Storage roots of cassava (Manihot esculenta Crantz), a major subsistence crop of sub-Saharan Africa, are calorie rich but deficient in essential micronutrients, including provitamin A β-carotene. In this study, β-carotene concentrations in cassava storage roots were enhanced by coexpression of transgenes for deoxy-d-xylulose-5-phosphate synthase (DXS) and bacterial phytoene synthase (crtB), mediated by the patatin-type 1 promoter. Storage roots harvested from field-grown plants accumulated carotenoids to ≤50 lg/g DW, 15- to 20-fold increases relative to roots from nontransgenic plants. Approximately 85%–90% of these carotenoids accumulated as all-trans-β-carotene, the most nutritionally efficacious carotenoid. β-Carotene-accumulating storage roots displayed delayed onset of postharvest …

Development And Characterization Of Rice Genotypes For Water Use Efficiency And Drought Resistance, Anuj Kumar
Development And Characterization Of Rice Genotypes For Water Use Efficiency And Drought Resistance, Anuj Kumar
Graduate Theses and Dissertations
Rice, the second largest staple food crop, uses 30% of global fresh water to complete its life cycle worldwide. Water deficits worldwide have become a serious problem affecting rice growth and ultimately grain yield. To solve the water problem globally, improvement of water use efficiency (WUE) and other drought resistance (DR) traits in rice genotypes would be a useful strategy using advanced genomics tools. In this study, our objectives were to 1) analyze phenotypes of the USDA rice mini-core collection (URMC) for WUE and DR related traits, 2) correlate drought response phenes for physiological traits and grain yield in the …
Development And Evaluation Of Environmentally-Sensitive Genetic Male Sterile Lines For Louisiana Hybrid Rice, Federico Molina Casella
Development And Evaluation Of Environmentally-Sensitive Genetic Male Sterile Lines For Louisiana Hybrid Rice, Federico Molina Casella
LSU Doctoral Dissertations
The Louisiana rice industry generates ~ 400 million dollars each year toward our state economy. Louisiana varieties are typically bred as pure lines, but hybrid (F1) varieties have the potential to increase grain yields 15% or more over conventional sources. A primary goal of the LSU AgCenter Hybrid Breeding program is to breed elite cross combinations (hybrid varieties) through development of environmentally sensitive male sterile and fertile pollinator lines. Extensive multi-location yield trials in Louisiana have demonstrated high grain and head rice yields for candidate hybrids, but improvements in maturity, lodging, and grain chalk are needed. To address …

Inheritance Study Of Resistance To The Herbicide Quizalofop-P-Ethyl In Rice And Development Of Provisia Hybrid Rice Germplasm, Jose Roberto Camacho Montero
LSU Doctoral Dissertations
In 2017, ProvisiaTM quizalofop-p-ethyl (QFE) resistant rice became a new alternative for the chemical control of in-season red rice (Oryza sativa L. sp. spontaneum) infestations in Louisiana rice farms. Until then, Clearfield® imidazolinone resistant rice was the only alternative that US farmers had for the efficient in-season chemical control of red rice. ProvisiaTM rice is a BASF Corporation patented technology that can serve as an alternative for rotation with Clearfield® rice. This strategy seeks to prevent outcrosses between red rice and herbicide resistant varieties, and consequently extend the life span of both technologies. This …

Tan spot (syn. yellow spot and yellow leaf spot) caused by Pyrenophora tritici-repentis is an important foliar disease of wheat in Australia. Few resistance genes have been mapped in Australian germplasm and only one, known as tsn1 located on chromosome 5B, is known in Australian breeding programs. This gene confers insensitivity to the fungal effector ToxA. The main aim of this study was to map novel resistance loci in two populations: Calingiri/Wyalkatchem, which is fixed for the ToxA-insensitivity allele tsn1, and IGW2574/Annuello, which is fixed for the ToxA-sensitivity allele Tsn1. A second aim was to combine …

