The Effect Of Diphtheria Toxin On Nitric Oxideinduction From Raw264.7 Murine Macrophages, Cemalettin Aybay, Gülnur Tarhan, Turgut İmi̇r, Başak Kayhan

In this study the effect of diphtheria toxin (DT) on nitric oxide (NO) production from RAW 264.7 murine macrophages was investigated. Griess reagent was used to determine NO production by measuring nitrite levels in the culture supernatants. Corynebacterium diphtheriae G12/6 strain-produced DT (Limes flocculation activity and immunodiffusion assays were positive) demonstrated a dose-dependent effect on RAW 264.7 macrophages to induce NO production. L-NAME, an L-arginine analogue, inhibited NO production from DT-stimulated RAW 264.7 macrophages. We examined the effect of global cerebral ischemia (Two vessel occlusion plus systemic hypotension method) for 15 minutes on the hippocampal CA1 and CA3 neurones in rats, and compared with turn prefence scores. 14 of 27 animals were ambidexter while 9 had left and 4 had right turn preference. The left turns were significantly different from rights (t:2,26 df:26 p:0,032). Male rats prefered 62,73% and females 56,88% left turns. Erythrosine (FD&C Red No.3, di sodium salt of 2,4,5,7-tetra-iodofluorescein); is used extensively as a colour additive in foods, drugs and cosmetics. Previous studies have shown that some of the food additives, depending on their long-term and short-term use, have potential carcinogenic, toxic and teratogenic effects. Diethylstilbestrol (DES), nonsteroid in structure, is an artificial estrogen derivative and is used as an anabolic agent. It is known that diethylstilbestrol causes genetic disorders and toxic effects leading up to carcinoma even in amounts below its residual levels. For this purpose, a dose of 60µm/kg/day of DES dissolved in 0,1 ml corn oil has been administered to rats by oral gavage for varying periods of 20, 50 and 100 days periods. Raw milk samples obtained from the Foundation of Milk Industry, three private dairy companies and individual dairy farms in Sivas, were tested for the presence of Listeria species. A total of 100 raw milk samples were analysed by Food and Drug Administration (FDA) and two-stage enrichment techniques. Two-stage enrichment procedure was found to be more effective than FDA method in isolating Listeria species from raw milk. The overall incidence of Listeria species in raw milk was 6%. In this study we have showed that zinc ion has an inhibitory activity on imipenem and meropenem suspectibility of Pseudomonas aeruginosa. When zinc ion concentration of Mueller-Hinthon agar (MHA) media was gradually increased the zone diameters of imipenem and meropenem gradually decreased. So we concluded that zinc concentration in the media might effect imipenem and meropenem susceptibility of Pseudomonas aeruginosa and might indicate false resistance to imipenem and meropenem. Therefore the use of these antipseudomonal antibiotics in Pseudomonas infections may be restricted.

In this study, the in vitro effects of some antibiotics on carbonic anhydrase (CA) isozymes have been investigated. Human erythrocyte CA-I and CA-II isozymes were separately purified by affinity chromatography. Inhibition effects of four different antibiotics on CA isozymes were determined by using the CO 2 -Hydratase method. I 50 values of antibiotics were obtained by drawing % Activity [Actibiotic] graphs. In this study the age at menopause and factors influencing age at menopause were investigated by a questionnaire in 282 women. The mean age of menopause was found to be 45.3 years and ranged from 35 to 54. Positive relationships were found between age at menopause and marital satus, body mass index, gravidity, parity, age at menarche, menstruation and menstruation bleeding period. Whereas negative relationships were found between age at menopause and smoking, age at first birth, abortion status and oral contraceptive use.

Paradoxical therapy consists of suggesting that the patient intentionally engages in the unwanted behaviour, such as performing complusive ritual or bringing on a conversion attack. Diabetic patients show hemobiological abnormalities such as increased platelet adhesiveness, platelet hyperaggregability, decreased platelet half life, hemorheological abnormalities and altered fibrinolysis, perhaps contributing to a procoagulative state. Gliclazide, a novel sulfonylurea in routine clinical use, was thought to have effects on prostanoid release and platelet function. We studied thromboxane A 2 metabolite; serum thomboxane B 2 (TXB 2 ) and the prostacyclin metabolite, 6-keto-PGF 1\alpha to assess the efficacy of gliclazide on these parameters. In this study, plasma fibronectin and urine glycosaminoglycane levels were determined spectrophotometrically in 20 Rheumatoid arthritis, 10 Ankilosing spondilites, 22 osteoarthrosis and 21 healthy control subjects. The mean levels of Fibronectin (\mug/ml) were 413.3±97 in patients with rheumatoid arthritis, 325.5±93.5 in ankilosing spondilites, 439.1±120.3 in osteoarthrosis and 302.2±60.1 in the control group. The ratio of urine glycosaminoglycan excretion to creatinine (g/mol) was 4.52±2.64 in patients with rheumatoid arthritis, 4.18±1.52 in patients with ankilosing spondilites, 2.72±1.19 in osteoarthrosis and 2.21±1.02 in the control group. The production and detection of a mannoprotein adhesin (MPA) of the hyphal-form cells of C. albicans on plastic petri dishes was investigated. Using Concanavalin A-coated latex microspheres, the MPA was detected on the plastic surface on which C. albicans produced germ tubes. The adhesin was extracted using dithiothreitol and iodoacetamide. It did not inhibit the adhesion of the yeast-form C. albicans to buccal epithelial cells (BEC). This suggested that the MPA of the hyphal-form cells on the plastic was different from that on the yeast cells responsible for attachment to the BECs.

