Prior to the Deepwater Horizon oil spill, we lacked a comprehensive baseline of oil contamination in the Gulf of Mexico's sediments, water column, and biota. Gaps in prespill knowledge limit our ability to determine the aftereffects of the Deepwater Horizon blowout or prepare to mitigate similar impacts during future oil spill disasters. We examined spatiotemporal differences in exposure to and metabolism of polycyclic aromatic hydrocarbons (PAHs) in 2 hake species (Urophycis spp.) to establish a current baseline for these ecologically important, abundant, and at-risk demersal fishes. This dataset contains hepatic polycyclic aromatic hydrocarbons (PAHs) concentrations in Red Hind (Epinephelus guttatus) collected aboard R/V Weatherbird II cruises (WB1603, WB1701, WB1731) in the southern Gulf of Mexico (Mexico and Cuba) from 2015-09-18 to 2017-05-23. The dataset includes the location, date, concentrations of various PAHs in liver tissue (ng/g w.w.), and the total or sum of the low and high molecular weight PAHs (ng/g w.w.). PAHs were extracted from homogenized liver tissues using QuEChERS EMR (Agilent Technologies, Bond Elut). This dataset contains a histological examination of livers of golden tilefish (Lopholatilus chamaeleonticeps) collected aboard the R/V Weatherbird II cruises WB-1304, WB-1403, WB-1502, WB-1602, WB-1603, and WB-1701 in the Gulf of Mexico from 2012-08-14 to 2016-09-06. The dataset includes the identified presence of histological changes and images of the gross and histological evaluations. Formalin-fixed tissues were individually evaluated for color, texture, size, shape, and consistency. Photographs were taken with a Sony-Cybershot camera (Sony Corporation, Minato, Tokyo, Japan). This dataset contains pathological findings in red snapper (Lutjanus campechanus) livers collected aboard the multiple Weatherbird II cruises WB1304 (2012-08-13 to 2012-08-25), WB1403 (2013-08-15 to 2013-08-30), WB1502 (2014-08-05 to 2014-08-20), WB1602 (2015-08-15 to 2015-08-30), WB1736 (2017-07-18 to 2017-08-01) in the north central region of the Gulf of Mexico, post-Deepwater Horizon. This dataset contains the concentrations of polycyclic aromatic hydrocarbons (PAHs) in liver tissue of golden tilefish (Lopholatilus chamaeleonticeps) collected aboard multiple R/V Weatherbird II cruises WB1304 (2012-08-13 to 2012-08-25), WB1403 (2013-08-15 to 2013-08-30), WB1502 (2014-08-05 to 2014-08-20), WB1602 (2015-08-15 to 2015-08-30), WB1603 (2015-09-10 to 2015-10-02), WB1701 (2016-08-02 to 2016-09-09), and WB1736 (2017-07-18 to 2017-08-01) in the Gulf of Mexico during the Gulf of Mexico-wide surveys from 2012-2017. The malaria pre-erythrocytic stages have been identified as an ideal therapeutic target, but complex in vitro models for Plasmodium vivax and Plasmodium falciparum lack the efficiency needed for rapid screening and evaluation of new vaccines and drugs, especially targeting the P. vivax hypnozoite. To address this challenge, we employed a multi-parameter approach using “omics’” to identify pre-erythrocytic targets and biomarkers, guide phenotypic therapeutic screening, and study parasite functionality with innovative bioassays using highcontent screening. PANcreatic-DERived factor (PANDER) is a member of a superfamily of FAM3 proteins modulating glycemic levels by metabolic regulation of the liver and pancreas. The precise PANDER-induced hepatic signaling mechanism is still being elucidated and has been very complex due to the pleiotropic nature of this novel hormone. Our PANDER transgenic (PANTG) mouse displays a selective hepatic insulin resistant (SHIR) phenotype whereby insulin signaling is blunted yet lipogenesis is increased, a phenomena observed in type 2 diabetes. Malaria is a devastating global health issue that affects approximately 200 million people yearly and over half a million deaths are caused by this parasitic protozoan disease. PANcreatic-DERived factor (PANDER, FAM3B) has been shown to regulate glycemic levels via interactions with both pancreatic islets and the liver. Although PANDER is predominantly expressed from the endocrine pancreas, recent work has provided sufficient evidence that the liver may also be an additional tissue source of PANDER production. At physiological levels, PANDER is capable of disrupting insulin signaling and promoting increased hepatic glucose production. As shown in some animal models, strong expression of PANDER, induced by viral delivery within the liver, induces hepatic steatosis. However, no studies to date have explicitly characterized the transcriptional regulation of PANDER from the liver. …

Malaria is a critical and global public health problem, affecting over 200 million people every year, resulting in over 500,000 deaths. A vaccine is not currently available and only one drug, primaquine, is effective against the dormant stages of Plasmodium vivax. Preclinical assessment of novel therapeutic drugs and vaccines is hampered by the lack of an in vitro liver model for P. falciparum and P. vivax. PANcreatic-DERived Factor (PANDER), or FAM3B, is a 235-amino acid protein strongly expressed within and secreted from the endocrine pancreas. Research surrounding PANDER has revealed a large role for the protein in maintaining glucose homeostasis, as evidenced by several Ad-PANDER overexpressing murine models, our lab's pancreas-specific PANDER transgenic overexpressor, and most recently our mixed genetic C57/129J PANDER knockout (PANKO) mouse. However, PANDER's overall role in glycemic regulation and glucose homeostasis has yet to be studied in a purebred C57BL/6J PANDER knockout model.
