Parkinson’s Disease (PD) is an idiopathic disorder with no known cure. With number of cases steadily rising around the world, it is imperative to turn to the underlying cellular and molecular mechanisms of the disease manifestation and neurodegeneration to craft novel modes of therapy. VPS35 is one of the few genes that have identified and definitively linked to familial PD. The particular mutation that has been associated is known to cause dysfunction of a key cellular process known as autophagy. The extracellular microenvironment contributes significantly to a cell’s function and behavior. For instance, cell-cell interactions, cell-substrate interactions, and physical forces are all factors of the extracellular environment that can alter cellular behavior. Cells can receive these signals and forces through various membrane channels and receptors that transmit the signals from the extracellular to the intracellular space. Canonical Notch signaling is induced by ligand interactions with neighboring cells, but recent evidence has revealed that Notch signaling can occur through a variety of extracellular stimuli including hyperglycemia, hypoxia, multiple growth factors, fluid shear stress, and extracellular matrix (ECM) composition. Numerous applications of nanomaterials have led to their use in electronics, optics, and medicine. However, creating materials at such a small size brings them on the same scale as many biomolecules and cellular components, altering their interactions with biological systems. The study of evolutionary innovations, or novel traits, is integral to understanding evolution yet is poorly understood. By studying the regions between genotype networks that produce the same phenotype, we can better characterize the process by which innovation occurs. The overarching goal of this study is to assign fitness values to the overlapping genotype network of two catalytic RNA molecules, or ribozymes. Properly characterizing this region requires the study of thousands of individual sequences, which is achievable through the use of high-throughput sequencing analysis. Genetic structure is useful for inferring patterns of selection, gene flow and connectivity, and can define management units that aid in interpretation of spatially-specific trends and species management. American kestrels (Falco sparverius) are a widespread, generalist species with fully migratory, partial migrant, and resident populations. In many parts of their range, kestrels show evidence of declining population trends; however, it has been difficult to identify threats to kestrels because of differences in regional trends. Presented is the first field survey and laboratory-based study focused on Harpellales gut fungi found in Culicidae (mosquito) larvae collected from various parts of Idaho. Overall, 34 sites were sampled ranging from urban storm drains and irrigation puddles to pristine stream-side puddles. These sites yielded 17 different species of mosquitoes and three previously described species of gut fungi. Three species of mosquitoes were the first recorded observations as hosts of the following gut fungi: Culiseta alaskaensis was infested with Zancudomyces culisetae (from Renwyck Creek), Culex tarsalis with Smittium culicis (from Cottonwood Creek), and Ochleratus sp. with Smittium minutisporum (from Bear …

Fitness landscapes or adaptive landscapes represent the mapping of genotype (sequence) to phenotype (function or fitness). Originally proposed as a metaphor to envision evolutionary processes and mutational interactions, the fitness landscape has recently transitioned from theoretical to empirical. This is due in part to advances in DNA synthesis and high-throughput sequencing. This allows for the construction and analysis of empirical fitness landscapes that encompass thousands of genotypes. These landscapes provide tractable insight into mutational pathways, the predictability of evolution or even the evolution of life. Protected areas are a staple in conservation, but human activities outside of protected areas drive species interactions, compositions, and distributions. Research is especially needed in these multi-use landscapes to maintain habitat connectivity for entire wildlife communities between protected areas. Yet, such research is lacking in areas it is needed most, such as in sub-Saharan Africa, where human populations are expected to double by 2050. Cellular signaling pathways provide cells with the means to sense their environment and communicate with other cells. The Notch signaling pathway is comprised of a set of protein machines which work in unison to coordinate cellular processes in response to stimuli coming from neighboring cells and changing microenvironmental conditions. Outdoor recreation, as the intersection between physical exercise and nature, provides a multitude of psychological and physiological benefits to human well-being. Though many studies have reported qualitative stress reduction from outdoor recreation, few have focused on quantitative measurements of stress across recreational activity types, intrapersonal differences, and environmental variables. To determine whether outdoor recreation affects physiology, we collected 190 paired salivary cortisol and testosterone samples and 157 surveys from 88 hikers, 81 mountain bikers, and 44 off-highway vehicle (OHV) motorists. After recreation, cortisol concentrations were significantly reduced in hikers and OHV motorists, but cortisol and testosterone concentrations increased in mountain …

Great Lakes coastal wetlands are some of the most crucial ecosystems to biodiversity in the Great Lakes Basin, yet suffer increasing degradation due to invasive plants. Wetland plant invasions can be controlled in their initial stages, but early detection of invasive plants using field surveys are often untenable due to budget constraints. Quorum sensing is an intercellular mechanism used by many bacterial pathogens to regulate behaviors, such as motility, virulence, and antibiotic production. Disruption of quorum sensing is shown to be deleterious to bacterial pathogenicity, without causing lethality. In Gram-negative bacteria, acyl-homoserine lactone (AHL or acyl-HSL) signal molecules are synthesized by AHL synthase enzymes using acylated-acyl carrier protein (acyl-ACP) and S-adenosyl-L-methionine (SAM) as their substrates. Pantoea stewartii EsaI, which causes Stewart’s wilt in corn, and Yersinia pestis YspI, which causes bubonic plague are AHL synthases that use 3-oxohexanoyl-ACP and 3-oxooctanoyl-ACP as the acyl-substrate to synthesize 3-oxohexanoyl-HSL and 3-oxooctanoyl-HSL signals, respectively. Migratory birds face threats throughout the annual cycle, and cumulative effects from linkages between the breeding and non-breeding grounds may impact species at the population level. Long-billed Curlews (Numenius americanus) are a migratory shorebird of conservation concern associated with grasslands that show breeding population declines at some regional and local scales. Curlews exhibit high site fidelity to breeding territories, but also spend approximately 75% of the year on the wintering grounds. Therefore, localized population declines could indicate localized threats, in the breeding or wintering grounds.
