Fetal Whole-Body Fluid, Cardiovascular And Endocrine Dynamics : Computer Analysis Of Regulatory Mechanisms, Peter Stuart Gold

The dynamics of transcapillary fluid and protein distribution and endocrine regulation of cardiovascular function were studied in chronically instrumented fetal sheep during the third trimester. Average experimental changes in vascular pressures, blood volume, plasma hormone concentrations, hematocrit and plasma protein concentration were analyzed using mathematical modeling and parameter optimization techniques in order to determine values for the major parameters which regulate fluid, protein and endocrine dynamics and to determine endocrine dose-response relationships to changes in cardiovascular function. Selenium-Induced Enhancement Of Hematoporphyrin Derivative Phototoxicity In Murine Bladder Tumor Cells, Jeffrey M. Tosk

The phototoxicity of hematoporphyrin derivative (Hpd) to murine bladder tumor (MBT-2) cells was studied in vitro. It was observed that selenium in the form of sodium selenite enhanced Hpd-sensitized photodamage in MBT-2 cells under conditions where the selenite alone was non-toxic. Sodium selenite was found to enhance the fluorescence emission of Hpd and augment the Hpd-sensitized photooxidation of tryptophan. The data indicate that sodium selenite is able to disaggregate Hpd, thereby enhancing Hpd-sensitized phototoxicity.

Nine male marathon runners were exercised to exhaustion to determine the effects of a 27-hour fast on endurance performance. Each subject completed two exercise tests at 72% peak oxygen uptake (Vo₂max), one following a 27-hour fast and one three hours after a pre-exercise meal, in random order. Fasting caused a 44.7±5.8% (±SE) decrease in endurance performance (p2(9.3±2.0%), heart rate (8.4±2.4%), rating of perceived exertion (RPE), and psychological fatigue, evident within the first 60 minutes of exercise. Three local (Tamias panamintinus, Ammospermophilus leucurus and Spermophilus variegatus) and five extralocal (Tamias minimus, Tamias palmeri, Marmota flaviventris, Spermophilus townsendii, and Spermophilus lateralis) sciurid species (Sciuridae) were identified from the late Pleistocene-early Holocene Kokoweef Cave paleofauna. This fauna was the first documentation of T. minimus, T. palmeri and S. townsendii from the Pleistocene of the Mohave Desert. The 30-ft sedimentary sequence apparently recorded the mid-to late-Wisconsian introduction of S. lateralis into the Kokweef fauna but otherwise documented general taxonomic stability. The ecological diversity of the fauna was consistent with--but did not require--the hypothesis of more equable late Pleistocene climates, and …

A high percentage of fat in the diet has been implicated through epidemiological studies in several human cancers. Studies in experimental animals have produced evidence that increasing the fat in the diet decreases the animal’s ability to restrict tumor development and growth. To facilitate the study of the expression of hsd genes in Escherichia coli K, hsdK::MudX (amp crop lac) operon fusion mutants have successfully been constructed and characterized for the first time. The results indicate that Promoters 1 and 2 for hsdK genes appear to be relatively weak, as reflected by about 50 units of beta-gal activity. Both promoters were active in all the conditions tested except during Beta-gal activity was higher in aerobic than in anaerobic growth; at 30°C and 37°C than at 42°C; and in L-broth early log phase of the culture. than in minimal media. There is evidence that specific nutrients may modify hormonal balance and contribute to breast cancer etiology. To investigate the relationship between dietary nutrients and plasma reproductive hormones and to determine hormone-nutrient interrelationships, we measured plasma estrogens, androgens. progesterone and prolactin levels in 10 premenopausal Seventh-day Adventist (SDA) vegetarian and 10 premenopausal SDA nonvegetarian women. Over three days in each of three consecutive months. diet records and fasting midluteal blood samples were collected. The nonvegetarians consumed significantly more protein, total and saturated fats, oleic and linoleic acids, and cholesterol than did the vegetarians. Hormonal status of the two groups did not …

