The aim of this study was to evaluate the serological diagnostic potential of the Brucella recombinant outer membrane (rOMP25, rOMP31) and periplasmic proteins (rBP26, rSOD) in a comparative way using an indirect enzyme-linked immunosorbent assay (i-ELISA). Rabbit and/or mouse antibodies to Brucella whole cell and/or soluble protein preparations recognized all recombinant proteins used, which confirms the expression of target antigens in E. coli in active form. The recombinant proteins showed different antigenicity to antibodies of cattle kept on a brucellosis-affected (endemic) farm and/or a new focus of infection. Background/aim: Changes in collagen metabolism and fibroblastic activity may play a role in the pathogenesis of brucellosis. The prolidase enzyme plays an important role in collagen synthesis. We aimed to investigate the association of prolidase levels with brucellosis.Materials and methods: Serum prolidase levels in 20 patients newly diagnosed with brucellosis were compared with levels in 30 healthy control subjects. Patients with brucellosis were reassessed 3 months later for prolidase, other laboratory measurements, and response to treatment. Background/aim: It was aimed to evaluate the results of Rose Bengal (RB), ELISA total tests (IgM and IgG), and the Brucella Coombs gel test (BCGT), which are used as screening tests, with the combined results of a tube agglutination test (standard Wright test: SWT) and a tube agglutination test with antihuman globulin (AHG TAT). Materials and methods: Samples from 97 patients prediagnosed with brucellosis (age ≥18 years) were screened with RB, ELISA, and BCGT. SWT < 160 samples and RB-negative but ELISA- or BCGT-positive samples were tested by AHG TAT. SWT ≥ 160 or AHG TAT ≥ 160 was taken as reference. Results: Thirty-two of 56 RB-positive samples and one RB-negative but ELISA- and BCGT-positive sample were found to be ≥160 with SWT or AHG TAT. Sensitivity, specificity, accuracy, and agreement (kappa) values according to SWT ≥ 160 or AHG TAT ≥ 160 positivity were as follows, respectively: RB 96.9%, 62.5%, 74.2%, and 0.509; ELISA total 100%, 60.9%, 74.2%, and 0.515; BCGT test 97%, 70.3%, 79.4%, and 0.594. Conclusion: Sensitivities of the screening tests are similar, but positivities should be confirmed by more specific tests. Background/aim: To determine the relationship between human leukocyte antigen (HLA) antigens and osteoarticular brucellosis by evaluating HLA Class I and II antigens in control subjects and patients developing osteoarticular brucellosis in Turkey.Materials and methods: The study included 28 patients with osteoarticular involvement diagnosed with brucellosis and 100 controls. Trás-os-Montes e Alto Douro, in the northeastern region of Portugal, previously exhibited the highest brucellosis prevalence in the entire small ruminant population of Portugal. Consequently, a vaccination program of the whole population with Brucella melitensis Rev-1 was carried out from 2001 to 2004, and further compulsory Rev-1 vaccination of 3- to 6-month-old lambs and kids was carried out between 2005 and 2007. The prevalence of brucellosis decreased by 71.42% in 2004, with a 2-fold decrease occurring from 2005 to 2007. The reduction of brucellosis prevalence was statistically significant (P < 0.001) according to flock size. By species, brucellosis prevalence decreased 14.5-fold in sheep and 5.5-fold in goats in 2004. Regarding flock constitution, it decreased by 77% for pure flocks and 73% in mixed flocks in 2004. Regarding the animals involved in production, brucellosis prevalence decreased during the 7-year period. It was concluded that Rev-1 vaccination of the whole population was effective in decreasing brucellosis prevalence. These results contribute to the scarce information available regarding the effect of Rev-1 vaccination on different characteristics of flocks. They may be used to improve the efficiency of brucellosis eradication programs within livestock management.

