The effects of the presence of the mud snail Ilyanassa obsoleta (Say) upon the benthic community of Broad Bay, Virginia were examined through artificial removal and exclusion of the snail and artificial increase of the snail population. The enclosure treatment exhibited a significantly higher number of macrofaunal individuals than the control. The control had a significantly higher number of meiofaunal individuals than either treatment. There were no significant differences for the chlorophyll a. The increase in macrofauna and corresponding decrease in meiofauna were probably due to cage effects and not snail densities. The presence of the fences can act as …

N,N'-Diphenylguanidine (DPG) is a potentially toxic compound used in the vulcanization of rubber products. Distribution of DPG in hybrid mice (C57BL/6J interbred with DAB2) was determined following intraperitoneal injection. Urine, fecal material, whole blood and tissues were collected at various times after administration of DPG and the extracts analyzed for DPG by high pressure liquid chromatography. Recovery of DPG from spiked urine and whole blood exceed 90%, however, the recovery of DPG from spiked tissue samples was approximately 70%.

The evaluation of human semen is very important for diagnosis of male infertility. Combined with the analysis of seminal plasma, a comprehensive biochemical screening of human spermatozoan proteins may be of assistance in providing qualitative and quantitative information to the physician to assist with evaluation.

In this study, human spermatozoa were ruptured by sonication. The soluble spermatozoan proteins were separated from the cellular debris by centrifugation and then analyzed in dissociated forms by two dimensional electrophoresis. The separated proteins were visualized by a silver stain method.

The significance of sea nettle abundance on lower levels of the Chesapeake Bay food chain was examined in a field study and by the analysis of medusa gut contents. In the field study, the abundance of four levels of the food chain (Chlorophyll (a), copepods, the ctenophore Mnemiopsis leidyi, and the Schyphomedusa Chrysaora guinguecirrha) were monitored twice weekly at four stations from May 10 through September 30, 1982. The copepod standing stock declined sharply in late May when M. leidyi appeared, but rebounded a month later when C. guinguecirrha medusae reduced the ctenophore population. The spatial and temporal distribution of phytoplankton was measured in relation to frontal areas located in the entrance to the Chesapeake Bay. Phytoplankton biomass, taxonomy, and several physical parameters were measured on fifteen sampling cruises between September,1981 and February, 1983.

Several statistical tests revealed that phytoplankton biomass did not accumulate at the fronts and that generally the distribution of phytoplankton biomass was homogenous across the frontal region. The cell count data indicated that the community structure of the phytoplankton assemblage was usually very similar on both sides of the front. The objective of this study has been to develop an hypothesis explaining comparable aerial and underwater visual acuity of the bottlenose dolphin, Tursiops truncatus. This objective was accomplished by integrating the findings of other dolphin researchers working in the fields of comparative psychology, histology and ophthalmology. Their combined results enabled the formulation of a schematic dolphin eye. It is proposed that a dolphin lens, similar to the spherical lens of teleost fish with a rigid core of uniformly high refractive index and a margin of radially decreasing refractive index, could function as a bifocal lens and compensate for the eye's …

A study was conducted to determine the potential impact of open ocean disposal of sediments dredged from a highly industrialized seaport. Sediments from three potential dredge sites along the Southern Branch of the Elizabeth River, Hampton Roads, Virginia were evaluated for acute toxicity. The suspended particulate fractions of dredged materials were tested in a series of 96-hour, static bioassays using the calanoid copepod, Acartia tonsa Dana. Significant mortalities were observed for all three sites with the sediments from the most heavily industrialized site producing immediate and severe mortality. This study has particular application to the blue crab fisheries in Chesapeake Bay, an economically important industry whose successful management has been hindered by relatively poor understanding of the population dynamics. Recent biological studies have indicated a systematic spawning behavior which seems designed for ejection of larvae seaward from the bay entrance where they spend their pre-metamorphosis stages in the neuston. A physical mechanism for retention of the larvae in sufficient proximity to the bay entrance for their return at the proper time which involves the action of wind stress in shallow waters is proposed. The purpose of this research project was to develop assay procedures for determination of zinc in erythrocytes. Three methodologies were developed. The methods were evaluated on the basis of within-run and between-run precision. The most reproducible method, the 213.856 nm flame methodology, was used to determine red blood cell zinc concentrations in more than 200 samples obtained from the Neonatal Intensive care Unit at the Children's Hospital of the King's Daughters.

The caridean shrimp Hippolysmata wurdemanni Gibbes was studied from a histological and a behavioral basis and was found to be a protandric hermaphrodite which later adopts simultaneous functional hermaphroditism. When the animal reaches a certain size, the androgenic gland degenerates, the male secondary sexual characteristics are lost, and vitellogenesis occurs in the anterior portion of the gonad. The animals do not lose the ability to function as males as evidenced by the presence of mature sperm in the gonad and the gonadal lumen of gravid shrimp, and the ability of both members of an isolated pair of shrimp to repeatedly …

The in situ growth and feeding behavior of juvenile spot, Leiostomus xanthurus Lacepede, was investigated for populations in the nursery area of the James River, Virginia. Growth of juvenile spot was found to be fairly rapid (11.34 mm/month, standard length) during their first summer in the nursery grounds, but it levelled off in the autumn. The mean size of spot after the first year was calculated to be 195 mm total length. The weight-length relationship was log W = -5.018 + 3.246(log L), where W = wet weight in grams and L = standard length in millimeters.

