In Silico Design And In Vitro Development Of A Highly Accurate Test To Detect Brucella Species, Hosein Safari, Setareh Shahnazari, Maysam Mard-Soltani, Somayeh Tavakoli, Bahman Khalesi

Turkish Journal of Medical Sciences

Background: Conventionalmethods of detecting Brucella spp. suffer from technical and biological complications. Besides, newly characterized species of the genus Brucella could be neglected by previously designed polymerase chain reaction (PCR) tests. Therefore, a more accurate PCR-based test seems to be imminently needed. Materials and methods: Blood samples were collected from 39 patients diagnosed with brucellosis and 25 healthy controls. Multiple sequence alignments (MSA) were performed on 500 Omp2-related protein and gene sequences. Thereafter, specific primers were designed and synthesized for the regions with highest conservancy. The collected samples were assessed by PCR test. To overcome the cross-reactivity issue, PCR thermal …

Background/aim: Changes in collagen metabolism and fibroblastic activity may play a role in the pathogenesis of brucellosis. The prolidase enzyme plays an important role in collagen synthesis. We aimed to investigate the association of prolidase levels with brucellosis.Materials and methods: Serum prolidase levels in 20 patients newly diagnosed with brucellosis were compared with levels in 30 healthy control subjects. Patients with brucellosis were reassessed 3 months later for prolidase, other laboratory measurements, and response to treatment. Results: The levels of serum prolidase were significantly higher in brucellosis patients compared with those of healthy controls. Prolidase, sedimentation, and C-reactive protein levels …

Comparison Of Standard Agglutination Tests, Enzyme Immunoassay, And Coombs Gel Test Used In Laboratory Diagnosis Of Human Brucellosis, Emi̇ne Emel Koçman, Memnune Selda Erensoy, Meltem Taşbakan, Meltem Çi̇çekli̇oğlu

Turkish Journal of Medical Sciences

Background/aim: It was aimed to evaluate the results of Rose Bengal (RB), ELISA total tests (IgM and IgG), and the Brucella Coombs gel test (BCGT), which are used as screening tests, with the combined results of a tube agglutination test (standard Wright test: SWT) and a tube agglutination test with antihuman globulin (AHG TAT). Materials and methods: Samples from 97 patients prediagnosed with brucellosis (age ≥18 years) were screened with RB, ELISA, and BCGT. SWT < 160 samples and RB-negative but ELISA- or BCGT-positive samples were tested by AHG TAT. SWT ≥ 160 or AHG TAT ≥ 160 was taken as reference. Results: Thirty-two of 56 RB-positive samples and one RB-negative but ELISA- and BCGT-positive sample were found to be ≥160 with SWT or AHG TAT. Sensitivity, specificity, accuracy, and agreement (kappa) values according to SWT ≥ 160 or AHG TAT ≥ 160 positivity were as follows, respectively: RB 96.9%, 62.5%, 74.2%, and 0.509; ELISA total 100%, 60.9%, 74.2%, and 0.515; BCGT test 97%, 70.3%, 79.4%, and 0.594. Conclusion: Sensitivities of the screening tests are similar, but positivities should be confirmed by more specific tests. Positive samples from screening tests should be tested with AHG if the SWT value is

Materials and methods: The diagnosis was made with clinical, serological and/or cerebrospinal fluid, and electromyographic examinations. Results: All of the patients except one improved quickly after specific antimicrobial treatment. Furthermore, electrophysiological examinations revealed normal findings in 6 patients. Conclusion: This clinical form of brucellosis is mostly reversible if it is diagnosed earlier and treated effectively.

Comparison Of The Diagnostic Value Of The Standard Tube Agglutination Test And The Elisa Igg And Igm In Patients With Brucellosis, Mustafa Ertek, Hali̇l Yazgi, Zülal Özkurt, Ahmet Ayyildiz, Mehmet Parlak

Turkish Journal of Medical Sciences

Background and Aims: Brucellosis is endemic in Turkey. Since it affects many organs and the symptoms are non-specific, the diagnosis by clinical findings is difficult and may be easily missed. Many serological tests have been used for the diagnosis of human brucellosis. This study compared the diagnostic value of the Brucella standard tube agglutination test (SAT) with that of ELISA (Brucella specific IgG and IgM) tests in patients with Brucella bacteremia. Patients and Methods: Thirty-two patients with brucellosis who had positive blood and/or bone-marrow cultures for Brucella species, and 20 healthy individuals as controls were included in the study. Results: …

Turkish Journal of Medical Sciences

The aim of this work was to evaluate changes in lymphocyte subpopulations, especially helper and cytotoxic T cells, in acute brucellosis patients undergoing treatment. Forty-three acute brucellosis patients were included in the study. Twenty healthy subjects served as controls. Total lymphocytes and the CD3^+, CD4^+, CD8^+, CD19^+ and CD (16+56)^+ subpopulations were counted by two-color flow cytometric analysis. The CD4+ counts in patients before and after treatment were not statistically different (p = 0.7), but healthy subjects had significantly more of these cells (p = 0.001 and p = 0.001 compared to pre- and post-treatment patients, respectively). The CD8^+ counts …