This thesis is based on a semiquantitative analysis of four nuclear relay centers [nucleus tractus solitarius (NTS), parabrachial nucleus (PBN), ventroposteromedial nucleus (VPM), gustatory neocortex (GNC)] in the chorda tympani nerve pathway. Metabolic changes were examined by the 2-deoxyglucose method in which one chorda tympani nerve was chemically stimulated and its contralateral side lesioned. Coronal sections of four taste centers were prepared for autoradiography and Nissl staining. Densities of the autoradiograms were determined by means of a video based image processor. A restriction map of DNA from bacteriophage P3-C₁ for the endonucleases BamH I, Bgl II, EcoR I, Hind III, Sac I and Sca I is described in this study. The phage DNA was determined to be a linear double stranded molecule with a unit genomic length of 31 kb. After electrophoresis in 0.5% agarose gels, the undigested DNA appeared as three molecules of 31, 62 and 93 kb in length. Sixty-four 35 day old Sprague Dawley Harlan rats were divided into four groups of 16 rats each and categorized as the control, orthopaedic expansion, surgery without graft and surgery with graft respectively. A general monomer-dimer equilibrium system involving ligand interactions is presented. Cooperativity features of specific limited models are analyzed by selecting the appropriate family of equilibrium constants from this general scheme. Each system is then characterized in terms of Hill coefficient dependency on alterations in values of equilibrium constants and total acceptor concentration. This method permits comparison of predicted cooperativity trends between systems. Contrasting reports concerning cooperativity dependencies for certain defined equilibrium systems are compared and the discrepancies resolved. The number of epithecal (external) growth lines was surveyed over 124 days for Montastrea annularis growing at different depths (3.5 m to 61 m). There was a significant (P< .01) decrease in the number of lines with depth. Although the linear regression is significant, there is evidence for a logarithmic or quadratic relationship, and more data from greater depths are needed to determine which of these gives a better fit.

The number of growth lines formed during light-dark cycles of 16, 24 and 32 hours were compared for M. annularis, M. cavernosa, Porites astreoides and Tubastrea aurea. The variability among colonies from the same species was in many cases more significant than that among light treatments. This is a serious problem for anyone counting lines in corals, especially from a few specimens; therefore conclusions based on such counts seem hazardous on …

Alcoholic Hyaline (AH) and keratin intermediate filaments possess ultrastructural, biochemical and immunochemical similarities. This study compares human AH, human stratum corneum keratin (HSCK) and normal human liver cytokeratin (HCK) by quantitative immunochemical means, using polyclonal as well as monoclonal antibodies. Enzyme-linked immunosorbent assays (ELISAs) were employed, as well as "immunoblotting" and immunocytochemical techniques. HCK was isolated using Triton X-100 and "high-salt” buffers and exhibited seven major polypeptide bands on SDS-PAGE (41-58 kd). Antisera were made toward HCK and the two major HCK bands. The physiologic role of the parotid glands is more complex than its recognized salivary function. Various investigators have suggested an intriguing relationship between the parotids and the pancreas, both endocrine and exocrine in nature.

We studied in rats the contribution of salivary glands in the regulation of β-cell function in response to an acute intra-arterial glucose challenge in parotidectomized, submandibulariectomized and totally sialoadenectomized animals. Fifteen days post-parotidectomy or total sialoadenectomy, the β-cell response, at 2 minutes post-glucose infusion, diminishes and stabilizes at about 40% of the preoperative value. The ⁵¹Chromium release assay was used to measure natural killer (NK) cell activity associated with Corynebacterium parvum (CP) injection against YAC-1 and MBT-2 targets. The peritoneal exudate cells from intraperitoneal treatment of CP at different dosages consistently gave higher cytotoxicity than their respective spleen cell fractions. In contrast, CP given intravenously showed the reversed effect. Our results demonstrate that the route of administration and dosage of CP are two crucial variables for determining the effectiveness of this immunostimulant. In vitro manipulations of nordihydroguaiaretic acid, indomethacin, and prostaglandin E2 on CP-stimulated NK cells suggest that the lipoxygenase pathway is involved …

Continuous administration of estrogens for 7-9 months, both steroidal and nonsteroidal, to male BALB/c mice, leads to the formation of testicular Leydig cell tumors. Three days following the subcutaneous implantation of a pellet of estrogen in cholesterol, there is a peak in the incorporation of ³H-tymidine into the DNA of the interstitial cells. These effects are hypothesized to be mediated by the estrogen receptor system in the Leydig cell.