The Japanese chestnut (Castanea crenata) carries resistance to Phytophthora cinnamomi, the destructive and widespread oomycete causing ink disease. The European chestnut (Castanea sativa), carrying little to no disease resistance, is currently threatened by the presence of the oomycete pathogen in forests, orchards and nurseries. Determining the genetic basis of P. cinnamomi resistance, for further selection of molecular markers and candidate genes, is a prominent issue for implementation of marker assisted selection in the breeding programs for resistance. In this study, the first interspecific genetic linkage map of C. sativa x C. crenata …

Sinorhizobium fredii is a fast-growing rhizobial species that can establish a nitrogen-fixing symbiosis with a wide range of legume species including soybeans (Glycine max). In soybeans, this interaction shows a high level of specificity such that particular S. fredii strains nodulate only a limited set of plant genotypes. Here we report the identification of a dominant gene in soybeans that restricts nodulation with S. fredii USDA193. Genetic mapping in an F2 population revealed co-segregation of the underlying locus with the previously cloned Rfg1 gene. The Rfg1 allele encodes a member of the Toll-interleukin receptor/nucleotide-binding site/leucine-rich repeat class of …

Obtaining corn hybrid seeds (Zea mays L.) with high vigour depends on the parental lines and the direction of the cross, and this relates to seed desiccation tolerance and composition. This research studied reciprocal crosses between pairs of proprietary, elite parent lines (L1 and L5; L2 and L4) producing hybrid seeds with different qualities attempting to correlate vigour with seed composition, focusing on storage proteins, starch and soluble sugar amounts. Four corn hybrid seed lots produced from reciprocal crosses were compared (HS 15 with HS 51, and HS 24 with HS 42) by assessing germination, vigour, and seedling emergence …

Identification Of Hydrated And Dehydrated Lipids And Protein Secondary Structures In Seeds Of Cotton (Gossypium Hirsutum) Line, Diwas Kumar Silwal, Nsoki Phambu, Bharat Pokharel, Ahmad Naseer Aziz
Chemistry Faculty Research
Cottonseeds from two parents (TM-1 and 3-79) and their 17 progeny (chromosomal substitution) lines were analyzed for various secondary structures of proteins and moisture content of lipids, separately in hulls and kernels. Fourier transform infrared spectroscopy (FTIR) was used on mature seeds from Upland cotton (G. hirsutum) progeny lines and parents. Based on secondary structures of proteins and hydration levels of lipids, differences were observed among the cottonseeds. The two progeny lines – CS-B12sh and CS-B22sh retained lipid moisture content and protein secondary structures similar to both parents, while CS-B06, CS-B15sh and CS-B16 remained distinct from either parent. On the …

Production of high quality seeds is of fundamental importance for successful crop production. However, knowledge of the effects of increased temperature resulting from global warming on seed quality of alpine species is limited. We investigated the effect of maternal environment on seed quality of three cultivars of the leguminous forage species Vicia sativa, giving particular attention to temperature. Plants of each cultivar were grown at 1700 and 3000 m a.s.l., and mass, germination, electrical conductivity (EC) of leakage and longevity were determined for mature seeds. Seeds of all three cultivars produced at the low elevation had a significantly lower …

