Turkish Journal of Medical Sciences

Background/aim: Methotrexate (MTX) is a folic acid antagonist that is widely used to treat osteosarcoma, leukemia, breast cancer, and autoimmune and inflammatory diseases. The most important concerns with MTX are its poor solubility and high toxicity, particularly in liver cells. To enhance its solubility and to minimize its toxicity, we encapsulated MTX in niosomes and investigated its hepatotoxicity mechanisms using genetic biomarkers. Materials and methods: Niosomes were successfully prepared using a modified thin film method, and the prepared monodisperse smallsized formulation was subsequently characterized. In vitro cytotoxicity studies were performed both in hepatocarcinoma (HEP3G) and healthy liver (AML12) cell lines. …

Melatonin, Aging, And Covid-19: Could Melatonin Be Beneficial For Covid-19 Treatment In The Elderly?, Güler Öztürk, Kazi̇me Gonca Akbulut, Şevi̇n Güney

Turkish Journal of Medical Sciences

The aim of this review is to summarize current studies on the relationship between melatonin and aging. Nowadays, age-related diseases come into prominence, and identifying age-related changes and developing proper therapeutic approaches are counted as some of the major issues regarding community health. Melatonin is the main hormone of the pineal gland. Melatonin is known to influence many biological processes in the body, including circadian rhythms, the immune system, and neuroendocrine and cardiovascular functions. Melatonin rhythms also reflect the biological process of aging. Aging is an extremely complex and multifactorial process. Melatonin levels decline considerably with aging and its decline …

Physiological And Pharmacological Roles Of Melatonin In The Pathophysiologicalcomponents Of Cellular Injury After Ischemic Stroke, Ertuğrul Kiliç, Berrak Çağlayan, Mustafa Caglar Beker Jan 2020

Physiological And Pharmacological Roles Of Melatonin In The Pathophysiologicalcomponents Of Cellular Injury After Ischemic Stroke, Ertuğrul Kiliç, Berrak Çağlayan, Mustafa Caglar Beker

Turkish Journal of Medical Sciences

Apart from its metabolic or physiological functions, melatonin has a potent cytoprotective activity in the physiological and pathological conditions. It is synthetized by the pineal gland and released into the blood circulation but particularly cerebrospinal fluid in a circadian manner. It can also easily diffuse through cellular membranes due its small size and lipophilic structure. Its cytoprotective activity has been linked to its potent free radical scavenger activity with the desirable characteristics of a clinically- reliable antioxidant. Melatonin detoxifies oxygen and nitrogen-based free radicals and oxidizing agents, including the highly toxic hydroxyl-and peroxynitrite radicals, initiating cellular damage. However, the cytoprotective …

Ilp-2 Modeling And Virtual Screening Of An Fda-Approved Library:A Possible Anticancer Therapy, Saeed Khalili, Hemn Mohammadpour, Mahideh Shokrollahi Barough, Parviz Kokhaei

Turkish Journal of Medical Sciences

Background/aim: The members of the inhibitors of apoptosis protein (IAP) family inhibit diverse components of the caspase signaling pathway, notably caspase 3, 7, and 9. ILP-2 (BIRC-8) is the most recently identified member of the IAPs, mainly interacting with caspase 9. This interaction would eventually lead to death resistance in the case of cancerous cells. Therefore, structural modeling of ILP-2 and finding applicable inhibitors of its interaction with caspase 9 are a compelling challenge. Materials and methods: Three main protein modeling approaches along with various model refinement measures were harnessed to achieve a reliable 3D model, using state-of-the-art software. Thereafter, …

Utility Of M30, An Apoptotic Serum Marker, In Liver Diseases, Aki̇f Altinbaş, Şahi̇n Çoban, Ömer Başar, Osman Yüksel Jan 2015

Utility Of M30, An Apoptotic Serum Marker, In Liver Diseases, Aki̇f Altinbaş, Şahi̇n Çoban, Ömer Başar, Osman Yüksel

Turkish Journal of Medical Sciences

The aim of this paper is to evaluate the role of apoptosis in some common liver diseases, and the utility of M30, an apoptotic serum marker, in the diagnosis of the severity of underlying hepatic injury. As is widely known, apoptosis is programmed cell death, and its deregulation results in an uncontrolled inflammatory process leading to upregulation of liver fibrogenesis. Both extrinsic and intrinsic pathways are crucial in apoptosis, and caspase cleavage of cytokeratin proteins occurs in both. Therefore, the measurement of caspase-cleaved cytokeratin fragments could be a novel method to assess the intensity of apoptotic cell numbers in epithelial …

To investigate the renal expression of apoptosis regulatory proteins b-cell lymphoma 2 (bcl-2) and bcl-2 associated X (bax) in children with Henoch–Schönlein nephritis (HSN) Materials and methods: Renal expression of antiapoptotic bcl-2 and proapoptotic bax proteins were studied immunohistochemically in 22 children with HSN. The kidneys of 5 children who died due to nonrenal reasons were used as the control. The patients were compared according to their renal histopathology and prognosis. Results: Nephrotic or nephrotic-nephritic renal involvement was the most common presentation. Histopathological findings were Class I in 1, Class II in 5, Class III in 10, Class IV in …

To investigate the effect of vardenafil on ischemia–reperfusion (I/R) injury in rat ovary. Materials and methods: A total of 21 Wistar rats were divided into 3 groups. In group 1, ovary torsion was not performed and no drug was administered. In group 2, 1 h of ischemia and 2 h of reperfusion were performed and no drug was given. In group 3, 1 h of ischemia and 2 h of reperfusion were performed and 1 mg/kg vardenafil was administered after 30 min of ischemia. Right ovaries were surgically removed in all groups. Malondialdehyde (MDA) levels and activities of superoxide dismutase …

To evaluate the effects of Helicobacter pylori on cell proliferation and apoptosis in gastric cancer (GC) both at tumoral and nontumoral sites and in gastritis. Materials and methods: Biopsies from 96 patients (gastric cancer = 52, gastritis = 44) were examined immunohistochemically and histopathologically. Results: In H. pylori-positive GC patients the average proliferative index (PI) in tumor and nontumor sites was 40.7% and 24.8%, while in H. pylori-negative patients it was 32.9% and 17.8%, respectively. The PI was significantly higher in the tumor than in the nontumor sites, regardless of H. pylori infection. Cell proliferation was also significantly associated with …
