Penelitian ini dilakukan untuk mengkaji pengaruh pemberian molibdenum dan inokulasi rhizobium terhadap berat akar, berat nodul efektif dan hasil hijauan legum. Rancangan percobaan menggunakan Rancangan Acak Lengkap (RAL) pola faktorial 3 faktor (4 x 2 x 2) dengan 2 kali ulangan. Penelitian bertujuan untuk mengetahui respon broiler terhadap pemberian ransum mengandung dedak padi hasil fermentasi oleh Aspergillus ficuum yang diukur melalui konsumsi ransum, pertambahan bobot badan, konversi ransum, persentase karkas dan imbangan efisiensi protein. Penelitian menggunakan 300 ekor ayam broiler final stock Arbor Acres yang dibagi ke dalam 30 plot. Ransum disusun iso-protein dan iso-energi dengan kandungan protein kasar 22% dan energi metabolis 3100 kkal/kg. Penelitian bertujuan untuk mengetahui fermentabilitas dan kecernaan ransum yang mengandung berbagai kompleks minyak-mineral in vitro. Yeasts merupakan mikroba potensial untuk dikembangkan sebagai penghasil komponen bioaktif seperti enzim, antimicrobial agent sebagai biopreservasi, serta pengahasil pewarna makanan (pigment). Penelitian merupakan studi ekploratif potensi yeasts yang diisolasi dari daging ayam dan sapi. Ditemukan isolate yeasts yang memiliki karakteristik spesifik dengan warna merah. Jumlah total yeasts pada daging ayam 2.105 cfu/gram sedangkan pada daging sapi 1.105 cfu/gram. Hasil pemurnian isolate diperoleh dua isolate yang memiliki kemampuan daya hambat yang terhadap Pseudomonas aerugenes dan Staphylococus aureus serta tidak meiliki aktivitas penghambatan terhadap Escherichia coli dan Candida albicans. pH akhir media selama fermentasi 25 0C, 48 jam adalah 5,5 dan 5,6 …

Penelitian ini bertujuan untuk mengetahui pengaruh penambahan jus buah mangga dan apel terhadap keasaman, total bakteri dan sineresis stirred yoghurt mangga dan apel yang dihasilkan dan selama proses penyimpanan. Penelitian ini dilakukan dengan tiga perlakuan yaitu stirred yoghurt mangga, stirred yoghurt apel dan kontrol. Stirred yoghurt yang dihasilkan disimpan selama 7 hari. Hasil penelitian menunjukkan stirred yoghurt mangga (210 menit, pH=4,3) dan apel (210 menit, pH=4,2) terkoagulasi lebih cepat dan pada pH yang lebih rendah dibandingkan dengan control (250 menit, pH=4,4). Penelitian ini dilaksanakan di laboratorium Mikrobiologi dan Pengolahan Limbah Fakultas Peternakan universitas padjadjaran. Tujuan penelitian ini adalah untuk mengetahui jumlah total bakteri dan koliform pada lumpur hasil ikutan pembentukan gasbio dari feses sapi perah. Penelitian dilakukan dengan metode eksplorasi pada peternak sapi perah yang membuat instalasi gasbio di desa Haurngombong, Kecamatan Pamulihan-Sumedang. Hasil penelitian menunjukkan jumlah bakteri total adalah 41,82 x 1012 cfu/ml lumpur, sedangkan jumlah 8,23 MPN/ml lumpur. Kata kunci : bakteri total, koliform, lumpur, gasbio, feses sapi perah.

