Aquaculture is a globally important industry that faces significant challenges due to infectious diseases, which can result in substantial financial losses. In 2011, a new strain of Edwardsiella ictaluri emerged as a major pathogen affecting zebrafish in the ornamental fish industry, leading to high mortality rates and posing a severe threat to the sector. Vaccines provide potential benefits for disease prevention in aquaculture, particularly live-attenuated vaccines (LAVs), which induce strong host immune responses. Bacterial attenuation for vaccine development has evolved from serial passage to more targeted genetic modification techniques. However, regulatory constraints limit the availability of licensed LAVs for aquaculture. …

This is a three-study dissertation in which we aimed to broaden our knowledge of the mechanisms contributing to a 45° clockwise visuomotor adaptation by including variations in workspace. We provide behavioral and in one study physiological outcomes as evidence to support our conclusions. In the first experiment, we observed the adaptation of movement parameters such as pathlength, movement time, resultant velocity, and normalized jerk across groups trained with rotated visual feedback with both the left and right hands. Workspace location and hand differentially affected movement trajectory length. The group that practiced the task with their nondominant, left hand showed larger …

Epilepsy is a complex group of neurological disorders affecting approximately 65 million people worldwide. Animal models reveal the activation of cannabinoid receptor 1 and cannabinoid receptor 2 reduces the severity of seizures associated with epilepsy. Trials of phytocannabinoids in mammals demonstrate their efficacy of reducing the severity of epileptic seizures. Numerous terpenoid compounds are present in Cannabis plants but most have yet to be clinically investigated. This research examined the effect of biodegradable, polymeric, lignin-based nanoparticles (LNPs, 113.8±3.4, negatively charged) and zein nanoparticles (ZNP, 141.6±3.9, positively charged) on soybean plant health. The LNPs were synthesized from lignin, covalently linked to poly(lactic-co-glycolic) acid by emulsion evaporation. ZNPs were synthesized by nanoprecipitation. Soybeans grown hydroponically were treated with three concentrations (0.02, 0.2, and 2 mg/ml) of NPs at 28 days after germination. The neurohypophysial hormone oxytocin is involved in the regulation of social behaviors, including social recognition, pair bonding, and sex-specific parental behaviors in a variety of species. Oxytocin triggers these social behaviors by binding to oxytocin receptors (OXTR) in various parts of the brain. Oxytocin-induced sex-typical behavior, therefore, suggests a sexual dimorphic distribution of OXTR in the brain. Obesity is a global epidemic characterized by an expansion of adipose tissue. Adipose tissue, composed of adipocytes and a stromal vascular fraction, is an endocrine organ that regulates whole body homeostasis. Obesity leads to the dysregulation of adipocytes and is often associated with increased susceptibility to metabolic diseases such as Type II diabetes mellitus (T2DM). Understanding the mechanisms by which adipocyte function and development are regulated is crucial. The primary focus of this dissertation was to understand the motor control strategy used by our neuromuscular system for the multi-layered motor tasks involved during smartphone manipulation. To understand this control strategy, we recorded the kinematics and multi-muscle activation pattern of the right limb during smartphone manipulation, including grasping with/out tapping, movement conditions (MCOND), and arm heights.

The Obstetrical Dilemma (OD) theory has become canon in biological anthropology. The OD posits that i] dystocia results from bipedal mothers and encephalized infants, ii] contrasting selection for bipedality and obstetrics hinders locomotive efficiency, and iii] the contradicting requirement of the fetus being small enough to pass through the birth canal yet being cognitively advanced enough to cling to its mother after birth. Females, theoretically, exhibit deficient gait efficiency for the sake of successful childbirth. As of 2018, the United States National Institutes of Health estimate that over half a billion people worldwide are affected by autoimmune disorders. Though these conditions are prevalent, treatment options remain relatively poor, relying primarily on various forms of immunosuppression which carry potentially severe side effects and often lose effectiveness overtime. Given this, new forms of therapy are needed. We propose small-interfering RNA (siRNA) for hypervariable regions of the T-cell receptor β-chain gene (TCRb) as a highly targeted, novel means of therapy for the treatment of autoimmune disorders.

