneuropathy was determined. Ten years later, he was admitted again due to progression of weakness in the four extremities. Pyramidal …

Serum fibronectin, haptoglobin, ceruloplasmin, and transferrin concentrations were measured in 44 term neonates with culture proven sepsis and a control group of 37 normal neonates of similar age, sex and weight. The white blood cell count, total neutrophil count, and the ratio of band to neutrophils were also determined. While the serum fibronectin and haptoglobin concentrations were significantly different (P

The pattern of distribution of fibronectin adhesins on yeast and hyphal-form Candida albicanswas studied with gold-labelled fibronectin and transmission electron microscopy. The germ-tube parts of the hyphal-form cells were the major binding sites for gold-labelled fibronectin, and the attached yeast-cell parts and yeast-form cell remained unlabelled. Therefore, the walls of the germ tube parts of hyphal-form cells might be sites of important interactions between C. albicans and host tissues containing fibronectin.

Ascites, being an accumulation of excess fluid within the peritoneal cavity, is most frequently encountered in patients with cirrhosis or certain other diseases such as congestive cardiac failure, peritoneal tuberculosis and some primary or secondary peritoneal tumours. This study included 17 patients (5 females, 12 males) with malignant ascites (MA) and 23 patients (13 females, 10 males) with nonmalignant ascites (NMA). We aimed to determinet the values for total sialic acid (SA), pH, cholesterol, fibronectin, total protein concentrations and lactate dehydrogenase (LDH) activity in ascitic fluid for the differential diagnosis of ascites. While total SA, fibronectin, cholesterol, LDH and total …

In this study, plasma fibronectin and urine glycosaminoglycane levels were determined spectrophotometrically in 20 Rheumatoid arthritis, 10 Ankilosing spondilites, 22 osteoarthrosis and 21 healthy control subjects. The mean levels of Fibronectin (\mug/ml) were 413.3±97 in patients with rheumatoid arthritis, 325.5±93.5 in ankilosing spondilites, 439.1±120.3 in osteoarthrosis and 302.2±60.1 in the control group. The ratio of urine glycosaminoglycan excretion to creatinine (g/mol) was 4.52±2.64 in patients with rheumatoid arthritis, 4.18±1.52 in patients with ankilosing spondilites, 2.72±1.19 in osteoarthrosis and 2.21±1.02 in the control group. Plasma Fibronectin levels were significantly higher in rheumatoid arthritis and osteoarthrosis when compared to the control group …
