The Virome Of Peony And The Population Structure Of Its Most Prominent Viruses, Cullen Shaffer Dec 2020

The Virome Of Peony And The Population Structure Of Its Most Prominent Viruses, Cullen Shaffer

Graduate Theses and Dissertations

Peony (Peonia lactiflora, Pall.) is a popular ornamental that has been cultivated for millennia. Due to its popularity, plant material is frequently moved across international borders allowing for the spread of viruses. The virome of several peony plants was investigated and four viruses; namely Amazon lily mild mottle virus (ALiMMV), Cycas necrotic stunt virus (CNSV), Gentian Kobu-sho associated virus (GKaV) and Lychnis mottle virus (LycMoV) were detected for the first time in the Western Hemisphere. Incidence ranged from a few plants for ALiMMV to near universal infection for CNSV. GKaV was found in individuals that were infected with Lemoine’s disease …

Fungicide Resistance: Surveillance, Risk Assessment And Evolution In Two Soil-Borne Pathogens, Nikita Gambhir Dec 2020

Fungicide Resistance: Surveillance, Risk Assessment And Evolution In Two Soil-Borne Pathogens, Nikita Gambhir

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Fungicide-resistant pathogens are an increasing threat to fungicide efficacy and plant health. The goal of this dissertation was to advance the foundational knowledge required to prevent and detect fungicide resistance development in the seedling disease pathogen, Rhizoctonia zeae and the white-mold pathogen, Sclerotinia sclerotiorum. Corn and soybean fields in 12 states (IA, IL, IN, KS, KY, MI, MN, MO, ND, NE, SD, and WI) were surveyed for R. zeae. In vitro fungicide sensitivity was determined for 91 isolates to fludioxonil, sedaxane, and/or prothioconazole. Rhizoctonia zeae was sensitive to all fungicides (EC50 < 3 µg/ml) except azoxystrobin (EC50 > 100 µg/ml). In planta application of …

Population Structure And Reproductive Biology Of Monilinia Vaccinii-Corymbosi (Reade) Honey In Lowbush Blueberry In Maine, Katherine A. Ashley May 2020

Population Structure And Reproductive Biology Of Monilinia Vaccinii-Corymbosi (Reade) Honey In Lowbush Blueberry In Maine, Katherine A. Ashley

Electronic Theses and Dissertations

Lowbush blueberry, Vaccinium angustifolium Aiton, is both an economically and culturally important crop in Maine, being one of the few endemic crops to North America. The fungus Monilinia vaccinii-corymbosi (Mvc) causes mummy berry disease and is a significant pathogen of both highbush (Vaccinium corymbosum) and lowbush blueberries. While impacts of this disease are not regularly documented, it is estimated that 30-50% of the yield in an unmanaged field can be lost because of Mvc. This disease is typically managed with fungicides or burning of the field during years when the field is pruned, however, the impacts to the …

A Basic Ddradseq Two‐Enzyme Protocol Performs Well With Herbarium And Silica‐Dried Tissues Across Four Genera, Ingrid E. Jordon-Thaden, James B. Beck, Catherine A. Rushworth, Michael D. Windham, Nicolas Diaz, Jason T. Cantley, Christopher T. Martine, Carl J. Rothfels Apr 2020

Premise

The ability to sequence genome‐scale data from herbarium specimens would allow for the economical development of data sets with broad taxonomic and geographic sampling that would otherwise not be possible. Here, we evaluate the utility of a basic double‐digest restriction site–associated DNA sequencing (ddRADseq) protocol using DNAs from four genera extracted from both silica‐dried and herbarium tissue.

Methods

DNAs from Draba, Boechera, Solidago, and Ilex were processed with a ddRADseq protocol. The effects of DNA degradation, taxon, and specimen age were assessed.

Results

Although taxon, preservation method, and specimen age affected data recovery, large phylogenetically informative …

Characterization Of The Transcriptional Divergence Between The Subspecies Of Cultivated Rice (Oryza Sativa), Malachy T. Campbell, Qian Du, Kan Li, Sandeep Sharma, Chi Zhang, Harkamal Walia Jan 2020

Characterization Of The Transcriptional Divergence Between The Subspecies Of Cultivated Rice (Oryza Sativa), Malachy T. Campbell, Qian Du, Kan Li, Sandeep Sharma, Chi Zhang, Harkamal Walia

Agronomy & Horticulture -- Faculty Publications

Background: Cultivated rice consists of two subspecies, Indica and Japonica, that exhibit well-characterized differences at the morphological and genetic levels. However, the differences between these subspecies at the transcriptome level remains largely unexamined. Here, we provide a comprehensive characterization of transcriptome divergence and cis-regulatory variation within rice using transcriptome data from 91 accessions from a rice diversity panel (RDP1).

Results: The transcriptomes of the two subspecies of rice are highly divergent. Japonica have significantly lower expression and genetic diversity relative to Indica, which is likely a consequence of a population bottleneck during Japonica domestication. We leveraged high-density genotypic data and …

Patterns Of Genetic Diversity In Highly Invasive Species: Cogongrass (Imperata Cylindrica) Expansion In The Invaded Range Of The Southern United States (Us), Rima D. Lucardi, Lisa E. Wallace, Gary N. Ervin Jan 2020

Patterns Of Genetic Diversity In Highly Invasive Species: Cogongrass (Imperata Cylindrica) Expansion In The Invaded Range Of The Southern United States (Us), Rima D. Lucardi, Lisa E. Wallace, Gary N. Ervin

Biological Sciences Faculty Publications

The spatial expansions of invasive organisms in the novel range are generally expected to follow an isolation-by-distance relationship (IBD) if the invasion is biologically driven; however, many invasions are facilitated anthropogenically. This research focused on the extant expansion patterns of cogongrass (Imperata cylindrica). Cogongrass is a widespread invasive species throughout the southern United States (US). Patterns of infestation vary among US states. Cogongrass is pyrogenic, and its invasion threatens softwood (Pinus spp.) plantations, a substantial economic market for this US region. Over 600 individuals were sampled from seven invaded US states, using amplified fragment length polymorphisms (AFLPs) …

Snp Population Genetic Data For The Seagrass Halodule Uninervis From Pilbara Region, Western Australia, Kathryn Margaret Mcmahon, Richard Evans

Research Datasets

This dataset contains 80 SNP loci from 15 locations with 25-47 samples per locations. Each column contains data from two alleles. All genotyped samples are included. Prior to population genetic analysis clone mates were identified and removed.
