This dissertation study investigates the impact of the US biofuel policies related to greenhouse gas (GHG) emission regulation, tax credit and renewable fuel standard (RFS2) mandate over production and consumption of ethanol as well as technical and environmental performance of corn ethanol plants. The study develops analytical models and provides quantitative estimation of the impact of various biofuel policies in each of the three chapters.

Chapter 1 of this dissertation examines the tradeoff between achieving the environmental goal of minimizing life cycle GHG emissions and minimizing production costs in recently built dry-grind corn ethanol plants. The results indicate that the …

Proline dehydrogenase catalyzes the FAD-dependent oxidation of proline to Δ1- pyrroline-5- carboxylate, which is the first step of proline catabolism. Here, we report the structures of proline dehydrogenase from Deinococcus radiodurans in the oxidized state complexed with the proline analog L-tetrahydrofuroic acid and in the reduced state with the proline site vacant. The analog binds against the si face of the FAD isoalloxazine and is protected from bulk solvent by the α8 helix and the β1-α1 loop. The FAD ribityl chain adopts two conformations in the E-S complex, which is unprecedented for flavoenzymes. One of the conformations is novel for …

Fusarium head blight (FHB), caused mainly by Fusarium graminearum, is a devastating disease of wheat and other small grain cereals. FHB lowers grain yield and quality and contaminates grain with mycotoxins, predominantly deoxynivalenol (DON) and its acetylated derivatives 3-ADON and 15-ADON. Forty one Fusarium isolates collected from grain elevators and wheat fields in Nebraska in 2009 and 2010 were sequenced for molecular identification. Forty isolates were identified as F. graminearum and one isolate was identified as F. culmorum. To develop a dispersive Raman spectroscopic method for measuring amylose-amylopectin ratios of corn starch mixtures, 67 mixtures were prepared by randomly mixing waxy and normal corn starches. Amylose contents were measured using a dual wavelength iodine binding colorimetric method. Raman data were collected from 250 to 3200 cm^-1 using optimized instrument parameters. Partial least-squares (PLS) and principal components regression (PCR) were used to prepare multivariate calibration models; however, PLS commonly outperformed PCR. Truncating the spectra to 250 to 2000 cm^-1 improved the results (r² of validation = 0.831, SEP = 2.90%). Removal of a cold water swelling …

Population characteristics and long-term population trends of pallid sturgeon Scaphirhynchus albus in the lower Missouri River are relatively unknown. As recovery efforts continue, understanding and quantifying these characteristics and trends are critical for species recovery and future management decisions. Therefore, the objectives of this study were to determine the pallid sturgeon population characteristics, predict changes to the pallid sturgeon population based on different management and life history scenarios, and examine trot line catch dynamics in the lower Missouri River. Glutaredoxin belongs to the oxidoreductase family with cytosolic glutaredoxin 1 (Grx1) and mitochondrial gluraredoxin 2 (Grx2) isoforms. Of the two isozymes, the function of Grx2 is not well understood. This paper studied the effect of Grx2 deletion on cellular function using primary lens epithelial cell cultures isolated from Grx2 gene knockout (KO) and wild type (WT) mice. We found that both cell types showed similar growth patterns and morphology, and comparable mitochondrial glutathione pool and complex I activity. Cells with deleted Grx2 did not show affected Grx1 or thioredoxin (Trx) expression but exhibited high sensitivity to oxidative stress. Under treatment …

The primary goal in development of at-risk species conservation assessments is to compile biological and ecological information that may assist conservation practitioners in making decisions regarding the conservation of species of interest. The Nebraska Natural Legacy Project recognizes the ghost tiger beetle, a.k.a. white tiger beetle, (Cicindela lepida) as a Tier I at-risk species of high conservation priority. Some general management recommendations are made here regarding ghost tiger beetles; however, conservation practitioners will need to use professional judgment to make specific management decisions based on objectives, location, and a multitude of variables. Object. Concussive head injuries have received much attention in the medical and public arenas, as concerns have been raised about the potential shortand long-term consequences of injuries sustained in sports and other activities. While many student athletes have required evaluation after concussion, the exact definition of concussion has varied among disciplines and over time. The authors used data gathered as part of a multiinstitutional longitudinal study of the biomechanics of head impacts in helmeted collegiate athletes to characterize what signs, symptoms, and clinical histories were used to designate players as having sustained concussions.

