Internalization and trafficking of ligand-receptor complex rely on a particular set of proteins, e.g. small GTPase protein Rab5 and its activators called guanine nucleotide exchange factors. Rab5-activating protein 6 (RAP6), a Vps9-containing protein, may participate in Rab5-mediated insulin signaling and receptor trafficking. A dicarbonyl compound methylglyoxal was found to alter insulin signaling in preadipocytes. This dissertation aimed to investigate the association of RAP6 activity on 3T3-L1 preadipocyte differentiation and those driven by methylglyoxal. Overexpression of RAP6 inhibited preadipocyte differentiation, Ser473-phosphorylation of Akt1, and expression of adipogenic marker PPARγ, but not C/EBPα. Trends in adult obesity have been used to motivate key public health policies in the United States. While these analyses provide important insights into the broad historical contours of the obesity epidemic in the U.S., they shed less light on the proximate mechanisms that have generated these changes and that will ultimately determine the long-term course and pace of change in obesity rates. This dissertation seeks to examine obesity in different contexts throughout the life course. Through empirical analyses, separate stages of the life course are considered: namely childhood through adolescence, young adulthood, and adulthood. By using a life course perspective, it is possible to consider longitudinal and intergenerational approaches to these questions, which will update and inform the current debates surrounding obesity.

This study observed the relationship between television consumption and child obesity through the theoretical construct of Putnam’s displacement hypothesis. It did so by updating the previous research through the investigation of the displacement of both physical activity and advertisements on contemporary television platforms. The inclusion of Putnam’s displacement hypothesis was to provide a foundational framework, not found in previous literature, to study two important paradigms that were represented in previous research: 1) the displacement of physical activity; and 2) the displacement of traditional advertisement exposure.

We extend the existing literature on food taxes targeting obesity. We systematically incorporate the implicit substitution between added sugars and solid fats into a comprehensive food demand system and evaluate the effect of taxes on sugars and fats. The approach conditions how food and obesity taxes affect total calorie intake. The proposed methodology accounts for the ability of consumers to substitute leaner low-fat and low-sugar items for rich food items within the same food group.
