PURPOSE: Healthy people with a family history (FH+) of type 2 diabetes (T2D) display impaired metabolic and microvascular function prior to glucose intolerance, and are at greater risk for developing T2D. While mechanisms to explain this disparity are lacking, it is possible that intestinal permeability plays a role, as it is also linked with insulin resistance, glucose intolerance, and chronic inflammation. Lipopolysaccharides (LPS) act as an outer membrane component of gram-negative bacteria in intestines and play a role in inflammation and chronic disease when in circulation, thus serving as a surrogate measure of intestinal permeability. PURPOSE: The primary objective of the present study was to determine the role of physical activity in the relationship between sitting time and insulin resistance in 7,092 randomly selected U.S. adults. METHODS: Cross-sectional data from the National Health and Nutrition Examination Survey (NHANES), 2011-2018, were used to answer the research question. NHANES data for 2019-2020 were not available because of COVID. Participants were selected using a multi-stage, random sampling strategy. Subjects reported average sitting time, quantified as the number of minutes spent per day sitting. Insulin resistance was determined using HOMA-IR (homeostatic model assessment). Obesity and type 2 diabetes are associated with impaired glucose homeostasis and blood lipid profiles. Further, a family history of diabetes (FH) increases the risk for development of insulin resistance. However, it is unclear whether a FH impairs glucose tolerance, blood glucose and lipid profiles in young, healthy, normoglycemic adults. PURPOSE: To investigate whether a FH impairs glucose tolerance and blood lipid profile in healthy, sedentary Hispanic males.
