Detection Of Listeria Species In Minced Beef, Ufuk Tansel Şi̇reli̇, İrfan Erol

Turkish Journal of Veterinary & Animal Sciences

In recent years there has been an increase in the number of outbreaks of human listeriosis and contaminated foods have been considered to be a potential source of infection. In this study, a total of 100 minced beef samples were obtained from different butchers and supermarkets in Ankara and examined for the presence and degree of contamination with Listeria species. The minced beef samples were collected in aseptic conditions and transported to the laboratory in cold storage. The method suggested by USDA - FSIS was used for the isolation and identification of Listeria spp. In addition, the degree of contamination …

Isolation And Preparation Of Monoclonal Antibody To Ovineimmunoglobulin M, Hüseyi̇n Yilmaz, Aysan Tantaş, Ati̇lla Ilgaz, Kenton L. Morgan Jan 1999

Isolation And Preparation Of Monoclonal Antibody To Ovineimmunoglobulin M, Hüseyi̇n Yilmaz, Aysan Tantaş, Ati̇lla Ilgaz, Kenton L. Morgan

Turkish Journal of Veterinary & Animal Sciences

Ovine immunoglobulin M was isolated from sheep serum using gel filtration and affinity chromatography. Polyclonal and monoclonal antibodies against this isotype were prepared. The specificity of the monoclonal antibody was investigated using SDS-Page, immunoblotting, an enzyme-linked immunosorbent assay (ELISA) and affinity chromatography. On immunoblotting, monoclonal anti-IgM reacted with a 70 kD protein which corresponded to ovine IgM.

Isolation And Serological Characterization Of Highly Virulent Viruses Frominfectious Bursal Disease Outbreaks In Turkey, Olcay Türe, Fethi̇ye Çöven Jan 1999

Isolation And Serological Characterization Of Highly Virulent Viruses Frominfectious Bursal Disease Outbreaks In Turkey, Olcay Türe, Fethi̇ye Çöven

Turkish Journal of Veterinary & Animal Sciences

Infectious bursal disease (IBD) field viruses were isolated from the bursa Fabricius (BF) materials collected from the IBD infected or suspected flocks in different geographical regions of Turkey during 1992-1994. The field viruses were adapted to embryonated spesific-pathogen-free (SPF) eggs and chicken embryo fibroblast (CEF) cell cultures by serial blind passages and characterized by virus neutralization test (VN). In this study, out of 60 BF samples that were diagnosed positive for IBDV by agar gel precipitation (AGP) test, only 20 isolates adapted and grew in CEF cell cultures and showed cytopathic effect (CPE) typical to IBDV. During further characterization of …
