Trypanosoma brucei is the protozoan parasite that causes African Sleeping Sickness in humans and nagana, a wasting disease in cattle. T. brucei completes its life cycle in two hosts, mammals and the tsetse fly insect vector. Due to the geographical restriction of the tsetse fly, the disease is endemic in sub-Saharan Africa. Both the insect and mammalian forms of the parasite need fatty acids to anchor their surface proteins. Soil disturbance from the cultivation and intensive management of orchards in hot and humid subtropical climates can reduce soil organic matter (OM). This research explored how the addition of OM amendments impact soil and tree parameters in two field studies and how proximity to OM may elicit unique genomic response of peach roots in a split-root pot study. The first field study applied two rates of compost (1x and 2x) and subsequently reduced fertilizer applications annually to a virgin and replant orchard location. The effect of long-term frozen storage of Atlantic salmon and sliced peaches was studied to determine methods to optimize quality. The first experiment was conducted to determine the quality changes in Atlantic salmon stored at different freezer temperatures over 12 months. Fresh and pre-frozen salmon were placed in five different freezers set at –7°C, –12°C, –18°C, –29°C and –77°C and evaluated for quality at 30, 90, 180, 270 and 360 days of storage. In general, higher quality was retained to a greater extent in salmon held at –29°C and –77°C for 360 days compared to the other storage temperatures. No …

Mini-ring is a disease in ultradwarf bermudagrass (UDBG) [Cynodon dactylon (L.) Pers. × C. transvaalensis (Burtt-Davy)] putting greens caused by Waitea zeae (Voorhees) J.A. Crouch & Cubeta, (formerly Rhizoctonia zeae). Symptoms typically resemble frog-eye patches that are 10 to 40 cm in diameter with a bronze to orange outer ring and green center. In the southeastern United States, mini-ring symptoms appear in late-summer and generally persist until UDBG dormancy in late-fall. Arsenic is an environmental contaminant commonly found in food and drinking water. Exposure to arsenic during embryonic development has been linked to reduced muscle growth, disrupted muscle development and locomotor activity, impaired neurodevelopment, reduced IQ, impaired memory and learning deficits. While the mechanisms responsible for developmental changes following in utero exposure to arsenic are not well known, one possibility is that arsenic might disrupt proper cellular differentiation. Bone fracture healing includes complex and sequence of dynamic events to restore the integrity and biomechanical properties of the bone. While most of the fractures heal without any problem, healing is sometimes compromised. Two significant fracture healing complications are orthopedic implant-associated infections and non-unions/delayed union. These can be interrelated causes as well. Implant-associated infection can cause implant loosening, and as a result, it can delay the fracture healing process. In this dissertation, we investigated the behavior and ecology of the American alligator (Alligator mississippiensis) in coastal, residential resort communities with an overarching goal of identifying behavioral patterns of alligators that can inform risk management strategies. Toxoplasma gondii is an obligate intracellular parasite that can infect a wide range of mammalian hosts. T. gondii can cause severe disease in immunocompromised individuals as well as pregnant women, in which congenital toxoplasmosis can lead to spontaneous abortion and fetal blindness. T. gondii infects up to roughly one-third of the global human population and is the causative agent of toxoplasmosis. Given this health burden of toxoplasmosis and that antibiotics currently used for the treatment of infection lead to strong side effects, identification of parasite-specific targets are necessary for novel therapeutic strategies.

In this dissertation we describe methods for measuring infection relevant biochemical analytes using radioluminescent and ultrasound luminescent materials. Films and nanoparticles fabricated with europium doped gadolinium oxysulfide (Gd2O2S:Eu3+) are used to quantitatively measure radiolabeled pharmaceutical concentration, specifically tritium labeled vancomycin (3H-vancomycin). Europium and dysprosium doped strontium aluminate is used to fabricate an ultrasound modulated, pH sensing film. These methods are indicated for theranostic evaluation of implant associated infection. Kefir grains were purchased from online, home kitchen vendors in the United States (n = 22), inoculated into UHT milk, and incubated at 25°C for 22 hours. Results indicated lactobacilli, lactococci and yeast in all samples. Six out of 11 samples contained coliform, and all samples contained pseudomonal and staphylococcal species. Using targeted genome sequencing (16S) of kefir grain samples 1, 2, and 3 revealed that lactobacilli were the predominant genera of bacteria. ITS sequencing revealed different fungal species in each grain. Results indicated potential presence of menaquinone-4 and menaquinone-7 by UPLC-MS/MS analysis.
