Functional Analysis Of The Acetic Acid Resistance (Aar) Gene Cluster In Acetobacter Aceti Strain 1023, Elwood Mullins, T Joseph Kappock

Vinegar production requires acetic acid bacteria that produce, tolerate, and conserve high levels of acetic acid. When ethanol is depleted, aerobic acetate overoxidation to carbon dioxide ensues. The resulting diauxic growth pattern has two logarithmic growth phases, the first associated with ethanol oxidation and the second associated with acetate overoxidation. The vinegar factory isolate Acetobacter aceti strain 1023 has a long intermediate stationary phase that persists at elevated acetic acid levels. Many food plants accumulate oxalate, which humans absorb but do not metabolize, leading to the formation of urinary stones. The commensal bacterium Oxalobacter formigenes consumes oxalate by converting it to oxalyl- CoA, which is decarboxylated by oxalyl-CoA decarboxylase (OXC). OXC and the class III CoA-transferase formyl- CoA:oxalate CoA-transferase (FCOCT) are widespread among bacteria, including many that have no apparent ability to degrade or to resist external oxalate. The EvgA acid response regulator activates transcription of the Escherichia coli yfdXWUVE operon encoding YfdW (FCOCT), YfdU (OXC), and YfdE, a class III CoA-transferase that is ~ 30% identical to YfdW. Indiana sweet corn acreage harvested for fresh market averaged 5,633 acres annually from 2010-2012, with a yield of 71 cwt/acre (160 crates or 3.5 tons per acre) and annual value of $13.7 million (USDA NASS, 2013). Indiana ranked 19th among states for production of fresh market sweet corn and produced about 0.8% of the nation’s total in 2012. The 2007 USDA Ag Census reported 603 Indiana farms producing sweet corn for fresh markets and 51 farms selling to processors. Sweet corn fields for fresh market sales are located throughout the state. In northern Indiana, bicolor corn is most commonly grown. …

Crowdsourcing services allow employing human intelligence in tasks that are difficult to accomplish with computers such as image tagging and data collection. At a relatively low monetary cost and through web interfaces such as Amazon’s Mechanical Turk (AMT), humans can act as a computational operator in large systems. Recent work has been conducted to build database management systems that can harness the crowd power in database operators, such as sort, join, count, etc. The fundamental problem of indexing within crowdsourced databases has not been studied. This project enabled students in an introductory biology course to learn biological information literacy. The section of the course described utilized a peer-led team learning (PLTL) model in which 50 students met twice a week. The students also divided into teams that met weekly for a workshop facilitated by a peer leader. The first time the course was taught in this way information literacy tasks were added to the weekly problem sets that the students worked on with their teams each week. By examining recorded footage of team meetings, it was determined that the students did not always see the …