Identifying interspecies changes in gene regulation, one of the two primary sources of phenotypic variation, is challenging on a genome-wide scale. The use of paired time-course data on cold-responsive gene expression in maize (Zea mays) and sorghum (Sorghum bicolor) allowed us to identify differentially regulated orthologs. While the majority of cold-responsive transcriptional regulation of conserved gene pairs is species specific, the initial transcriptional responses to cold appear to be more conserved than later responses. In maize, the promoters of genes with conserved transcriptional responses to cold tend to contain more micrococcal nuclease hypersensitive sites in their …
Genomic And Physiological Approaches To Improve Drought Tolerance In Soybean, Avjinder Kaler
Genomic And Physiological Approaches To Improve Drought Tolerance In Soybean, Avjinder Kaler
Graduate Theses and Dissertations
Drought stress is a major global constraint for crop production, and improving crop tolerance to drought is of critical importance. Direct selection of drought tolerance among genotypes for yield is limited because of low heritability, polygenic control, epistasis effects, and genotype by environment interactions. Crop physiology can play a major role for improving drought tolerance through the identification of traits associated with drought tolerance that can be used as indirect selection criteria in a breeding program. Carbon isotope ratio (δ13C, associated with water use efficiency), oxygen isotope ratio (δ18O, associated with transpiration), canopy temperature (CT), canopy wilting, and canopy coverage …
Dissecting The Genetic Basis Of Salt Tolerance In Rice (Oryza Sativa), Malachy Thomas Campbell
Dissecting The Genetic Basis Of Salt Tolerance In Rice (Oryza Sativa), Malachy Thomas Campbell
Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research
Excessive salt is a major factor limiting crop productivity. Rice is an important food crop worldwide, and provides a major source of calories for billions of people in tropical and subtropical developing countries. In many regions throughout Southeast Asia, rice is grown in low-lying coastal regions, which are prone to ingress by seawater. Rice is highly sensitive to salt stress, with mild salinity resulting in large reductions in yield. Improving salt tolerance in rice is essential for ensuring food security for many individuals throughput the developing world.
In this work, the overall goals were to characterize a diverse collection of …

Phenotypic Variation And Genetic Purity Of The Original 'Prime-Jim®' X 'Arapaho' Population, Loren M. Luther
Horticulture Undergraduate Honors Theses
In 2013, Castro et al., produced the first linkage map for primocane-fruiting blackberries using the cross ‘Prime-Jim®’ x ‘Arapaho.’ This mapping population has been maintained since the original experiment with the hopes of conducting future studies. Further research was determined to be needed on the population to better characterize additional phenotypic traits, and since nine years had passed since the seedlings were established, there was a need to assess the continued genetic purity of the population using molecular methods. Phenotypic data was collected to analyze the variation of the population including soluble solids content, titratable acidity, average weight, shape, and …

Seed oils of many Cuphea sp. contain .90% of medium-chain fatty acids, such as decanoic acid (10:0). These seed oils, which are among the most compositionally variant in the plant kingdom, arise from specialized fatty acid biosynthetic enzymes and specialized acyltransferases. These include lysophosphatidic acid acyltransferases (LPAT) and diacylglycerol acyltransferases (DGAT) that are required for successive acylation of medium-chain fatty acids in the sn-2 and sn-3 positions of seed triacylglycerols (TAGs). Here we report the identification of a cDNA for a DGAT1-type enzyme, designated CpuDGAT1, from the transcriptome of C. avigera var pulcherrima developing seeds. Microsomes of camelina (Camelina …

High And Low Yielding Soybean Lines From An Irrigated Selection Environment: Performance Evaluation In Irrigated And Droughted Environments, Jorge E. Perez Arocho
Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research
Drought is the most significant abiotic stress affecting agricultural production. Improving seed yield under water stress is an important breeding objective. Soybean cultivars that tolerate water stress could help improve and stabilize production in water-stressed environments. “Drought” needs to be defined, because water stress can vary in intensity, timing, and duration. Other factors including ambient temperature, soil texture, depth, and fertility will influence the impact of limited water on crop productivity. This research addresses the impact of water stress on soybeans adapted to the north-central US, where the majority of soils are relatively deep, fertile silt loams or silty clay …

Soya bean (Glycine max (L.) Merr.) is sought after for both its oil and protein components. Genetic approaches to add value to either component are ongoing efforts in soya bean breeding and molecular biology programmes. The former is the primary vegetable oil consumed in the world. Hence, its primary usage is in direct human consumption. As a means to increase its utility in feed applications, thereby expanding the market of soya bean coproducts, we investigated the simultaneous displacement of marine ingredients in aquafeeds with soya bean-based protein and a high Omega-3 fatty acid soya bean oil, enriched with alpha-linolenic …

