Increased age and obesity diminish motivation, perseverance, and spatial memory function. Aerobic exercise interventions have successfully rescued some of these processes. However, in older and heavier populations aerobic exercise is not as sustainable due to high risk of injury. Resistance exercise consists of physical activity where maximum oxygen consumption is not increased and has been proposed as a safe and effective intervention for this population. Here, we used touchscreen-based cognitive testing to elucidate the influence of resistance exercise on motivation and spatial memory in aged, diet-induced obese mice, using a water restriction procedure I develop in Chapter 2. Gut Microbial Metabolite Indole: A Stimulator Of Enteroendocrine Cell Differentiation Via Activation Of Aryl Hydrocarbon Receptor, James Hart

Enteroendocrine cells (EECs) regulate energy balance and glucose homeostasis by releasing hormones in response to food intake. Dysregulated EEC differentiation is observed in obesity, while gut microbiota metabolites influence this process. Here, we investigated the role of indole, a biologically active gut microbial metabolite, in EEC differentiation through aryl hydrocarbon receptor (AhR) activation. Human intestinal organoids derived from jejunal mucosal biopsies were exposed to indole. Indole treatment significantly increased mRNA levels of chromogranin A, an EEC marker. The effect was reversed by an AhR antagonist, indicating AhR involvement. Indole also upregulated AhR target gene mRNA levels. These findings highlight the …

While many weight loss interventions have resulted in significant weight loss, maintaining that weight loss long-term continues to be challenging for most, with long-term weight loss maintenance (LTWLM) rates as low as 20% among participants of clinical weight loss studies. Those who struggle with LTWLM exhibit many shared characteristics, including lower mental health composite scores; high levels of perceived stress; and lower rates of adherence to WLM behaviors. One explanation for this might be that emotion regulation (ER) skills are stronger among those who are successful at LTWLM, and therefore more successful at self-regulating for the goal of LTWLM.

Brown adipose tissue (BAT) has become a promising target for obesity treatment and prevention. However, effective dietary factors to promote BAT mass and function have not been identified. 17,18-epoxyeicosatetraenoic acid (17,18-EEQ) and 19,20-epoxydocosapentaenoic acid (19,20-EDP) are two prominent epoxy fatty acids (EpFAs) produced from n-3 polyunsaturated fatty acid eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), respectively. This dissertation fills an important research gap in the study of obesity policymaking by characterizing to what extent the phenomenon of dismantling is happening at the state level for obesity-related policies targeting the context of consumption, identifying determinants, and characterizing actors and processes involved. The five-year rate of dismantling of these policies is close to one-third for any attempts and one-tenth for enacted dismantling. Food and beverage tax policies face a particularly high rate of dismantling. Fatty acid oxidation inhibition is one approach to reducing blood glucose levels in type II diabetes. Skeletal muscle specific Carnitine Palmitoyltransferase 1b knockout mice (Cpt1bm-/-) cannot transport long-chain fatty acids into the mitochondria to be oxidized in order to produce energy. Cpt1bm-/- mice have debilitated fat oxidation, less fat mass and improved glucose utilization compared to control C57BL/6 mice fed a 25% fat diet.

Objective: This study’s purpose was to examine dietary quality, macronutrient intake, and micronutrient adequacy among long term weight loss maintainers (WLM) in a commercial weight management program.

Methods: Participants were 1,207 WLM in WW (formerly Weight Watchers) who had maintained a 9.1 kg or greater weight loss (29.7 kg on average) for 3.4 years, and had an average BMI of 28.3 kg/m2. A control group of weight stable adults with obesity (Controls; N=102) had a BMI of 41.1 kg/m2 and 2.3 kg or less weight change over the previous five years.

The aim of this study is to evaluate weight-related eating behaviors of participants with clinically significant weight loss (CSWL) in a proprietary weight loss program. A crosssectional sample of participants (n=1,454) enrolled in a proprietary weight-loss program that includes meal replacements and health coaching were queried for weight-related eating behaviors (using Weight Related Eating Questionnaire) of routine restraint (RR), compensatory restraint (CR), susceptibility to external cues (SEC), and emotional eating (EE) in relation to CSWL (defined as having achieved a weight loss greater than 10% of starting weight). Men and women have similar rates of obesity but the combined prevalence of overweight and obesity is higher among men. Men who are overweight are a high-risk group for many obesity-related chronic diseases, as they are more likely to carry excess weight in the abdomen, which is generally more harmful than weight stored in the lower body. Men are also less likely than women to perceive themselves as overweight, and thus are less likely to initiate weight loss through organized weight loss programs. The relationship between tibial plateau dimensions and knee vertical loading may explain the loading related injuries OW/OB children experience.

Background: The added sugar intake of children and adolescents in Canada warrants more research due to its contribution to obesity, diabetes, and other chronic and mental health issues.

Objective: To assess the trends and correlates of added sugar intake in Canadians aged 6 to 17.

Methods: Data were provided by the Canadian Community Health Survey-Nutrition 2004 and 2015. Statistical analysis included multivariable regression and t-tests.

Obesity is an increasing epidemic which during 2015-2016 afflicted 39.8% of adults and 18.5% of youth in the United States. Not only can obesity affect quality of life, but it can lead to life-threatening diseases such as type 2 diabetes, heart disease, stroke, and some cancers. Animal and human research have demonstrated a pattern of gut microbiota perturbation in overweight and obesity, characterized by a reduction of beneficial bacterial species and increase in harmful species. Obesity prevalence in the United States continues to increase and is associated with health consequences such as type 2 diabetes, hypertension, atherosclerosis, and hyperlipidemia. Among many contributing factors to obesity, fructose may be one of the major reasons as it disrupts the antioxidant system thereby resulting in an accumulation of reactive oxidative species and leading to obese conditions. The enzyme, isocitrate dehydrogenase 2 (IDH2), reduces nicotinamide adenine dinucleotide phosphate from the TCA Cycle, hence might be implicated with not only energy metabolism but also cellular redox homeostasis.
