Anaesthetic drugs are commonly administered to fish in aquaculture, research and veterinary contexts. Anaesthesia causes temporary absence of consciousness and may reduce the stress and/or pain associated with handling and certain invasive procedures. The rainbow trout (Oncorhynchus mykiss) is a widely-used model species with relevance to both aquaculture and natural ecosystems. This study sought to establish the relative acute impact of commonly used anaesthetics on rainbow trout when used for anaesthesia or euthanasia by exploring their effects on aversion behaviour and stress physiology. A potentially painful experience may modify normal behavioural responses. To gauge the importance of pain relative to predation or social status, we presented competing stimuli, a predator cue or an unfamiliar social group, to two groups of noxiously treated rainbow trout, Oncorhynchus mykiss. In the predator cue experiment, fish were classified as bold or shy. Noxiously stimulated fish did not show antipredator responses, suggesting that pain is the imperative. In the social status experiment, noxiously stimulated fish held individually and undisturbed showed an increase in respiration rate and plasma cortisol. The effect of feeding frequency on nutrient digestibility and growth performance was investigated in rainbow trout (Oncorhynchus mykiss) fed a high lipid diet. The experimental diet contained 25.2% crude lipid and 40.7% crude protein. Two feeding frequencies were tested: 2 meals per day (Group I) and 6 meals per day (Group II). Fish were fed a restricted daily ration (1.5% of body weight) divided into equal meals. t the end of the experiment, the weight gain in Group I and II was 84.6% and 90.1%, respectively. The effects of diets including various natural carotenoid sources and synthetic astaxanthin on the pigmentation and growth of rainbow trout (Oncorhynchus mykiss) were investigated. Rainbow trout weighing about 111 g were fed diets containing 1.6%, 2.4% and 3.2% marigold flower; 4.4%, 6.6% and 8.8% red pepper; 100 mg kg^{-1} synthetic astaxanthin; and a control diet for 60 days. At the end of the experiment, synthetic astaxanthin provided the highest carotenoid accumulation in the fish flesh (6.42 mg kg^{-1}) in the groups. Hazelnut meal as a substitute for soybean meal in extruded feeds of rainbow trout was investigated. The performance of extruded feeds containing hazelnut meal as a substitute for 20% of soybean meal (Diet 2) and 30% of soybean meal (Diet 3) was compared to a nutritionally balanced control (Diet 1) (a commercial trout feed formulation). Leteux-Meyer mixture (LMM) contains formalin and malachite green and can be externally applied to fish as an antimicrobial agent. To determine the effects of prophylactic treatment with LMM on plasma lysozyme levels and the secondary stress indices, alterations in plasma glucose, chloride, sodium, potassium and hematocrit were monitored in healthy rainbow trout, Oncorhynchus mykiss W. Fish were i) exposed to 25 ppm Leteux-Meyer mixture for 60 min and ii) the same treatment followed by 60 min recovery in clean freshwater. The effects of melatonin hormone on rainbow trout (Oncorhynchus mykiss) erythrocyte carbonic anhydrase enzyme purified by using Sepharose 4B-L tyrosine-sulfanylamide affinity gel chromatography were investigated in vitro and in vivo. Carbonic anhydrase from Oncorhynchus mykiss erythrocytes was purified with a specific activity of ~400 EU/mg protein and ~200-fold in a yield of ~20. Melatonin did not inhibit carbonic anhydrase enzyme in vitro. Carbonic anhydrase activity of the control group was determined as 1597.7 ± 429.0 EU/g hemoglobin in vivo. Inhibitory effects of some antibiotics on glucose 6-phosphate dehydrogenase from the erythrocytes of rainbow trout (Oncorhynchus mykiss Walbaum, 1792) were investigated. For this purpose, initially erythrocyte glucose 6-phosphate dehydrogenase was purified 1271.19-fold in a yield of 70.40% by using ammonium sulfate precipitation and 2',5'-ADP Sepharose 4B affinity gel. A constant temperature (+4 ºC) was maintained during the purification process. Enzyme activity was determined with the Beutler method by using a spectrophotometer at 340 nm. This method was utilized for all kinetic studies. Thiamphenicol, amikacin, gentamicin, and netilmicin were used as antibiotics. All the antibiotics exhibited inhibitory effects on the enzyme. …

