Traumatic brain injuries (TBIs) are a common form of head injury, with mild TBIs (mTBIs) making up 75-90% of the TBI severity scale. It’s been suggested that repetitive mild TBIs (rmTBIs) may enhance effects of Alzheimer’s Disease (AD). With dementia linking TBI and AD together, we investigated the memory hub of the brain - the hippocampus. Astrocytes and microglia are two glial cells that respond to neuroinflammation and therefore imaged in hippocampal regions of the 3xTg-AD mouse brain. We hypothesized that AD brain pathology (i.e., neuroinflammation) would be accelerated in 3xTg-AD mice following rmTBIs early in life. Analyses revealed that …

Models of settlement patterns in the northwest Great Basin describe a decrease in residential mobility, intensified use of upland spring ecozones, and an increase in diet breadth during the Middle and Late Archaic. Here, I present data collected from the Sage Hen Springs site in northwestern Nevada during a Phase II testing project conducted by the BLM and an analysis of these data focusing on patterns of subsistence and mobility strategies throughout the Archaic. Results of this analysis support existing models of lifeways in the northwest Great Basin at the small scale and point to climatic factors as influences on …

The glenohumeral joint (GH) is commonly conceptualized as a ball-and-socket joint [1], and its center of rotation (COR) is presumed to coincide with the geometric center of the medial-superior region of the humeral head [2]. Recent research has endorsed improvements in COR estimation through invasive and noninvasive techniques, including cadaver studies, stereophotogrammetry, and motion capture (MOCAP) systems. Despite increased interest in wearable technology within human movement analysis, the problem of COR estimation employing MOCAP systems and its validation against bi-planar fluoroscopy remains relatively unexplored.

Osteoarthritis (OA) is a form of arthritis that develops in the joint due to overuse and aging causing pain, discomfort, and disability. Total Knee Arthroplasty (TKA) is a surgical procedure performed when OA symptoms are severe with an estimated 600,000 patients in the United States currently receiving TKA. Studies have reported dissatisfaction of the knee for 14-39% of patients. This study collected knee kinematics before and after surgery using stereo radiography for precise measurement of gait and deep knee flexion activities. Understanding the interplay between nucleic acids and protein aggregation is integral to the understanding of proteostasis, aging, and neurodegenerative disease progression. Nucleic acids are known to modulate the aggregation of PrP, tau, ⍺-synuclein, and other disease relevant proteins. Although the interactions between misfolded protein and nucleic acids can play a role in disease, this interaction may potentially be beneficial as well. Our group and others have shown nucleic acids can be powerful chaperones. The dugong (Dugong dugon) is an important marine mammal in Palau. However, current population dynamics are not well understood. This study aimed to connect scientific data with local knowledge by examining the social implications of changing fishing methods and their impact on dugong population dynamics in the face of climate change and fisheries commercialization. Through interviews with local fishers, it was found that destructive fishing methods like trawling and long lining have led to habitat loss and degradation for dugongs. While marker-based motion capture remains the gold standard in measuring human movement, accuracy is influenced by soft-tissue artifacts, particularly for subjects with high body mass index (BMI) where markers are not placed close to the underlying bone. Obesity influences joint loads and motion patterns, and BMI may not be sufficient to capture the distribution of a subject’s weight or to differentiate differences between subjects. Subjects in need of a joint replacement are more likely to have mobility issues or pain, which prevents exercise. Obesity also increases the likelihood of needing a total joint replacement. Small GTPases are a critical component to the spatiotemporal accuracy of intracellular trafficking pathways as they determine the specificity and direction of organelle transport. There exists over 150 small GTPases categorized into 5 sub-families and are employed across all cell types. Understanding how the early stages of sexual signal diversification proceed is of critical importance because the nature of these microevolutionary dynamics can directly shape species trajectories and macroevolutionary patterns. Unfortunately, studying signal diversification is challenging because signals involve complex interactions between behavioral, morphological, and physiological components, many of which can only be measured in real time. The diversification of animal communication systems is driven by many interacting factors. Unintended receivers play an important part in this process, yet little is known about their role in signal evolution. Flies of the genus Ormia are parasitoids of crickets and rely on acoustic cues to locate hosts. In Hawaii, selection imposed by Ormia ochracea has led to recent and rapid diversification in the songs of their host. Here, we compare neural and behavioral responses of Hawaiian flies to those of an ancestral population to understand the role of parasitoid sensory and behavioral variation in the evolution of host songs. …