Plants pollinated by hummingbirds or bats produce dilute nectars even though these animals prefer more concentrated sugar solutions. This mismatch is an unsolved evolutionary paradox. Here we show that lower quality, or more dilute, nectars evolve when the strength of preferring larger quantities or higher qualities of nectar diminishes as magnitudes of the physical stimuli increase. In a virtual evolution experiment conducted in the tropical rainforest, bats visited computer-automated flowers with simulated genomes that evolved relatively dilute nectars. Simulations replicated this evolution only when value functions, which relate the physical stimuli to subjective sensations, were nonlinear. Selection also depended on …

Background: Perennial growth in plants is the product of interdependent cycles of daily and annual stimuli that induce cycles of growth and dormancy. In conifers, needles are the key perennial organ that integrates daily and seasonal signals from light, temperature, and water availability. To understand the relationship between seasonal cycles and seasonal gene expression responses in conifers, we examined diurnal and circannual needle mRNA accumulation in Douglas-fir (Pseudotsuga menziesii) needles at diurnal and circannual scales. Using mRNA sequencing, we sampled 6.1 × 109 reads from 19 trees and constructed a de novo pan-transcriptome reference that includes 173,882 tree-derived transcripts. Using …

Soybean C3 photosynthesis can suffer a severe loss in efficiency due to photorespiration and the lack of a carbon concentrating mechanism (CCM) such as those present in other plant species or cyanobac-teria. Transgenic soybean (Glycine max cv. Thorne) plants constitutively expressing cyanobacterial ictB (inorganic carbon transporter B) gene were generated using Agrobacterium-mediated transformation. Although more recent data suggest that ictB does not actively transport HCO3-/CO2, there is nevertheless mounting evidence that transformation with this gene can increase higher plant photosynthesis. The hypothesis that expression of the ictB gene would improve photosynthesis, biomass production and seed yield in soybean …

Under nitrogen deprivation, the green alga C. reinhardtii showed substantial triacylglycerol (TAG) accumulation and up-regulation of a gene, GPD2, encoding a multidomain enzyme with a putative phosphoserine phosphatase (PSP) motif fused to glycerol-3-phosphate dehydrogenase (GPD) domains. Canonical GPD enzymes catalyze the synthesis of glycerol-3-phosphate (G3P) by reduction of dihydroxyacetone phosphate (DHAP). G3P forms the backbone of TAGs and membrane glycerolipids and it can be dephosphorylated to yield glycerol, an osmotic stabilizer and compatible …

Predictions suggest that current crop production needs to double by 2050 to meet global food and energy demands. Based on theory and experimental studies, overexpression of the photosynthetic enzyme sedoheptulose-1,7-bisphosphatase (SBPase) is expected to enhance C3 crop photosynthesis and yields. Here we test how expression of the cyanobacterial, bifunctional fructose-1,6/sedoheptulose-1,7-bisphosphatase (FBP/SBPase) affects carbon assimilation and seed yield (SY) in a major crop (soybean, Glycine max). For three growing seasons, wild-type (WT) and FBP/SBPase-expressing (FS) plants were grown in the field under ambient (400 μmol mol−1) and elevated (600 μmol mol−1) CO2 concentrations [CO …

- Few transcription factors have been identified in C4 grasses that either positively or negatively regulate monolignol biosynthesis.
- Previously, the overexpression of SbMyb60 in sorghum (Sorghum bicolor) has been shown to induce monolignol biosynthesis, which leads to elevated lignin deposition and altered cell wall composition. To determine how SbMyb60 overexpression impacts other metabolic pathways, RNA-Seq and metabolite profiling were performed on stalks and leaves.
- 35S::SbMyb60 was associated with the transcriptional activation of genes involved in aromatic amino acid, S-adenosyl methionine (SAM) and folate biosynthetic pathways. The high coexpression values between SbMyb60 and genes assigned to these pathways indicate …