Many chemical anesthetics and drugs have been used in fish stripping, surgery and transportation, but their undesirable side effects on the body metabolism during treatment are not usually taken into consideration. In this study, we investigated the inhibition effect of some chemical anesthetics, such as Tranquil and tricaine methanesulfonate (MS-222), and hypothermia on glucose 6-phosphate dehydrogenase (G6PDH) activity. The aim of this study was to isolate Edwardsiella ictaluri from diseased rainbow trout obtained from a commercial farm in the Ankara region, Turkey. Growth of the microorganism after 36 h of incubation was higher and purer at 28 ºC than at 37 ºC on all the organs (liver, spleen, kidney) of the 10 fish examined. D1, D2 and D3 diets containing 15.5, 12.5 and 9.0 MJ kg^{-1} digestible energy concentrations were offered to rainbow trout (Oncorhynchus mykiss Walbaum, 1792), and the rates of gastric evacuation and feed intake were determined. Gastric evacuation determinations were achieved by slaughtering eight fish every 6 h until no residue was found in the cardiac stomach. Return of appetite experiments were conducted by re-feeding groups of trout every 6 h following the first feeding. Considering the relation between the endocrine and immune systems, the effects of zeranol, a synthetic oestrogen compound, on the growth rate and nonspecific immune system in rainbow trout (Oncorhynchus mykiss, Walbaum) were investigated. For this reason, fish were fed a diet containing 0, 2.5, 5, 10 and 20 ppm zeranol for 21 days. Fish were exposed to three different sublethal doses in order to determine the effects of Cypermethrin on some biochemical parameters (Ca-Calcium, P-Phosphorus, Na-Sodium and TP-Total Protein) of rainbow trout (Oncorhynchus mykiss). In this study, the population variability in chromosome numbers of rainbow trout (Oncorhynchus mykiss) cultured in 10 farms in the Northeast Black Sea Region of Turkey is examined. From the analysis of 468 anterior kidney tissue cells from 118 specimens, 7 distinct diploid numbers, namely 2n=58, 2n=59, 2n=60, 2n=61, 2n=62, 2n=63 and 2n=64, were found. The most frequent karyotypes with proportions of 25.2% and 24.8% were 2n=60 and 2n=62 respectively. Extensive Robertsonian polymorphisms were determined. Growth performances, daily food consumption and feed conversion ratios of rainbow (Onchorynchus mykiss) and brook trout (Salvelinus fontinalis) reared as single and mixed species were compared. During the experimental period, which lasted for about a growing season (December - June, 217 days), mean % weight increment of single species brook trout group were maximum (from 18.2 to 165.6 g), followed by mixed rainbow (26.8 to 241.6 g) and brook trout (17.0 to 151.8 g), and single species rainbow trout (27.5 to 207.6 g). The possibilities of replacing slaughterhouse byproduct meals (blood meal, poultry-by products, meat and bone meal) into menhaden fish meal in salmonid (rainbow trout fry) diets were economically analyzed in this study. Fish meal levels of diets were balanced as 30% and 45%, and 10 test diets with average 34% (first five diets) and 41% (last five diets) protein and about 3500 kcal/kg metabolic energy were made by replacing 0, 25, 50, 75 and 100% slaughterhouse byprolucts into these categories of fish meal. In this study reproductive performance of female rainbow trout broodstock ( Oncorhynchus mykiss) in a commercial trout farm in Eastern Black Sea Region were evaluated during 1995 spawning season. Mean total and relative fencundity values were 2304±425 egg/individual and 1364±281 egg/kg, respectively, while mean egg dia meter was 5.2±0.21 mm. Fertilisation rate on average was 73.7% of total fencundity. 84.6% of the fertilised eggs was eyed and 96.0% of those eyed was hatched.