Exocytosis is an essential process for intercellular communication in eukaryotic cells. This process involves significant changes in membrane curvature, and lipids and curvature-sensing proteins can assist these processes. One protein in particular, phospholipase D1 (PLD1), and its product, the lipid phosphatidic acid (PA), are involved in multiple exocytic processes. However, PLD1 and PA’s role in this process has remained unclear. In this work, PLD1 and the production of PA were visualized during exocytosis, and PA localization to regions of membrane curvature was established. Together, these results support the hypothesis that PLD1 production of PA stabilizes negative curvature during membrane fusion. …

Proteins can react with reactive oxygen species (ROS) and reactive nitrogen species (RNS) to form post-translational modifications (PTMs), which can affect protein structure and function. The formation of 3-nitrotyrosine (NTyr) and dityrosine (DiTyr) upon reaction of proteins with ROS/RNS are two common PTMs studied due to their stability and irreversibility, as well as their ability to enhance the allergenicity of pollen allergens upon formation. The aberrant fibrous, extracellular, and intracellular proteinaceous deposits in cells, organs and tissues are referred to as amyloids. These deposits are dominated by β-sheet structures that have been implicated in several neurodegenerative diseases and cancer. In this work, the types of amyloidosis studied include Parkinson’s disease (PD) using UA196 and NL5901 strains of Caenorhabditis elegans (C. elegans), Alzheimer’s disease (AD) using GMC101 strain of C. elegans, and cancer-associated mutant p53 aggregation in MIA PaCa-2 mutant cells. Mild traumatic brain injury (mTBI), yielding a Glascow Coma Scale of 13-15, is the most commonly occurring severity of TBI. Pathology from mTBI consists of blood brain barrier disruption, neuroinflammation, oxidative stress, excitotoxicity, mitochondrial dysfunction, protein aggregation, axonal degeneration, and resulting neuronal death. These processes deplete the body’s endogenous antioxidant system. We report a retrospective analysis of antioxidant blood biomarkers in patients with a history of mTBI from a local sports medicine clinic, Resilience Code. We found persistent sex-specific antioxidant depletions in mTBI patients associated with worsened symptomology.

The role of EHD2 in terms of Cav1 interaction had not been previously characterized and was the main goal of this research. Cav1 has been shown to lead to the ubiquitination of Rac1. Because of this, the role of EHD2 in Rac1 regulation was investigated. An increase in long-term migration in endothelial cells was observed and suggested that EHD2 impacts the RhoA pathway instead of the Rac1 pathway. Cell corpse clearance is key for animals to avoid inflammation and autoimmune disease. To better understand the “eat me” signal of phosphatidylserine (PS) exposure for non-apoptotic corpse clearance, we attempted to disrupt the externalization of PS on C. elegans polar bodies. The second polar body undergoes a non-apoptotic form of cell death, loses membrane integrity, externalizes PS and is cleared by embryonic cells during early development. During apoptotic and necrotic cell death, PS asymmetry is disrupted by lipid scramblases, which translocate phospholipids between the leaflets of the lipid bilayer. Specific leaf area (SLA) reflects a plant’s carbon investment per area and relates to generalized leaf economics spectrum growth strategies (LES). To fill gaps of knowledge about intraspecific SLA variation and why it is often inconsistent with the LES, we studied the response of two riparian tree species along the urban-rural gradient in Northeastern Colorado: an invasive, nitrogen-fixer with broad tolerances and a nitrogen-limited and shade-intolerant native. Fires impact ecosystems globally and due to climate change, there are shifts in fire regimes that impact ecological communities which provide essential ecosystem services. Focusing on arthropods, fire can influence this ubiquitous animal group in various way. We performed a comprehensive meta-analysis evaluating how fires impact differing arthropod functional groups. We found that overall, fire negatively effects community level responses for most functional groups with herbivores as the only exception showing some positive effects of fire. Patterns of morphological divergence across species’ ranges provide insight into local adaptation and speciation. Here, we compare phenotypic divergence among 4,221 crickets from 337 populations of two related species of field cricket, Gryllus firmus and G. pennsylvanicus and their hybrids. We find that these species differ across their geographic range in key morphological traits, such as body size and ovipositor length, and we directly compare phenotype with genotype for a subset of crickets demonstrating nuclear genetic introgression, phenotypic intermediacy of hybrids, and essentially unidirectional mitochondrial introgression. We discuss how these morphological traits relate to life history differences between the species. …

Among bony vertebrates, the melanocortin-2 receptor ortholog is unique among the family of five melanocortin receptors on the basis that it is dependent on its accessory protein, MRAP1, for trafficking and activation, and is selective for activation by ACTH alone. Previous studies on the MC2R orthologs of select cartilaginous fish, the elephant shark (Callorhinchus milii) and the red stingray (Dasyatis akajei), revealed divergent traits in a less obligatory relationship on MRAP1 and its ability to be activated by ACTH or the MSH-sized peptides.