Brucellosis is an important disease in developing countries. We aimed to determine the epidemiologic, clinical, and laboratory characteristics of brucellosis, which still has a high morbidity in Turkey. Materials and methods: Seventy-two patients with brucellosis, monitored at our clinic from January 2004 to July 2010, were reviewed retrospectively. In the present study, 178 blood samples from buffaloes were tested against indirect enzyme linked immunosorbent assay (I-ELISA), rose bengal plate test (RBPT), microagglutination test (MAT), modified microagglutination test (mMAT), and polymerase chain reaction (PCR) to select the most suitable test for efficient and effective diagnosis of bovine brucellosis. To evaluate the epidemiology, clinical features, and complications of brucellosis. Materials and methods: Between January 2005 and December 2008, 161 patients with brucellosis who were admitted to our clinic were evaluated prospectively. Results: Of the patients, 97 (60.2%) were male, and the mean age was 47 ± 17 years (range: 16-79). There were 99 patients (61.5%) classified as acute, 36 (22.4%) classified as subacute, and 6 (3.7%) classified as having a chronic infection. Many reports suggest that a decline in vaccine productivity may be due to antigenic shifts in the circulating Brucella melitensis. To present 8 patients with polyradiculopathy or polyradiculoneuritis caused by brucellosis. Polyradiculopathy or polyradiculoneuritis are occasional neurological involvements of brucellosis. Materials and methods: The diagnosis was made with clinical, serological and/or cerebrospinal fluid, and electromyographic examinations. Results: All of the patients except one improved quickly after specific antimicrobial treatment. Furthermore, electrophysiological examinations revealed normal findings in 6 patients. Conclusion: This clinical form of brucellosis is mostly reversible if it is diagnosed earlier and treated effectively.

Brucellosis is a prevalent disease in Turkey. The object of this study was to assess the findings in brucellosis cases with and without complications. Materials and methods: A total of 324 patients diagnosed with brucellosis between 1999 and 2005 were reviewed retrospectively. The diagnosis of brucellosis was established serologically and/or by positivite blood culture. Epidemiologic features, clinical symptoms, and laboratory findings of the patients were assessed in both complicated and noncomplicated brucellosis cases by univariate and multivariate analysis. A total of 1500 serum samples were collected from cattle, buffaloes, and humans. The Rose Bengal plate test (RBPT) and enzyme-linked immunosorbent assay (ELISA) detected Brucella antibodies in 56 (10.18%) and 44 (8%) of the cattle, respectively. In samples collected from buffaloes 9.38% and 6.92% were positive, whereas 14% and 11% of the samples from humans were found to be positive by RBPT and ELISA, respectively. Among village workers a higher prevalence of brucellosis was recorded in females than in males. Abattoir-associated personnel also had a higher incidence of brucellosis. Aim: Brucellosis is a worldwide zoonotic disease that remains an important public health problem in rural Turkey. The aim of the present study was to identify Brucella species and biotypes, and to assess the antimicrobial susceptibility of isolates from blood samples. Materials and Methods: The study included 46 Brucella isolates from the Kırıkkale region of central Anatolia. The identification and biotyping of the isolates were based on conventional methods. The seroprevalence of Brucellosis in human, sheep, and cattle populations was investigated in rural regions of Kırıkkale in Turkey. Serum samples were obtained from 1436 humans, and 3.2% (46) of the population was found to be positive by Rose Bengal Plate test (RBPT) and 3.0% (43) gave positive results with the standard tube agglutination test (STAT). Aim: Brucellosis is a systemic infection in which any organ or system of the body can be involved. We evaluate here complications and involvement of Brucella infection in Erzurum, Turkey. Materials and Methods: We analyzed retrospectively 216 adult (89 female and 127 male) cases of brucellosis who were diagnosed and treated in the Infectious Disease Clinic of Atatürk University Research Hospital from 1985 to 2002. This study was planned in order to make good