It has been reported that the Java tilapia, Oreochromis mossambicus, displays hypersensitivity to a substance it produces when biomass levels in a flow-through culture system exceed 20 g/1, resulting in reduced growth and high mortality. Experiments on the growth of this species in small tanks were conducted in order to determine whether O. mossambicus produces a growth-inhibiting compound under crowded conditions. This species was successfully maintained at biomass levels of 38 g/1 and 57 g/1 with a total mortality of only 4.5%.

The purpose of this study was to determine whether homing occurred in the southern flying squirrel, Glauaomys volans, and if so, the greatest homing distance. Information on the squirrel's ecology, reproductive biology and use of artificial nest boxes was also obtained. The study was conducted from November 1978 through March 1981, using nest boxes placed in a deciduous forest in southeastern Virginia.

For the homing experiments, twenty squirrels were released a total of 29 times along three lines at distances of 250 m to 1.6 km from the homesites. Squirrels were found to home successfully up to 1,000 m.

Diversity and density of small rodents in the Dismal Swamp are believed to be low. Reasons for this may be excessive predation, heavy interspecific pressure from large rodents, lack of suitable habitat, low food availability or flooding.

Rodent populations were evaluated using live-traps and pitfall traps in four different Dismal Swamp plant communities. Habitat was compared on the basis of phytomass studies previously reported. Flood levels were recorded during live-trapping sessions. Mast from trees was collected in modified mast collectors, and fed to Peromyscus leucopus in the laboratory.

Nicotinamide adenine dinucleotide glycohydrolase (NADase) (E.C. 3.2.2.5) from human seminal plasma (HSP) was partially purified through a multistep purification procedure, including affinity chromatography on Amicon Red Matrex Gel A. The NADase activity was assayed by a fluorometric method with nicotinamide 1,N6 -ethenoadenine dinucleotide as the substrate. The enzymatic cleavage of etheno-NAD by NADase was accompanied by an eight-fold increase in fluorescence. This large increase in fluorescence provided a sensitive way to follow the rate of etheno-NAD hydrolysis.

NADase was found to be less specific for the purine moiety of NAD, since etheno-NAD was accepted as a substrate. The enzyme …

Little is known about the aquatic macroinvertebrate community structure in coastal salt marshes. This study investigated the aquatic macroinvertebrate community structure in a coastal Virginia salt marsh. Sixteen samples were taken weekly from two different sites on Smith Island, Virginia, from April to mid-October 1982. Bimonthly samples were taken during the months March and November due to low density of organisms and reduced growth rates at these times.

A total of 46 taxa were collected with 17 occurring regularly, indicating a relatively diverse fauna. Wide fluctuations in the physical parameters were observed; however, temporal changes in the macroinvertebrate community cannot …

Ocean Scallops (Placopecten magellanicus) were sampled from stations in and around the Philadelphia Disposal Site. Each sample was dissected and aliquots of adductor muscle and reproductive tissue were chemically extracted for Polychlorinated Biphenyls (PCBs). A perchlorinated procedure outlined by Diosady et al., 1972, Aromour, 1973 and Crist et al., 1977 was employed to increase detection levels and simplify quantitation of PCBs. PCB levels were reported in dry weight and lipid weight of both adductor muscle and reproductive tissue. Findings indicated residual levels or PCB/lipid weight increased as lipid levels decreased in both tissue types. Levels detected in all adductor muscle …

The phytoplankton that characterized the Smith Island pond was diverse, with several species of diatoms, dinoflagellates and blue-green algae dominant throughout the year. A total of 146 spp. were noted, predominantly composed of ultra and nanoplankters, that includes representatives from coastal waters. The island ponds are considered eutrophic habitats whose composition is influenced by overwash conditions and in turn introduce populations to surrounding pools and water channels.

Effects of storms on benthic infaunal communities have thus far been inferred rather than documented; especially lacking are studies examining immediate effects. To this end, the water column and 2 subtidal benthic sites were sampled before and after Storm David in September 1979. There were large post-storm increases in the numbers of infaunal species and individuals in the water column, presumably due to turbulent benthic boundary conditions. At the benthic stations, there were no pre vs. post-storm differences in the density of infauna; however, the number of species decreased at 1 station. An inexpensive, reliable thermistor device capable of monitoring infaunal foraging activity was built and used in the laboratory to examine effects of temperature on sediment reworking by the maldanld polychaete Clymenella torquata. The worm defecated at least once per hour 55.0 % (12.0 "C) to 88.2 % (5.5 "C) of the time. Defecation rate was linearly and positively related to temperature. Estimated volume of sediment ejected per defecation (0.015 ml) did not change with temperature. Results are compared with previous reworking studies of C. torquata.

Human Seminal Plasma was evaluated using a High Resolution Two-Dimensional Electrophoresis technique adapted by this laboratory. Seminal plasma from healthy volunteers with recently proven fertility were characterized using this technique to establish "normal" protein distribution patterns in the two-dimensional polyacrylamide gel slab. In addition, time studies were performed on selected specimens to determine the effects of liquefaction on seminal plasma protein composition within the first several hours after collection. Split ejaculates were collected for identification and elaboration of accessory organ components in the whole seminal plasma specimen.
