Shoot Organogenesis And Plant Regeneration In Begonia Coptidifolia, Beiyi Guo, Yuping Xiong, Hai Ren, Teng Wu, Jaime A. Teixeira Da Silva, Songjun Zeng, Guohua Ma

Turkish Journal of Agriculture and Forestry

An efficient regeneration protocol was established for a rare plant species, Begonia coptidifolia. Using leaf explants, adventitious shoots were induced on Murashige and Skoog (MS) basal medium using various concentrations (1.0-10.0 mM) of three cytokinins, 6-benzyladenine (BA), thidiazuron (TDZ) and kinetin (KIN). However, adventitious shoots could not be induced on MS medium supplemented with auxins [α-naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D)] tested in the 1.0-10.0 mM range. The highest number of shoots per explant (31.2) was induced on MS medium with 5.0 mM BA and 1.0 mM NAA. Although roots were induced in ½MS medium free of plant growth …

Culture Media And Growth Regulators Influence Callus Induction And Plant Regeneration Of Mature Embryos Of Orchardgrass (Dactylis Glomerata L.), Selçuk Kodaz, Kami̇l Hali̇loğlu, Arash Hossein Pour, Murat Aydin Jan 2021

Culture Media And Growth Regulators Influence Callus Induction And Plant Regeneration Of Mature Embryos Of Orchardgrass (Dactylis Glomerata L.), Selçuk Kodaz, Kami̇l Hali̇loğlu, Arash Hossein Pour, Murat Aydin

Turkish Journal of Agriculture and Forestry

To establish an efficient in vitro regeneration protocol for orchardgrass (Dactylis glomerata L.), type of medium, several plant growth regulators and their various concentrations were evaluated in a factorial experiment for callus induction and plant regeneration. Endosperm supported mature embryo were used as explants and optimum response in callus induction was observed on Chu medium with 12 mg/L dicamba (3.6-dichloro-2-methoxybenzoic acid). The highest responding embryogenic callus (15%) was found with Murashige and Skoog medium and 8 mg/L 2.4-D (2,4-dichlorophenoxy acetic acid). The highest regeneration efficiency was achieved on MS medium (17 plantlets) with 8 mg/L 2,4-D. Endosperm-supported mature embryos of …

Optimization Of Agrobacterium-Mediated Transformation And Regeneration For Crispr/Cas9 Genome Editing Of Commercial Tomato Cultivars, Zafer Seçgi̇n, Musa Kavas, Kubi̇lay Yildirim Jan 2021

Optimization Of Agrobacterium-Mediated Transformation And Regeneration For Crispr/Cas9 Genome Editing Of Commercial Tomato Cultivars, Zafer Seçgi̇n, Musa Kavas, Kubi̇lay Yildirim

Turkish Journal of Agriculture and Forestry

Tomato (Solanum lycopersicum) is the second most important horticultural crop worldwide that is widely used as a model plant in genetic manipulation of Solanaceae. CRISPR/Cas9 system has been successfully utilized in several studies for genome edition of model tomato cultivars. However, these genome editing systems should be also optimized for commercial tomato cultivar for direct application of genome editing in field conditions. In this study, we have optimized an Agrobacterium-mediated gene transfer and regeneration system for CRISPR/Cas9 genome editing in two commercial tomato cultivars for the first time. The effect of explant type, genotype, pre-transformation time, Agrobacterium concentration, infection time, …