Usaha perbibitan pada peternakan sapi perah memegang peranan penting dalam penyediaan stock bibit sapi perah. Dalam kerangka budidaya sapi perah, perbibitan atau breeding merupakan bagian yang tidak terpisahkan dari siklus feeding, breeding, dan management. Ke tiga faktor tersebut sangat penting dalam usahaternak sapi perah. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan berbagai tingkat asam sulfat (H2So4) dan asam formiat (HCOOH) pada proses pikel terhadap kualitas kulit jadi (leather) Domba Garut. Kualitas Kulit jadi didasarkan pada kadar keasaman (pH), kadar krom (Cr2O3), dan kadar air (H2O). Penelitian ini dilakukan secara eksperimen menggunakan pola faktorial dengan rancangan dasar rancangan acak lengkap, menggunakan 48 lembar kulit Domba Garut jantan ukuran 90. Penelitian ini bertujuan mempelajari pengaruh suplemen Zn-Proteinat, Cu-Proteinat dan Ca-Minyak dalam ransum berbasis pucuk tebu diamoniasi terhadap pertumbuhan dan efisiensi ransum pada domba jantan persilangan Priangan x Barbados. Percobaan dilakukan terhadap 5 ekor domba jantan yang sedang tumbuh dengan menggunakan rancangan Bujur Sangkar Latin (BSL) 5x5, lima macam ransum yaitu R1 = campuran 50% pucuk tebu amoniasi + 50% konsentrat, R2 = R1 + 2% Zn-proteinat, R3 = R1 + 2% Cu-proteinat, R4 = R2 + 2% Cu-proteinat, dan R5 = R4 + Ca-minyak ikan. Penelitian ini bertujuan untuk mempelajari potensi pakan asal limbah tanaman pangan dan daya dukungnya terhadap populasi ternak ruminansia di wilayah kabupaten Garut. Penelitian dilakukan dengan cara survey dan pengumpulan data sekunder. Hasil penelitian menunjukkan bahwa Potensi pakan asal rumput alam dan jerami asal limbah tanaman pangan di seluruh wilayah kabupaten Garut adalah 414.211,1 ton bahan kering (BK) yang terdiri atas 151.395,6 ton BK asal rumput dan 262.815,5 ton BK asal limbah tanaman pangan. Daya dukung pakan terhadap populasi ternak adalah 124.706 satuan ternak (ST), yang terdiri atas 45.580,5 ST ternak ruminansia kecil dan 79.125,5 ST ternak ruminansia besar.

Penelitian ini bertujuan untuk mempelajari potensi pakan asal limbah tanaman pangan dan daya dukungnya terhadap populasi ternak ruminansia di wilayah kabupaten Sumedang. Penelitian dilakukan dengan cara survey dan pengumpulan data sekunder. Hasil penelitian menunjukkan bahwa Potensi pakan asal rumput alam dan jerami asal limbah tanaman pangan di seluruh wilayah kabupaten Sumedang adalah 284.784,72 ton bahan kering (BK) yang terdiri atas 86.937,65 ton BK asal rumput dan 197.847,07 ton BK asal limbah tanaman pangan. Tujuan penelitian untuk mengetahui suhu, waktu dan ratio volume air yang tepat selama pemanasan sampai terbentuknya gelatinisasi. Cangkang rajungan mengandung kitin yang persentasenya paling besar berupa polisakarida. Kitin dan derivat-derivatnya telah banyak digunakan dalam berbagai keperluan terutama untuk kesehatan. Penelitian ini dilakukan dengan tujuan untuk mengetahui pengaruh pemberian tepung rajungan sebagai sumber kitin terhadap efisiensi pakan dan kandungan lemak feces tikus putih Rattus norvegicus) strain Wistar. Penelitian menggunakan rancangan acak lengkap pola faktorial 2x5 dengan tiga kali ulangan. Faktor pertama adalah jenis kelamin dan kedua level tepung rajungan yaitu 0%, 5%, 10%, 15%, and 20%. Tujuan penelitian ini untuk mengetahui sejauh mana pengaruh lama penyimpanan semen cair ayam buras pada suhu 50C terhadap periode fertil dan fertilitas sperma. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan setiap perlakuan diulang 6 kali. Lama penyimpanan yang digunakan dalam penelitian ini adalah penyimpanan 1 jam (0 hari), 24 jam (1 hari) dan 48 jam (2 hari). Peubah yang diukur adalah periode fertil dan fertilitas sperma. Myeloid cells are highly heterogeneous and play key roles in establishing an immunosuppressive tumor microenvironment (TME), but the mechanisms controlling their differentiation remain unclear. Using orthotopic murine breast cancer models, we investigated how interleukin-1α (IL-1α) influences this process. Deletion of host Il1α resulted in rejection of transplanted tumors. Single-cell RNA sequencing identified CX3CR1⁺ macrophages as the main source of IL-1α in the TME. Il1α deficiency disrupted monocyte–macrophage differentiation, reducing CX3CR1⁺ macrophages and increasing iNOS⁺ macrophages. Myeloid cells from Il1α⁻/⁻ hosts induced lower PD-1 and CTLA-4 expression in co-cultured CD8⁺ T cells than those from wild-type hosts. CellChat analysis showed …