Rickettsia felis is the etiologic agent of flea-borne spotted fever (FBSF) in humans and a poorly described cause of fever in animals. It is transmitted by its primary arthropod vector and reservoir host, the cat flea Ctenocephalides felis. Known routes of Rickettsia felis transmission between Rickettsia felis-infected cat fleas and vertebrate hosts include cutaneous bites and contamination of cutaneous wounds with infective flea feces. The Janus Kinase - Signal Transducer and Activator of Transcription (JAK-STAT) signaling pathway transduces several signals crucial for development and homeostasis. Suppressors of cytokine signaling (SOCS) proteins control JAK-STAT signaling via a negative feedback loop. The transcription factor STAT5 is known to play a significant role in fat cell development and function, and several studies suggest that acetylation may affect STAT5 transcriptional activity. Soil-transmitted helminth (STH) infections impact billions of people worldwide. The traditional STH control approach is a morbidity control strategy implementing mass drug administration (MDA) programs targeting school-aged children (SAC). In Brazil, this control strategy has decreased STH prevalence to less than 20% in most of the country and providing an opportunity to transition from the morbidity control program and towards a surveillance and response system geared towards STH elimination. From within their ecologic niches, zoonotic viruses emerge from animal reservoirs into the edges and centers of human habitation to exploit opportunities for unabated transmission within immunologically–naïve populations. Our understanding of where, in whom, and how these viruses emerge is under direct challenge, driving the evolution of modern infectious disease epidemiology within a rapidly-connected global community. The objectives of these studies were to validate and confirm the structure of a previously developed questionnaire that measures the psychosocial constructs defined by the Self-Determination Theory (SDT) and then test this questionnaire in a nutrition education setting. The instrument, titled the Motivation to Prepare Healthy Foods Questionnaire (MPHFQ), included items that measured intrinsic motivation, perceived competence, relatedness, autonomy, and autonomy support. These studies evaluated the questionnaire’s validity and ability to measure changes in the SDT construct scores.

Melanoma accounts for 90% of skin cancer-related deaths in humans. Treatment options for metastatic melanoma in people is very limited. Melanoma is considered to be an immunogenic tumor, spurring interest in development of immunotherapies for the treatment of metastatic melanoma. Oncolytic virotherapy has been widely investigated. The first ever oncolytic virotherapy to receive FDA-approval is an HSV-1-based virus (Talimogene Laherperavec (T-Vec) or Imlygic) containing a transgene for human GM-CSF to enhance anti-tumor immune responses after injection. Amongst the nutrients present in HBM, the long-chain polyunsaturated fatty acids (LCPUFAs) are of high importance due to the pivotal role they play in infant cognitive and visual development, and growth. PURPOSE: To explore the hematological differences in African-American athletes with sickle cell trait (SCT) and healthy controls (CON) before preseason camp and the changes that occur after a full season. METHODS: Sixteen (n=8 SCT; n=8 position matched, CON) NCAA Division 1 athletes (12 football, 2 each women’s soccer and volleyball) had blood drawn before preseason camp, after and at the end of the season, and analyzed for Hb electrophoresis (Hb-A, Hb-A2, Hb-S, and Hb-F), complete blood count with differential, chemistry panel 26, and prothrombin time, activity and international normalized ratio (INR). Herpes simplex Typ-1 (HSV-1) virions utilize the microtubular network for intracellular transport during both virus entry and virus egress from infected cells. Spotted Fever Group Rickettsia are inoculated into the mammalian host during hematophagous arthropod feeding. Once in the bloodstream and during dissemination, the survival of these pathogens is dependent upon their ability to evade innate host defenses until a proper cellular target is reached. The establishment of a successful infection also relies on the ability of the bacteria to attach and invade target cells, as failure to do so results in destruction of the bacterium. Age-related macular degeneration (AMD) is one of the leading causes of blindness among the elderly worldwide. Retinal problems, specifically in the macula, hold potential for development of AMD. Macular pigment optical density (MPOD) gives a measure of the thickness of the macula and therefore health of the macula. Some of the populations at risk for development of AMD include being white, female, and having light eye color. Factors related to the development of the disease have been divided into modifiable and non-modifiable; non-modifiable include a genetic predisposition.
