Leptin Differentially Regulate Stat3 Activation In Ob/Ob Mouse Adipose Mesenchymal Stem Cells, Zhou Zhou, Manish Neupane, Hui R. Zhou, Dayong Wu, Chia-Cheng Chang, Naima Moustaid-Moussa, Kate J. Claycomb Dec 2012

Leptin Differentially Regulate Stat3 Activation In Ob/Ob Mouse Adipose Mesenchymal Stem Cells, Zhou Zhou, Manish Neupane, Hui R. Zhou, Dayong Wu, Chia-Cheng Chang, Naima Moustaid-Moussa, Kate J. Claycomb

Animal Science Publications and Other Works

Background

Leptin-deficient ob/ob mice exhibit adipocyte hypertrophy and hyperplasia as well as elevated adipose tissue and systemic inflammation. Multipotent stem cells isolated from adult adipose tissue can differentiate into adipocytes ex vivo and thereby contribute toward increased adipocyte cell numbers, obesity, and inflamm ation. Currently, information is lacking regarding regulation of adipose stem cell numbers as well as leptin-induced inflammation and its signaling pathway in ob/ob mice.

Methods

Using leptin deficient ob/ob mice, we investigated whether leptin injection into ob/ob mice increases adipose stem cell numbers and adipose tissue inflammatory marker MCP-1 mRNA and secretion levels. We also determined leptin …

Dynamics Of Domain Coverage Of The Protein Sequence Universe, Bhanu Rekapalli, Kristin Wuichet, Gregory D. Peterson, Igor B. Zhulin

Microbiology Publications and Other Works

Background

The currently known protein sequence space consists of millions of sequences in public databases and is rapidly expanding. Assigning sequences to families leads to a better understanding of protein function and the nature of the protein universe. However, a large portion of the current protein space remains unassigned and is referred to as its “dark matter”.

Results

Here we suggest that true size of “dark matter” is much larger than stated by current definitions. We propose an approach to reducing the size of “dark matter” by identifying and subtracting regions in protein sequences that are not likely to contain …

Abundance Of Female-Biased And Paucity Of Male-Biased Somatically Expressed Genes On The Mouse X-Chromosome, Bjorn Reinius, Martin M. Johansson, Katarzyna J. Radomska, Edward H. Murrow, Gaurav K. Pandey, Chandrasekhar Kanduri, Rickard Sandburg, Robert W. Williams, Elena Jazin Nov 2012

Microbiology Publications and Other Works

Background

Empirical evaluations of sexually dimorphic expression of genes on the mammalian X-chromosome are needed to understand the evolutionary forces and the gene-regulatory mechanisms controlling this chromosome. We performed a large-scale sex-bias expression analysis of genes on the X-chromosome in six different somatic tissues from mouse.

Results

Our results show that the mouse X-chromosome is enriched with female-biased genes and depleted of male-biased genes. This suggests that feminisation as well as de-masculinisation of the X-chromosome has occurred in terms of gene expression in non-reproductive tissues. Several mechanisms may be responsible for the control of female-biased expression on chromosome X, and …

Mycobacterium Avium Subsp. Paratuberculosis Lipophilic Antigen Causes Crohn’S Disease-Type Necrotizing Colitis In Mice, Eiichi Momotani, Hiroshi Ozaki, Masatoshi Hori, Shizuo Yamamoto, Takashi Kuribayashi, Shigetoshi Eda, Masahiro Ikegami Nov 2012

Animal Science Publications and Other Works

Background: A 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced murine colitis model was developed to investigate the pathogenesis and to evaluate a method of treating human Crohn’s disease. This experimental model rapidly induces colitis similar to human Crohn’s disease lesion in a reproducible manner. However, natural exposure of the human digestive tract to TNBS is unrealistic. A novel animal model based on realistic data is eagerly anticipated in future research on pathogenesis of CD. Method: We evaluated the potency of Map antigen molecules in an effort to develop a novel colitis model using a more realistic source than TNBS. We prepared the Map …

Transgressive Hybdrids As Hopeful Monsters, Dylan R. Dittrich-Reed, Benjamin M. Fitzpatrick Nov 2012

Transgressive Hybdrids As Hopeful Monsters, Dylan R. Dittrich-Reed, Benjamin M. Fitzpatrick

Faculty Publications and Other Works -- Ecology and Evolutionary Biology

The origin of novelty is a critical subject for evolutionary biologists. Early geneticists speculated about the sudden appearance of new species via special macromutations, epitomized by Goldschmidt’s infamous “hopeful monster”. Although these ideas were easily dismissed by the insights of the Modern Synthesis, a lingering fascination with the possibility of sudden, dramatic change has persisted. Recent work on hybridization and gene exchange suggests an underappreciated mechanism for the sudden appearance of evolutionary novelty that is entirely consistent with the principles of modern population genetics. Genetic recombination in hybrids can produce transgressive phenotypes, “monstrous” phenotypes beyond the range of parental populations. …

Nanoparticle Biofabrication Using English Ivy (Hedera Helix), Jason N. Burris, Scott C. Lenaghan, Mingjun Zhang, C. N. Stewart Oct 2012

Nanoparticle Biofabrication Using English Ivy (Hedera Helix), Jason N. Burris, Scott C. Lenaghan, Mingjun Zhang, C. N. Stewart

Plant Sciences Publications and Other Works

Background

English ivy (Hedera helix) is well known for its adhesive properties and climbing ability. Essential to its ability to adhere to vertical surfaces is the secretion of a nanocomposite adhesive containing spherical nanoparticles, 60–85 nm in diameter, produced exclusively by root hairs present on adventitious roots. These organic nanoparticles have shown promise in biomedical and cosmetic applications, and represent a safer alternative to metal oxide nanoparticles currently available.

Results

It was discovered that the maximum adventitious root production was achieved by a 4 h application of 1 mg/ml indole-3 butyric acid (IBA) to juvenile English ivy shoot …

Sharing And Re-Use Of Phylogenetic Trees (And Associated Data) To Facilitate Synthesis, Arlin Stoltzfus, Brian C. O'Meara, Jamie Whitacre, Ross Mounce, Emily L. Gillespie, Sudhir Kumar, Dan F. Rosauer, Rutger A. Vos Oct 2012

Sharing And Re-Use Of Phylogenetic Trees (And Associated Data) To Facilitate Synthesis, Arlin Stoltzfus, Brian C. O'Meara, Jamie Whitacre, Ross Mounce, Emily L. Gillespie, Sudhir Kumar, Dan F. Rosauer, Rutger A. Vos

Faculty Publications and Other Works -- Ecology and Evolutionary Biology

Background

Recently, various evolution-related journals adopted policies to encourage or require archiving of phylogenetic trees and associated data. Such attention to practices that promote sharing of data reflects rapidly improving information technology, and rapidly expanding potential to use this technology to aggregate and link data from previously published research. Nevertheless, little is known about current practices, or best practices, for publishing trees and associated data so as to promote re-use.

Findings

Here we summarize results of an ongoing analysis of current practices for archiving phylogenetic trees and associated data, current practices of re-use, and current barriers to re-use. We find …
