Role Of Sec61alpha In The Regulated Transfer Of The Ribosome-Nascent Chain Complex From The Signal Recognition Particle To The Translocation Channel, Weiqun Song, David Raden, Elisabet Mandon, Reid Gilmore

Elisabet Mandon

Targeting of ribosome-nascent chain complexes to the translocon in the endoplasmic reticulum is mediated by the concerted action of the signal recognition particle (SRP) and the SRP receptor (SR). Ribosome-stripped microsomes were digested with proteases to sever cytoplasmic domains of SRalpha, SRbeta, TRAM, and the Sec61 complex. We characterized protein translocation intermediates that accumulate when Sec61alpha or SRbeta is inactivated by proteolysis. In the absence of a functional Sec61 complex, dissociation of SRP54 from the signal sequence is blocked. Experiments using SR proteoliposomes confirmed the assembly of a membrane-bound posttargeting intermediate. These results strongly suggest that the Sec61 complex regulates …

Samples with titers of =>1:320 were considered positive. The rate of seroprevalence of antibodies was 88%. The increase in the rate of yearly seroprevalence of Borrelia burgdorferi antibodies in dogs was …

A Loop-Mediated Isothermal Amplification (Lamp) Assay For Rapid Detection Of Anaplasma Phagocytophilum Infection In Dogs, Chungchen Lee, Yungchang Lin, Chauloong Tsang, Yangtsung Chung Jan 2012

A Loop-Mediated Isothermal Amplification (Lamp) Assay For Rapid Detection Of Anaplasma Phagocytophilum Infection In Dogs, Chungchen Lee, Yungchang Lin, Chauloong Tsang, Yangtsung Chung

Turkish Journal of Veterinary & Animal Sciences

A simple and rapid diagnostic method of Anaplasma phagocytophilum infection was established, employing the loop-mediated isothermal amplification (LAMP) reaction. With 1 set of 4 specific primers targeting the citrate synthase (gltA) gene of A. phagocytophilum, the LAMP assay rapidly amplified the target DNA sequence in a single tube in 1 h, requiring only a water bath for reaction. When visualized by gel electrophoresis, the LAMP products appeared as a ladder-like pattern, with many bands of different sizes from 180 base pairs up to the loading well. The results obtained from testing 32 clinical blood samples of dogs demonstrated that the …

Methicillin Resistance In Staphylococcus Pseudintermedius, Chad Christopher Black Aug 2010

Methicillin Resistance In Staphylococcus Pseudintermedius, Chad Christopher Black

Doctoral Dissertations

Staphylococcus pseudintermedius affecting dogs is analogous to S. aureus on humans, acting as both normal flora and opportunistic pathogen. Methicillin resistance in S. pseudintermedius is recent, with the first documented occurrence of an isolate bearing the methicillin resistance gene, mecA, in 1999. This gene encodes penicillin binding protein 2a, which renders all beta-lactam drugs ineffective and functions as a “gateway” antibiotic resistance determinant. In the presence of ineffective antibiotics, opportunities for mutational events and acquisition of mobile genetic elements increase as microbial densities increase, often leading to multi-drug resistance. Methicillin-resistant S. pseudintermedius (MRSP) infections have become increasingly common. For …

The present study provides the results from a comparative study of the 3 commonly used methods for total protein (TP) measurement. The experiments were carried out with 6 dogs (4-7 year-old, weighing 12.8 ± 1.4 kg). Five blood samples were obtained by saphena venepuncture from all dogs, during the time course of the experimentally induced infection with Staphylococcus intermedius, administered subcutaneously at a dose rate of 5 ml of 1.10^9 CFU/ml within 14 days. TP concentration was measured by 2 macro protein techniques - biuret method (commonly used) and method of Lowry, and a modified version of biuret method (micro …

Supervillin (P205): A Novel Membrane-Associated, F-Actin-Binding Protein In The Villin/Gelsolin Superfamily, Kersi N. Pestonjamasp, Robert K. Pope, J. D. Wulfkuhle, Elizabeth J. Luna Mar 2008

Supervillin (P205): A Novel Membrane-Associated, F-Actin-Binding Protein In The Villin/Gelsolin Superfamily, Kersi N. Pestonjamasp, Robert K. Pope, J. D. Wulfkuhle, Elizabeth J. Luna

Elizabeth J. Luna

Actin-binding membrane proteins are involved in both adhesive interactions and motile processes. We report here the purification and initial characterization of p205, a 205-kD protein from bovine neutrophil plasma membranes that binds to the sides of actin filaments in blot overlays. p205 is a tightly bound peripheral membrane protein that cosediments with endogenous actin in sucrose gradients and immunoprecipitates. Amino acid sequences were obtained from SDS-PAGE-purified p205 and used to generate antipeptide antibodies, immunolocalization data, and cDNA sequence information. The intracellular localization of p205 in MDBK cells is a function of cell density and adherence state. In subconfluent cells, p205 …

A Potential Champ? [Distemper In Dogs], F C. Wilkinson

Journal of the Department of Agriculture, Western Australia, Series 4

THE mother of this pup was an eight-year-old sheep dog and in the eyes of her owner she was a champion and well worth her keep.

She was perhaps slowing down a little, so the best looking pup out of the last litter had been kept as a possible replacement.