In Nebraska, Greater Sage-Grouse has been considered an uncommon casual winter visitor in the extreme northwest (Sharpe et al. 2001). This species may now be extirpated in Nebraska due to habitat loss. There have been no known Nebraska observations of Greater Sage-Grouse in the last 25 years. The only documented record accepted by the NOURC of Greater Sage-Grouse in Nebraska (Accession #243) is a description of an adult male in September 1987 in extreme northwest Sioux County by Douglas Kapke (Grenon AG. 1990).

Nitrous oxide (N₂O) is a greenhouse gas primarily produced in soils by denitrifying and nitrifying organisms. Agricultural soils account for 70% of emissions in the United States, but little data is available for contributions from managed pasture ecosystems. This study focused on the production of N₂O in smooth bromegrass (Bromus inermis Leyss.) pastures established on silt loam soils in eastern Nebraska. Thirty smooth bromegrass plots (1.5m x 1.5m) were treated with five different fertilizer treatments (0, 45, 90, 135, and 180 kg N/ha) and two urine treatments (urine and no urine). Previous works demonstrated that various species of Leishmania promastigotes exhibit differential sensitivity to complement-mediated lysis (CML) during development. Upon exposure to normal human serum (NHS), cultures of Leishmania chagasi promastigotes recently isolated from infected hamsters (fewer than 5 in vitro passages) are CML-sensitive when in the logarithmic growth phase but become CML-resistant upon transition to the stationary culture phase. Visualization by light and electron microscopy revealed dramatic morphological differences between promastigotes from the 2 culture phases following exposure to NHS. Flow cytometric analysis demonstrated that surface deposition of the complement components C3, C5, and C9 correlated inversely with promastigote CML-resistance. …

Evidence suggests that selenium has cancer preventive properties that are largely mediated through selenoproteins. Our previous observations demonstrated that targeted down-regulation of the 15 kDa selenoprotein (Sep15) in murine colon cancer cells resulted in the reversal of the cancer phenotype. The present study investigated the effect of Sep15 knockout in mice using a chemically-induced colon cancer model. Homozygous Sep15 knockout mice, and wild type littermate controls were given four weekly subcutaneous injections of azoxymethane (10 mg/kg). Short-horned grasshoppers (Orthoptera: Acrididae) were studied in the field as well as in laboratory for their distribution, identification, preferred food plants and feeding damage. These grasshoppers are important potential pests of rangelands where they compete for forage with cattle. Examination of the 20 most common grasshoppers by levels 3 and 4 of Nebraska ecoregions revealed specific habitat use and may be helpful in predicting hotspots. The relative abundance of all species analyzed showed at least some significant differences at ecoregion level 4 and two species showed single ecoregion that differed from all others at level 3.

Understanding factors structuring fish assemblages in a particular area is valuable to both sport fishery management and native species conservation. Fish assemblages in the Black Hills are unique to South Dakota because they contain economically valuable introduced salmonids as well as native species of conservation need. Our objective was to examine the relationship between fish assemblages and geomorphic and reach-scale habitat features across multiple stream reaches in the Black Hills. Canonical correspondence analysis, a direct gradient ordination analysis, indicated that factors operating at multiple spatial scales interacted to structure fish assemblages. Largemouth bass (Micropterus salmoides) are opportunistic predators often feeding in littoral areas during crepuscular periods (Snow 1971). Adult largemouth bass primarily are piscivorous, whereas juveniles generally consume zooplankton, insects, and small fish (Zweiacker and Summerfelt 1974, Carlander 1977, Post 2003). Age-0 largemouth bass generally consume zooplankton and macroinvertebrates (Keast and Webb 1966, Keast 1985, Phillips et al. 1995, Post 2003) and shift to piscivory at 50-70 mm in length (Phillips et al. 1995, Olson 1996, Post 2003).
