Agricultural and Environmental Sciences Faculty Research

This study was conducted to evaluate the effect of substitution of soybean meal (SBM) with dehulled-micronized peas (Pisum sativum) in diets of guinea fowl broilers on their growth performance, carcass yields, and fatty acid composition of meat. One hundred forty 1-d-old guinea fowl keets were randomly assigned to 2 dietary treatments, which were fed from hatch to 12 wk. The birds were fed 2 wheat middling-based diets comprising a control diet, which contained SBM (78 g/kg) and a test diet containing dehulled-micronized peas (180 g/kg) as the main protein source. The substitution of SBM with peas had no adverse effect …

Tributyltin And Dibutyltin Alter Secretion Of Tumor Necrosis Factor Alpha From Human Natural Killer Cells And A Mixture Of T Cells And Natural Killer Cells, Kelsi Hurt, Tasia Hurd-Brown, Margaret Whalen

Chemistry Faculty Research

Butyltins (BTs) have been in widespread use. Tributyltin (TBT) has been used as a biocide in a variety of applications and is found in human blood samples. Dibutyltin (DBT) has been used as a stabilizer in polyvinyl chloride plastics and as a de-worming agent in poultry. DBT, like TBT, is found in human blood. Human natural killer (NK) cells are the earliest defense against tumors and viral infections and secrete the cytokine tumor necrosis factor-alpha (TNF-α). TNF-α is an important regulator of adaptive and innate immune responses. TNF-α promotes inflammation and an association between malignant transformation and inflammation has been …

Aggregation Behavior Of The Southern Chinch Bug (Hemiptera: Blissidae), Karla M. Addesso, Heather J. Mcauslane, Ron Cherry

Agricultural and Environmental Sciences Faculty Research

The southern chinch bug, Blissus insularis Barber, forms dense, multigenerational aggregations in St. Augustinegrass lawns leading to grass death from sap feeding. We conducted laboratory bioassays to better understand the signals responsible for the formation and maintenance of southern chinch bug aggregations. In small arena assays, chinch bugs demonstrated a stronger aggregation response over time and aggregated more often on or beneath St. Augustinegrass leaf blades than on or under artificial leaf-like shelters constructed from white or green paper. In Y-tube olfactometer assays, bugs of different age and sex were attracted to volatiles from mixed-sex chinch bug aggregations and showed …

Effects Of Precipitation Increase On Soil Respiration: A Three-Year Field Experiment In Subtropical Forests In China, Qi Deng, Dafeng Hui, Deqiang Zhang, Guoyi Zhou, Juxiu Liu, Shizhong Liu, Guowei Chu, Jiong Li

Biology Faculty Research

Background

The aim of this study was to determine response patterns and mechanisms of soil respiration to precipitation increases in subtropical regions.

Methodology/Principal Findings

Field plots in three typical forests [i.e. pine forest (PF), broadleaf forest (BF), and pine and broadleaf mixed forest (MF)] in subtropical China were exposed under either Double Precipitation (DP) treatment or Ambient Precipitation (AP). Soil respiration, soil temperature, soil moisture, soil microbial biomass and fine root biomass were measured over three years. We tested whether precipitation treatments influenced the relationship of soil respiration rate (R) with soil temperature (T) and soil …

Molecular Identification Of Synanthedonini Members (Lepidoptera: Sesiidae) Using Cytochrome Oxidase I, Jason A. Hansen, William E. Klingeman, J. Kevin Moulton, Jason B. Oliver, Mark T. Windham, Aijun Zhang, Robert N. Trigiano

Agricultural and Environmental Sciences Faculty Research

Many North American sesiid moths within Synanthodonini have been studied extensively because their feeding activity can cause detrimental economic and esthetic impacts to many commercially important ornamental and native plant species. Recent discoveries of nonnative clearwing moth pest introductions [e.g., Synanthedon myopaeformis (Borkh.)], reinforce the need for reliable and accurate molecular diagnostic tools that can be used by nontaxonomic experts, particularly when juvenile life stages are recovered from infested host-plant tissues. Cytochrome oxidase I (cox I) previously has been used to successfully identify species and resolve species complexes. In this study, the cox I phylogeny inferred from sequences generated from …

