PURPOSE: A retrospective chart review was conducted to determine if nutritional source of feeding and/or infant age at diagnosis of cystic fibrosis had any effect on nutritional status and subsequent growth accretion. Additionally, an attempt was made to identify predictors for poor growth in patients with undiagnosed cystic fibrosis.

METHODS: Data was collected from medical and clinic charts at Primary Children's Medical Center (PCMC), Salt Lake City, Utah, for subjects born between January 1, 1995 and December 31, 200 I, who were diagnosed with cystic fibrosis before 1 year of age. Approximately 10% of cystic fibrosis (CF) cases remain undiagnosed until age 18 or later and medical professionals are still unclear about the needs of these individuals. Adults with CF must either adapt to life with a chronic disease or transition into adulthood with CF. The purpose of this study was to address the impact of age-at-diagnosis on quality of life in a CF adult population.

Suboptimal growth and nutritional status are problematic for children with cystic fibrosis (CF). Optimal nutrition predicts better lung function and longevity. Daily nutrition therapy for children with CF requires adequate food resources, knowledge of appropriate nutrition and behavior management skills, and confidence in one's ability to correctly apply the necessary skills. The Mountain West Cystic Fibrosis Consortium Questionnaire (MWCFC-Q) was designed as an educational needs assessment for parents of children with CF. The goal was to identify areas of concern that could be targeted for educational intervention to ultimately improve children's growth and nutritional status.

The purpose of these studies was to further research on bone density in children with cystic fibrosis, particularly as it pertains to nutritional parameters and care. The first paper presented a comparison of a group of 50 children with cystic fibrosis to a group of 32 control children. There were no significant differences between the groups in any of the pertinent bone density measurements. Serum 25(OH) vitamin D was positively correlated with spine density z score in the cystic fibrosis group.

Cell cultures can be used to study ion transport processes. X-ray microanalysis of cell cultures at the cellular level gives interesting information that can complement electrophysiological and tracer studies. In this paper, methods for culturing and preparing a variety of epithelial and secretory cells (fibroblasts, insulinoma cells, bovine mammary epithelial cells, colon cancer cells) for X-ray microanalysis are presented. Results show that sometimes cell cultures are not homogeneous with respect to ion content or reaction to physiological stimuli. In colon cancer cell cultures, a "high K" and a "low K" cell subpopulation was found; these subpopulations also differed with respect …

In an attempt to produce an animal model for the disease cystic fibrosis (CF), mice were treated chronically with the diuretics amiloride and furosemide, in order to cause chronic inhibition of transepithelial ion transport. Experiments were carried out on adult mice (2 months treatment); in addition, pregnant mice were treated with diuretics, and tissue from offspring 2 and 7 days post partum was investigated. Since biliary cirrhosis is a common occurrence in CF, hepatocytes in the treated mice were investigated by X-ray microanalysis and by light and electron microscopy.

Defective transepithelial chloride and water transport is thought to be the cellular basis of the disease cystic fibrosis (CF). Therefore, it was attempted to develop an animal model for this disease by chronically inhibiting transepithelial chloride transport in experimental animals by long term treatment with high doses of diuretics. In the present study, changes in the salivary glands and pancreas after such treatment were investigated by X-ray microanalysis and electron microscopy.

Since energy dispersive X-ray microanalysis cannot distinguish between isotopes of the same element, alternative methods have to be used to get information similar to that obtained in experiments with radioactive tracers. In the present study, strontium was used as a tracer for calcium, and bromide as a tracer for chloride. Rats were injected with strontium chloride in vivo, and the uptake of strontium in the acinar cells of the submandibular gland was studied. Eventually a Sr/Ca ratio of 0.3 was reached. Subcellular ion distribution in villus, crypt, Paneth and smooth muscle cells of the mouse small intestine under resting conditions was investigated by X-ray microanalysis of ultrathin cryosections. In addition, the mass distribution was estimated by measuring the optical transmission of the compartments in transmission electron micrographs.

Each cell type is characterized by a special composition in terms of the major monovalent ions Na, K, and Cl. In particular, among crypt epithelial cells, those cells containing small secretion granula (termed crypt A cells) also display cytoplasmic ion concentrations significantly different from crypt epithelial cells lacking secretion granula (crypt B cells). Obstruction of salivary glands occurs in association with a number of pathological conditions. It has been suggested that the major changes found in the salivary glands of patients with cystic fibrosis are due to obstruction of the excretory duct by viscous mucus. In the present study, the effect of excretory duct obstruction on structure, elemental composition and function of rat submandibular gland was investigated. Obstruction was effected by infusion of a fast-hardening protein emulsion in the main excretory duct. X-ray microanalysis was used to study the elemental composition of major salivary glands in rat and man. The elemental composition of the acinar cells is influenced by a variety of factors. Stimulation of fluid secretion by cholinergic, α-adrenergic or peptidergic agonists leads to a sustained increase of the Na/K ratio. The elemental composition and ultrastructure of the acinar cells of rat submandibular gland, as well as flow rate and composition of pilocarpinestimulated submandibular saliva are significantly affected by thyroxine, but less by the synthetic glucocorticoid dexamethasone. The study presents preliminary qualitative findings of an investigation of grossly normal main and lobar bronchi at sites distant to well circumscribed tumour (n=15), adjacent to tumour (n=5) or of airways obtained during heart/lung transplantation in patients with cystic fibrosis (CF, n=3). In the normal airways the surface epithelium was on average 50 𝝁m thick, pseudostratified and rested on a roughly contoured basement membrane. A variety of cell types were identified although many were obscured by a dense covering of cilia, occasionally interrupted by foci of squamous metaplasia. Cystic fibrosis (CF) is a generally lethal, congenital, genetic disease of unknown etiology. It is likely that a defective regulation of ion and water transport in exocrine glands and possibly also in other epithelial cells has a central role in the pathogenesis of this disease. Calcium has been implicated in the basic defect underlying CF because of findings of abnormally high calcium levels in some secreted fluids and some cells of CF patients.

Possible alternatives to rapid freezing in liquid nitrogen of tissue for X- ray microanalysis of electrolytes at the cellular level were investigated. These alternatives might be used in cases where tissue becomes available for examination, e.g., at autopsy, but liquid nitrogen is not immediately available. Rat submandibular gland was used as a test tissue. Freezing of pieces of tissue in a conventional freezer at -80°C or even at -20°C retained the elemental distribution at the cellular level, and also retained the difference between a 'normal' and a 'pathological' (mimicked by an inject ion of a high dose of isoproterenol) situation. …