The pattern of distribution of fibronectin adhesins on yeast and hyphal-form Candida albicanswas studied with gold-labelled fibronectin and transmission electron microscopy. The germ-tube parts of the hyphal-form cells were the major binding sites for gold-labelled fibronectin, and the attached yeast-cell parts and yeast-form cell remained unlabelled. Therefore, the walls of the germ tube parts of hyphal-form cells might be sites of important interactions between C. albicans and host tissues containing fibronectin.

In order to show the neuronal processes, the Golgi-Cox impregnation technique was used. The first neuronal process development was observed in the cranial and caudal poles of the cerebral cortex during the 15th prenatal day and continued spreading from the lateral regions and poles towards the dorsocentral regions of the hemispheres. In this study, a new technique developed by us, called the "pad glove method" was used to measure sweat intensity in the palms of 15 patients with palmoplantar hyperhidrosis, and of 15 control subjects. Quantitative results in the hyperhidrosis group 2.1861±0.0836 grams/hour for the right hand and 2.2478±0.0961 grams/hour for the left hand, while in the control group were 0.3278±0.0419 grams/hour and 0.371±0.0501 grams/hours, respectively. The aims of this study were to determine the degree of lipid peroxidation in the sera of postmenopausal women and compare this with the lipid peroxidation of a premenopausal group, and to see if there was any correlation between lipid parameters and lipid peroxidation within the groups. We assayed the lipid profiles and malondialdehyde (MDA), a marker for lipid peroxidation, in a postmenopausal group (n=57, mean age=49.4±6.6), an age-matched male group (n=21, mean age=48.5±6.7) and a control group (n=31, mean age=27.7±5.2). In this study, we investigated first; the possible involvement of nitric oxide and antioxidant enzyme level in the pathogenesis of essential hypertension and second we investigated serum nitrite-nitrate and antioxidant enzyme activities and lipid profiles (total, HDL and LDL cholesterol, trygliceride) to determine the relationship between these parameters in atherosclerosis. 13 patients with coronary heart disease, 18 patients with essential hypertension and 16 age-matched health subjects were taken into this study. Hypervolemia is supposed to be the principal factor in hypertension in chronic hemodialysis adults. In this study, the role of intensive ultrafiltration (UF) treatment to reduce hypertension due to hypervolemia in hemodialysis children was investigated. The inferior vena cava (IVC) was also evaluated as an index of UF therapy. Twenty-three hemodialysis children with the mean age 10.6±3.1 years were classified due to their blood pressure status. Medullary thyroid carcinoma (MTC) may present in various clinical forms, either sporadic or a counterpart of hereditary endocrine disorders. The appropriate surgical procedure for MTC is total thyroidectomy combined with central neck clearance. The aim of this study was to document the initial clinical presentation, surgical treatment and clinical course of MTC in 27 patients. Thirteen (48.1%) cases were diagnosed as hereditary, either MEN 2a(7/27, 25.9%) or 2b syndrome (1/27, 3.7%) or familial non-MEN MTC (5/27, 18.5%), and fourteen cases were diagnosed as sporadic. Ascites, being an accumulation of excess fluid within the peritoneal cavity, is most frequently encountered in patients with cirrhosis or certain other diseases such as congestive cardiac failure, peritoneal tuberculosis and some primary or secondary peritoneal tumours. The importance of bone marrow biopsy in Hudgkin's Disase is well known. The incidence of bone marrow involvement in Hudgkin's Disase varies from 2% to 29%. The presence of myelofibrosis is not sufficent evidence for a definitive diagnosis of bone marrow involment. Patients having myelofibrosis as a first symptom without pripherallympadenopathy must be distinguished from patients with idiopathic myelofibrosis. In this study, we examined the frequency of bone marrow involvement and the degree of myelofibrosis in Hudgkin's Disase.
