The major causes of death in burn patients include multiple organ failure and infection. It is important for the clinician to understand the pathophysiology of burn injury and the effects it will have on the pharmacokinetics of a drug. The local and systemic inflammatory response to thermal injury is extremely complex, resulting in both local burn tissue damage and deleterious systemic effects on all other organ systems distant from the burn area itself. Thermal injury initiates systemic inflammatory reactions producing burn toxins and oxygen radicals and finally leads to peroxidation. Comparative Cytotoxicity Of Fludarabine In Human Peripheral Blood Mononuclear Cells And In The Promyelocytic Leukemia Cell Line (Hl60), A. Lale Doğan

Turkish Journal of Medical Sciences

Fludarabine is an inhibitor of DNA/RNA synthesis with antitumor activity on lymphoproliferative malignancies. We investigated the dose-dependent cytotoxic effects of the drug on human peripheral blood mononuclear cells, the HL60 cell line and the differentiated HL60 cells. Dimethylformamide was used for the neutrophilic differentiation of HL60 cells and the MTT assay was used to estimate drug cytotoxicity. Undifferentiated tumor cells had higher proliferation rates compared to the differentiated subline. There was 57% cell death in HL60 cells at a dose of 5 µg/ml of the drug. Recovery Of 1-Chloro-2,4-Dinitrobenzene Detoxification By N-Acetyl- L-Cysteine In Glutathione Predepleted Human Erythrocytes, Deni̇z Yildiz, C.Işil Kuran

Turkish Journal of Medical Sciences

Glutathione is an important thiol-containing compound involved in the detoxification process in erythrocytes. Its thiol group reacts with a variety of xenobiotics in a glutathione S-transferase catalyzed reaction to form conjugates that are effluxed from the erythrocytes by an ATP dependent transport mechanism. A well studied experimental system is the transport of the conjugate of glutathione and 1-chloro-2,4-dinitrobenzene. We investigated whether N-acetyl-L-cysteine protects the free-SH content and restores 1-chloro-2,4-dinitrobenzene detoxification in erythrocytes or replaces glutathione in detoxification process in glutathione predepleted erythrocytes. Norfloxacin is a poorly water soluble drug, and to improve its solubility it was complexed with b-cyclodextrin (BCD). Several ocular patches/inserts of norfloxacin-b-cyclodextrin were prepared in hydroxypropyl methyl cellulose (HPMC) matrix. The influence of rate controlling membranes made of ethyl cellulose (EC) alone and in combination with polyvinyl pyrrolidone K30 (PVP K30) in different proportions on drug release kinetics was studied. The data were subjected to regression analysis. Various physical characteristics of the films were evaluated. In vitro release studies were carried out in a fabricated flow through cell. Anterior openbite (AOB) is a malocclusion considered to be associated with symptoms and signs of temporomandibular disorders (TMD). An accurate, objective and repeatable method is needed in order to clarify the relationship between TMD and anterior openbite. The aim of this study is to examine the functions of the stomatognathic systems of 21 adolescents with AOB by means of electrovibratography (EVG), electrognathography (EMG), electroganthography (EGN), and transcranial and cephalometric radiographs. Nonconcentricity and mild asymmetry of the condyle fossa relationships were observed on transcranial radiographs. The Effects Of Melatonin On 6-Phosphogluconate Dehydrogenase: An In Vitro And In Vivo Study, İlhami̇ Gülçi̇n, Şükrü Beydemi̇r

Melatonin is known to influence a variety of biological processes including circadian rhythms, neuroendocrine, and cardiovascular and immune functions as well as thermoregulation. Melatonin is the chief secretory product of the pineal gland, although it is also produced in other organs. In this study, in vitro effects of melatonin on 6-phosphogluconate dehydrogenase from human erythrocytes and in vivo effects of melatonin on 6-phosphogluconate dehydrogenase from rat (Sprague-Dawley) erythrocytes were studied. Human erythrocyte 6-phosphogluconate dehydrogenase was purified in 3 steps, namely haemolysate preparation, ammonium sulphate fractionation (35-65%) and 2´,5´-ADP Sepharose-4B affinity gel chromatography. 6-Phosphogluconate dehydrogenase was purified with a recovery rate …

We investigated the morphometric characteristics of the acetabulum, the ligament of the femoral head (LFH) and the femoral head, and the relationships among them in 30 spontaneously aborted human fetuses. The maximum length and width of the LFH, the transverse and vertical diameters of the femoral head and the acetabulum, the height of the femoral head and the depth of the acetabulum were measured and the existence of any relationship among them was assessed statistically. The aim of this study was to determine the serum adenosine deaminase ( ADA ) levels in patients with brucellosis and compare them with the results of healthy individuals. Forty- eight subject were enrolled in this study : 34 patients with brucellosis and 14 healthy individuals. Serum ADA activity was assessed by spectrophotometer in patients with brucellosis and statistically compared with that of healthy individuals. A normal range of serum ADA was considered 5-35 IU/l. Chromosomal Aberrations In Radiation Waste Repository Workers Detected By Fish Painting And Giemsa Staining, Petya Atanasova, Valeria Borisova Hadjidekova, Savina Agova, Milen Iovtchev

We aimed to assess chromosomal aberration frequency by means of FISH chromosome painting and conventional Giemsa staining in peripheral blood lymphocytes of workers at a nuclear waste repository. The results obtained from the exposed group were compared with those from a control group from the administrative staff of the company. Data on health and social status were collected using a questionnaire. Conventional chromosomal analysis after Giemsa staining did not indicate any significant difference between radiation exposed and control workers. Analysis after FISH chromosome painting revealed a 2-fold increased in genomic frequency of stable chromosomal translocations in the exposed group over …
