Over recent years, the “microbiota-gut-brain axis” (MGBA) has garnered significant attention in the scientific community. Specifically, perturbations of the MGBA via stress and dietary intake have been linked to a wide-range of diseases including gastrointestinal diseases, metabolic diseases, mood disorders, and cognitive diseases. However, most studies have been solely conducted on mice models and have yet to consider the more complex, intricate systems that impact the human body. Regulation Of Adipocyte Differentiation And Metabolism: Rab5-Guanine Nucleotide Exchange Factors And Methylglyoxal, Praew Chantarasinlapin

Internalization and trafficking of ligand-receptor complex rely on a particular set of proteins, e.g. small GTPase protein Rab5 and its activators called guanine nucleotide exchange factors. Rab5-activating protein 6 (RAP6), a Vps9-containing protein, may participate in Rab5-mediated insulin signaling and receptor trafficking. A dicarbonyl compound methylglyoxal was found to alter insulin signaling in preadipocytes. This dissertation aimed to investigate the association of RAP6 activity on 3T3-L1 preadipocyte differentiation and those driven by methylglyoxal. Overexpression of RAP6 inhibited preadipocyte differentiation, Ser473-phosphorylation of Akt1, and expression of adipogenic marker PPARγ, but not C/EBPα. Trends in adult obesity have been used to motivate key public health policies in the United States. While these analyses provide important insights into the broad historical contours of the obesity epidemic in the U.S., they shed less light on the proximate mechanisms that have generated these changes and that will ultimately determine the long-term course and pace of change in obesity rates. We used data from the National Health and Nutrition Examination Survey (NHANES), Glenn Firebaugh's linear decomposition technique, and Kitagawa's algebraic decomposition method to decompose change in body mass index (BMI), obesity, and morbid obesity from 1971 through …