Effects Of Ddt And Triclosan On Tumor-Cell Binding Capacity And Cell-Surface Protein Expression Of Human Natural Killer Cells, Tasia Hurd-Brown, Felicia Udoji, Tamara Martin, Margaret M. Whalen Jun 2012

Effects Of Ddt And Triclosan On Tumor-Cell Binding Capacity And Cell-Surface Protein Expression Of Human Natural Killer Cells, Tasia Hurd-Brown, Felicia Udoji, Tamara Martin, Margaret M. Whalen

Chemistry Faculty Research

1,1,1-trichloro-2,2-bis(4-chlorophenyl)ethane (DDT) and triclosan (TCS) are organochlorine (OC) compounds that contaminate the environment, are found in human blood and have been shown to decrease the tumor-cell killing (lytic) function of human natural killer (NK) cells. NK cells defend against tumor cells and virally infected cells. They bind to these targets, utilizing a variety of cell surface proteins. The present study examined concentrations of DDT and TCS that decrease lytic function for alteration of NK binding to tumor targets. Levels of either compound that caused loss of binding function were then examined for effects on expression of cell-surface proteins needed for …

‘Ramata’: A New Dwarf Variegated Hedychium (Ornamental Ginger) Cultivar, Hamidou F. Sakhanokho, Anthony L. Witcher, Cecil T. Pounders, James M. Spiers

Agricultural and Environmental Sciences Faculty Research

Most Hedychium J. Koenig (Zingiberaceae) species have showy and fragrant flowers, which are used in leis and perfumery. Hedychium species are easy to grow and are mostly free of major diseases and pests. The essential oils of some species have been found to have insecticidal, antibacterial, and antifungal activities (Gopanraj et al., 2005; Jadhav et al., 2007; Medeiros et al., 2003). Hedychium plants prefer moist, fertile soil and are considered to be shade plants but will thrive in full sun if provided adequate moisture, and many species will grow and bloom throughout USDA zone 7 (Adams, 1999). Hedychium plants are …

Lipid Peroxides And Glutathione Status In Human Progenitor Mononuclear (U937) Cells Following Exposure To Low Doses Of Nickel And Copper, William Y. Boadi, Shalandus Harris, Justin B. Anderson, Samuel E. Adunyah May 2012

Lipid Peroxides And Glutathione Status In Human Progenitor Mononuclear (U937) Cells Following Exposure To Low Doses Of Nickel And Copper, William Y. Boadi, Shalandus Harris, Justin B. Anderson, Samuel E. Adunyah

Chemistry Faculty Research

Effects of Cu2+, Ni2+ or Cu2+ + Ni2+ on lipid peroxide and glutathione (GSH) levels in U937 cells were investigated. Cells were treated with 0, 5, 10, and 20 µM of Cu2+ and/or Ni2+ and H2O2 (0.01 mM) and incubated for 24 hours at 37°C. Lipid peroxides were measured by the thiobarbituric acid assay (TBA). GSH intracellular levels were assayed by the GSH assay kit from EMD/Calbiochem (San Diego, California, USA). Cu2+ or Ni2+ significantly (P < 0.01) increased lipid peroxides in a dose-dependent manner, compared to controls. The effect was more pronounced for Cu2+, compared to the Ni2+-treated samples. Cu2+ + Ni2+ increased lipid peroxides in a significant (P < 0.001), dose-dependent manner, compared to Cu2+ or Ni2+ alone (i.e., ratio of 2.5:1-fold for combined versus single treatments, respectively). Cu2+ or Ni2+ significantly decreased GSH levels in U937 cells, with the effect being pronounced for Cu2+. Cu2+ + Ni2+ metal ions significantly (P < 0.001) depleted cells of GSH in a dose-dependent manner. Ethylene diamine tetraacetic acid (EDTA) at 50 or 100 µM moderately reduced the Cu2+- or Ni2+-induced effects on GSH levels. Interestingly, GSH levels generally decreased to half (except for the combined metal dose of 20 µM at 100 µM EDTA) of its level at the highest metal concentration tested for both the single or combined treatments. In conclusion, multiple exposures of cells to metal ions may be lethal to cells, compared to their single treatments.