the lack of information about population-based data on the prevalence of brucellosis in the Kayseri region. This cross-sectional research was performed on 1850 individuals living in the rural regionaround Kayseri. People aged 15-85 in 9 districts in this rural areas were interviewed and blood samples were screened using the Rose Bengal Agglutination test. The overall prevalence of brucellosis was 3.4%. The aim of this work was to evaluate changes in lymphocyte subpopulations, especially helper and cytotoxic T cells, in acute brucellosis patients undergoing treatment. Forty-three acute brucellosis patients were included in the study. Twenty healthy subjects served as controls. Total lymphocytes and the CD3^+, CD4^+, CD8^+, CD19^+ and CD (16+56)^+ subpopulations were counted by two-color flow cytometric analysis. The CD4+ counts in patients before and after treatment were not statistically different (p = 0.7), but healthy subjects had significantly more of these cells (p = 0.001 and p = 0.001 compared to pre- and post-treatment patients, respectively). This study was designed to determine the seroprevalence of brucellosis and leptospirosis in aborted dairy cattle. For this purpose, serum samples were collected from 163 aborted dairy cattle that had no history of vaccination against either brucellosis or leptospirosis. The antibodies against B. abortus were detected in these serum samples as 68.1%, 65.6%, 58.9% and 55.2%, respectively by the Competitive Enzyme-Linked Immunosorbent Assay (C-ELISA), Complement Fixation Test (CFT), Rosebengal Plate Test (RBPT) and Serum Agglutination Test (SAT). In this study, the brain and other organs from an aborted bovine foetus were examined histopathologically and microbiologically. In addition, a serum sample from the aborting cow was examined serologically. A granulomatous inflammation, characterised by the infiltration of lymphocytes, plasma cells with some neutrophils and macrophages was observed in the meninges of the brain, liver and kidney. Brucella abortus biovar 1 was isolated in pure culture from the brain and other organs. The serum sample was positive for brucellosis. These findings indicate that Brucella abortus is a pathogen capable of inducing meningitis in bovine foetuses.

This study was conducted to determine brucellosis seropositivity in patients suspected of having brucellosis who sought treatment at the Central Laboratory of Dicle University Medical Faculty Hospital in southeast Turkey (Diyarbakır). 20,663 serum samples were collected during the study (1 August 2001-31 December 2002), and 14,480 sera were tested in a 12-month period on a seasonal basis by Rose-Bengal slide agglutination (RBSA), and positive sera were titrated by standard tube agglutination (STA). The aim of this study was to determine the serum adenosine deaminase ( ADA ) levels in patients with brucellosis and compare them with the results of healthy individuals. Forty- eight subject were enrolled in this study : 34 patients with brucellosis and 14 healthy individuals. Serum ADA activity was assessed by spectrophotometer in patients with brucellosis and statistically compared with that of healthy individuals. A normal range of serum ADA was considered 5-35 IU/l. In this study, we aimed to investigate the regional profile of bovine and ovine Brucellosis in Burdur by isolating Brucella spp. in milk and by detecting antibodies to Brucellae in milk samples. For this purpose, 404 milk samples from 101 cows and 226 milk samples from 113 ewes located in 14 different areas of Burdur were collected. The samples were examined by bacteriological and serological methods. Brucella spp. were not isolated from the milk samples. In this study,antibodies against Brucella melitensis in aborted sheep were investigated with enzyme-linked immunosorbent assay (ELISA), standard tube agglutination test (SAT), rose bengal plate test (RBPT), complement fixation test (CFT), 2-mercaptoethanol microagglutination test (2-ME-MAT)and dithiothreitol microagglutination test (DTT-MAT). A comparative study of the Rose Bengal test (RBT), Enzyme-Linked Immunosorbent Assay (ELISA) and complement fixation test (CFT) was performed to analyze sera of two Awassi flocks. Flock A was composed of 215 ewes, naturally infected with brucellosis. None of the animals were vaccinated. Flock B consisted of 336 non-infected non-vaccinated ewes.
