Site Directed Mutagensis Of Bacteriophage Hk639 And Identification Of Its Integration Site, Madhuri Jonnalagadda

Bacteriophages affect bacterial evolution, pathogenesis and global nutrient cycling. They are also the most numerous and diverse group of biological entities on the planet [1, 2, 3, 4, 5, 6]. Members of the Lambda phage family share a similar genetic organization and control early gene expression by suppressing transcription, a process known as antitermination. Transcription antitermination in Lambda is mediated by a phage-encoded protein whereas in lambdoid phage HK022, antitermination is mediated by a phage-encoded RNA molecules. Recent results suggest that another bacteriophage called HK639 also appears to use RNA-mediated antitermination. To characterize this newly identified phage we generated site …

The illegal bushmeat harvest has been identified as a reason for declining wildlife populations throughout much of Africa. For many years the trade was thought to exist primarily in Western Africa. The illegal use of bushmeat in Eastern Africa, including Kenya, went undocumented and unstudied. In 2004, the Born Free Foundation published a study which claimed illegal sale of wild game was substantial in butcheries throughout Nairobi, Kenya. Field studies were conducted during the summers of 2007 and 2008 at the Agricultural Research and Education Complex, Western Kentucky University, Warren County, KY and commercial production fields in Caldwell County, KY, Warrick County IN, and Vanderburgh County, IN. The goals of these studies were to further validate the Duncan grain yield model, the Russell aboveground biomass model, and to study the effect of inconsistent spacing within rows on Zea mays L. yield. Invasive species are an increasing threat to biological diversity as well as a leading cause of recent species’ extinctions. Invasives spread quickly and efficiently, and the U.S spends millions of dollars annually in the control and eradication of these species. More information is necessary in order to predict which species may become invasive. Rubus (Rosaceae) was chosen for study because this genus includes various ploidy levels, reproductive modes, and species that are invasive as well as native.

Only 6 autochthonous cases of human Chagas disease have been documented in the U.S.A., however, as many as 5% of immigrants from Latin America may be infected with the etiologic agent, Trypanosoma cruzi. The parasite has been isolated from a variety of wild mammals, particularly in the southeastern region of the U.S.A. The goal of our study was to determine if the sylvatic cycle of T. cruzi infection occurs in Kentucky, and, if present, to assess the prevalence of infection in Warren and Barren counties. Since the advent of techniques to investigate gene expression on a large scale, numerous circadian rhythms in mRNA abundance have been reported. These rhythms generally differ in amplitude and phase. First studies on circadian rhythms of transcription on a large scale are also emerging. We investigated to what extent the same circadian regulatory mechanism of transcription can give rise to rhythms in RNA amount that differ in phase solely based on a parameter that is not regulated by the circadian clock. Using a discrete-time approach, we modeled a sinusoidal rhythm in transcription with various constant exponential RNA decay rates. We …

The 1815 Log House is located on the campus of Western Kentucky University. Built in the early 1800's by Archibald Felts, the house was occupied by his descendants until 1968. The dogtrot floor plan, V-notched logs, and stone chimneys are some of the historical architectural features that can be viewed. It was donated to the Kentucky Library & Museum at WKU in 1980, and now serves as an on-site exhibit of early frontier life in Kentucky. Effects Of Exercise Induced Fatigue On Postural Balance: A Comparison Of Treadmill Versus Cycle Fatiguing Protocols, Katherine Wright

The purpose of this study was to examine the effects of muscle fatigue after a treadmill versus cycle ergometer incremental test on postural balance indices and recovery time recreationally trained individuals. The Bruce Treadmill Test and the Incremental Cycle Ergometer test were used to induce fatigue. Each subject (N=16) performed both exercises, but the order of the exercises was randomized. The subjects performed balance measures on a Biodex Balance System via the Dynamic Balance Test at level 5 and indices were recorded as given. Phopshorylase kinase (PhK) is a hexadecameric holoenzyme made up of four different subunits in the arrangement (αβγδ)4 and has total molecular mass of 1.3MDa. Alpha and β are regulatory subunits, γ is catalytic, and δ is an intrinsic molecule of calmodulin. PhK is a serine threonine kinase with glycogenolytic regulatory functions. Our lab has recently discovered that the γ subunit can be alternatively processed to produce a truncated form of 181 residues (γ-181). This variant of γ contains a phosphorylation site for PK-C, and its activity is influenced by this phosphorylation. We are using a LexA based yeast two hybrid …

The hypertrophied musculoskeletal system driving adduction of Sealopus aquatieus's forelimb supports the locomotion of these burrowing subterranean mammals. Due to the excessive bulk of the major muscles contributing to this action, it was thought that this particular species may represent a rare case of a mammalian species with a greatly enhanced force contribution from each muscle, correlating with a reduction in the potential for the forelimb to adduct at a high velocity. By assessing the lever ratios of five significant muscles contributing to forelimb adduction, combined with vector analyses for the angles of these muscles' insertions, a model was established …

As of 2006, the U.S. Environmental Protection Agency (EPA) changed the Maximum Contaminant Level (MCL) for arsenic from 50 parts per billion (ppb), to 10ppb because of links to cancer. Current remediation technologies are expensive; therefore, this change will result in increased economic pressure on rural communities with high levels of arsenic in their drinking water. Lowering of the standard has spurred the development of a novel remediation technology that has shown the ability to reduce arsenic in drinking water at the source, with the added benefit of low-cost disposal of a stable and benign waste product in ordinary landfills. …

Women's perceptions of technological changes in textiles and apparel and how the changes have affected daily life were examined using the transmissive reminiscence technique. Technological advances do contribute to use of less time and effort and offer a greater variety of products, but the products often have a shorter life expectancy or are kept in inventory for a shorter time because it is more expedient to replace rather than repair or recycle textile and clothing items. Care must be taken to cultivate cohesive relationships within the family and teach values that were once inherent in textiles/ apparel tasks. Otherwise advances …