Forkhead box class O1 (FOXO1) and fas-associated death domain (FADD) regulate cell death pathways and homeostatic processes such as cell cycle progression and apoptosis. FADD phosphorylation promotes nuclear localization of FOXO1, and FOXO1 regulates FADD expression. Therefore, it is plausible that FOXO1 and FADD have synergistic or antagonistic effects on cell cycle regulation and the response to anticancer drug treatment in cancer cells. In the present study, we report that AS1842856-mediated inhibition of FOXO1 reverses anticancer drug-induced cytotoxicity, while FADD knockdown increases anticancer drug-induced cytotoxicity in osteosarcoma (OS). Serious mental illnesses (SMIs), including major depressive disorder, bipolar disorder, and schizophrenia, have long been linked to cognitive decline, multiple chronic medical conditions, and premature mortality. These factors significantly contribute to the severe disability seen in SMIs, extending beyond the severity of psychopathology and indicating a premature aging phenotype associated with these conditions. The mechanisms that underlie the relationship between SMIs and the premature aging phenotype are not well understood, but recent evidence suggests that individuals with SMIs may exhibit accelerated biological aging. Purpose: This study examined the effects of sustained, isometric forearm flexion tasks anchored to ratings of perceived exertion of 2 (RPE2FT) versus 8 (RPE8FT) on the wavelet-based mechanomyographic (MMG) intensity responses from pretest and posttest maximal voluntary isometric contractions (MVICs) as well as the time course of changes in the MMG intensity responses throughout the sustained tasks. Reproductive medicine providers face ethical dilemmas often in their field, only some of which have standards of care. There are questions in the medical landscape pertaining to physician autonomy versus patient autonomy, which heavily apply to reproductive medicine providers. Patient autonomy is highly valued in medical ethics; however, physician autonomy should also be considered. GLUT1 facilitates a continuous flow of glucose across the inner and outer blood-retinal barriers (BRBs) to support vision. To understand the extent to which photoreceptors rely on glucose transport across the outer BRB, we generated a tamoxifen-inducible conditional knockout of Slc2a1 in the retinal pigment epithelium (RPE) (RPE-iΔGlut1). In the RPE-iΔGlut1 mice, rod photoreceptors exhibited impaired outer segment renewal and decreased the expression of proteins involved in phototransduction and ciliary transport. Proteins regulating the retinal stress response increased. Cone photoreceptors were functional and viable 15 months post-tamoxifen treatment in the RPE-iΔGlut1 mice, while 70% of the rods died. …

Sensory perception is shaped by experience, giving stimuli behavioral significance. Basal forebrain (BF) cholinergic neurons in mice, which are crucial for arousal and motivation, also regulate sensory processing. Within BF nuclei, glutamatergic (vGlut2BF) neurons receive cholinergic input and modulate behaviors, but their roles in encoding sensory significance remain unclear. Using in vivo calcium imaging, we found that vGlut2BF neurons initially poorly encoded odor identity. However, their response to conditioned odors increased following associative learning, and their population activity more distinctly encoded paired stimuli, reflecting emergent value representation. Introduction: Effective therapies are needed for patients with NSCLC with HER2-mutant tumors who progress on the HER2 antibody-drug conjugate trastuzumab deruxtecan (T-DXd), a standard-of-care treatment. A greater understanding of mechanisms mediating acquired T-DXd resistance and whether these tumors could benefit from HER2 tyrosine kinase inhibitors (TKIs) is needed.
Methods: Using preclinical models of acquired T-DXd resistance, LentiMutate scanning mutagenesis, and clinical analyses, we investigated mechanisms mediating acquired resistance to T-DXd and assessed the impact of each of these resistance mechanisms on cross-resistance to alternative HER2-targeting approaches.

Background: KMT2A partial tandem duplication (PTD) occurs in approximately 5-10% of acute myeloid leukemia (AML) cases and is associated with poor prognosis. While its cytogenetic and molecular features are well described, the immunophenotypic characteristics of AML with KMT2A-PTD remain incompletely defined.
Methods: We identified 47 cases of AML with KMT2A-PTD by optical genome mapping. All cases underwent flow cytometric immunophenotypic analysis and next-generation sequencing using an 81-gene panel.
Results: The cohort included 32 men and 15 women with a median age of 67 years (range, 19-87). Thirty-eight cases were de novo AML, and nine were secondary to myelodysplastic …

Epithelial-to-mesenchymal transition (EMT) is known to induce both stemness and mesenchymal properties, and our findings reveal that these two programs can be uncoupled. During EMT, epithelial cells transition from symmetric divisions producing differentiated daughter cells to self-renewing daughter cells. When we block cell division and induce EMT, cells gain mesenchymal properties but not stemness, suggesting the importance of cell division for gaining stemness. We identified ESRP1 as a key regulator of EMT-driven stemness, which get downregulated during EMT in a cell division-dependent manner. Overexpression of ESRP1 prevents the gain of stemness without affecting the mesenchymal program. Only the stemness and …

Patients with poorly differentiated thyroid cancer (PDTC) and anaplastic thyroid cancer (ATC) face a much poorer prognosis than those with differentiated thyroid cancers. Around 25% of PDTCs and 35% of ATCs carry the BRAFV600E mutation, which constitutively activates the MAPK pathway, a key driver of cell growth. Although combining BRAF and MEK inhibitors can shrink tumors, resistance often develops. The exact cause of this resistance remains unclear. Human epidermal growth factor receptor 2 (HER2) is an established therapeutic target in multiple solid tumors, particularly breast and gastric cancers. Significant advancements have been made in the development of HER2-targeted therapies, including monoclonal antibodies, tyrosine kinase inhibitors, antibody-drug conjugates (ADC), and novel bispecific antibodies. These agents have revolutionized the treatment landscape for HER2-positive metastatic cancers, resulting in improved progression-free and overall survival, and quality of life for patients. Acute myeloid leukemia (AML) is a clonal disorder characterized by immature blasts and arrested differentiation that primarily affects the bone marrow (BM) and occasionally presents as extramedullary (EM) disease. EM manifestations highlight AML's adaptability to distinct microenvironments, which we examined using spatial analyses of medullary and EM tissues. We describe a workflow for Visium-based spatial transcriptomics in medullary and EM AML, revealing insights into cell-cell communication and the spatial organization of AML hierarchies. In BM, monocytes and granulocyte-monocyte progenitors colocalized with leukemic populations, sharing molecular signatures with those in EM sample. CXCL12-CXCR4-mediated communication correlated with PI3K/AKT/mTOR signaling in inflammatory niches.

In this Opinions in the Profession article, we explore remediation treatment strategies using exercise for recovery from post-traumatic brain injury and aim to justify the use of such treatment in the occupational therapy profession to increase skills for functional gain. Common treatment strategies used by occupational therapists after brain injury are compensatory strategies, indicating the client learns a new way to perform the task. Though effective, it is also necessary to consider how remediation and neuroplasticity are essential. Despite considerable advances in identifying risk factors for obesity, gaps remain in our understanding about its etiology. Genetic variants explain only a small portion of variation in obesity-related traits such as body mass index (BMI). Epigenetic regulation, which controls gene expression and is influenced by environmental and genetic factors, may account for additional variability in BMI. Epigenetic studies of BMI have largely been conducted in European ancestry populations, despite the disproportionate burden of obesity in African Americans (AAs).
