The Role Of B Cell Activation State And Sex In Aryl Hydrocarbon Receptor Mediated Induction Of Chemokine Receptor 9 And Alpha4beta7 Expression In Vitro, Logan Bauerle
Master's Theses
Defense of mucosal tissues from microbial infection and allergy is reliant on continual production of antibodies. The aryl hydrocarbon receptor (AhR) is known to regulate B cell development and is associated with suppression of systemic humoral immunity. Recent attention has been paid to the role of the AhR in altering expression of cell adhesion molecules (CAMs). B cells express CAMs and chemokine receptors to migrate around the body for localized secretion of antibodies. AhR agonists promote B cell migration to the small intestine through upregulation of chemokine receptor 9 (CCR9) and integrin α4β7. Both the AhR …
The Interaction Of Sodium Fluoride And Sodium Selenate On The Developmental Toxicity To Xenopus Laevis And Ambystoma Maculatum Embryos, Shelby Wolfram
The Interaction Of Sodium Fluoride And Sodium Selenate On The Developmental Toxicity To Xenopus Laevis And Ambystoma Maculatum Embryos, Shelby Wolfram
Theses
Amphibians play critical roles in the environment's wellness. Fluoride (F) is a widely existing environmental pollutant. It is commonly known for being added to drinking water and as a topical dose on teeth. Selenium (Se) can be found in organic and inorganic forms in nature. Many different fertilizers, pesticides, insecticides, and fungicides contain sodium selenate in them. The range selenium has between deficiency and toxicity is narrow, meaning excess exposure could cause various adverse effects in aquatic organisms. Xenopus laevis is a standard model for developmental toxicity due to being minimal maintenance, cost efficient, and having transparent embryos. Xenopus makes …
Quantum Dots' Toxicity: A Multi-Level Investigation On The Impact Of Quantum Dots On The Actin Cytoskeleton, Nhi Le
MSU Graduate Theses
Quantum dots (QDs) are fluorescence nanomaterials with unique optical and physical properties. As such, they are highly sought after for their potential use in several biomedical and industrial applications. Despite their vast potential, recent studies have suggested that quantum dots are toxic to cells. Yet, the mechanism of quantum dots’ toxicity remains unclear. As such, my thesis aims to comprehensively examine the mechanism of quantum dots’ toxicity, emphasizing how quantum dots disrupt the actin cytoskeleton. In this study, I used RNA sequencing and mass spectrometry to investigate the influence of CdSe/ZnS QDs on the transcriptomic proteomic level of Saccharomyces cerevisiae …
Determination Of Genetic Variation In Opioid-Resistant Hospice Patients Using A Pharmacogenomics Approach, Daniel Bianculli
Determination Of Genetic Variation In Opioid-Resistant Hospice Patients Using A Pharmacogenomics Approach, Daniel Bianculli
Theses and Dissertations
Managing pain requires a delicate balance between the advantages of opioid analgesia and the possible adverse effects. This balance becomes even more difficult in hospice and palliative care environments where dosage regimens are frequently ambiguous. Some hospice patients experience opioid resistance, in which opioids no longer produce an analgesic effect on a patient. Evidence-based analyses frequently omit this vulnerable population, resulting in a need for concrete procedures. The emergence of pharmacogenomic analysis and personalized treatment presents a solution to this problem by using each patient's genomic data to interpret opiate dosing. This pharmacogenomic study aimed to single nucleotide polymorphisms (SNPs) …
Assessing Stormwater Management Pond Water Quality, Function, And The Potential Biotic Effects To Receiving Waters, Mitchell Elstone
Assessing Stormwater Management Pond Water Quality, Function, And The Potential Biotic Effects To Receiving Waters, Mitchell Elstone
Theses and Dissertations (Comprehensive)
The use of stormwater management ponds (SWMPs) has been increasing over the past five decades. However, an in-depth understanding of the daily performance of SWMPs and functionality during cold periods is limited. This is in part because mandated monitoring is relatively infrequent, and the assumption that SWMPs are inactive between storm events and during the winter. The goals of this research were to better understand daily stormwater (SW) characteristics, the performance of SWMPs based on current forms of evaluation and assess the potential for SWMP effluent to impact downstream biota. Influent and effluent samples from two SWMPs were collected daily …

Impact Of Cadmium Stress On Soil Virus Reproduction And The Persistence Of Viruses Under Abiotic Conditions, Zhibo Cheng
Doctoral Dissertations
Soil viruses are ubiquitous and greatly impact the structure and function of soil microbial communities, with their effects modulated by various environmental factors. This study investigates the inactivation of naturally occurring soil viruses in sterilized soil, as well as the effects of cadmium (Cd) exposure and carbon (C) and nitrogen (N) nutrient availability on the population dynamics of virus-host communities in agricultural soil.
Lab-scale slurry and unsaturated experiments were performed to examine virus inactivation in the absence of host bacteria in sterilized soil. In slurry experiments, virus abundance declined by over 90% after 10 days of incubation. The addition of …
Using A Toxic Aging Coin To Assess Hexavalent Chromium-Induced Neurotoxicity., Samuel Thomas Vielee
Using A Toxic Aging Coin To Assess Hexavalent Chromium-Induced Neurotoxicity., Samuel Thomas Vielee
Electronic Theses and Dissertations
We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. We address this crisis using a ‘toxic aging coin’ approach; heads examines how age impacts toxicity, tails examines how chemicals accelerate aging. This thesis applies the heads side for Cr(VI)-induced neurotoxicity across ages. We …

The Method Development And Validation Of Designer Benzodiazepines In Oral Fluid Using Solid Phase Extraction And Lc-Ms/Ms, Jenna L. Monsell
Student Theses
Oral fluid is a useful biological matrix. Collection is non-invasive, quick, observable, and its correlation between oral fluid and blood drug concentration make oral fluid an important matrix to use as a roadside test. Designer benzodiazepines have become increasingly popular over the years, adversely affecting motor skills and leading to driving under the influence. Many previous studies have focused on the method development and validation of designer benzodiazepines in blood and hair but fail to validate a method to detect designer benzodiazepines in oral fluid. This study focuses on the method validation of 13 designer benzodiazepines in oral fluid using …
The Impact Of Environmental Pollutants On Alcohol-Associated Liver Disease Pathogenesis., Tyler Charles Gripshover
The Impact Of Environmental Pollutants On Alcohol-Associated Liver Disease Pathogenesis., Tyler Charles Gripshover
Electronic Theses and Dissertations
Steatotic liver disease is brought upon by differing etiologies and its global prevalence is expected to keep increasing over the next several decades. Particularly, alcohol-associated liver disease (ALD) manifests from excessive alcohol consumption where the liver retains lipids and progresses with metabolic dysfunction. Previous studies have demonstrated that environmental pollutants, polychlorinated biphenyls (PCBs) promote metabolic dysfunction associated steatotic liver disease (MASLD), in diet-induced obesity models. However, it is unknown if environmental pollutants may also promote ALD pathogenesis. Thus, this dissertation focuses on characterizing the effects of PCB126 in a rodent ALD model followed by multiple ‘Omics approaches to identify mechanisms …
Hair And Drugs: The Impact Of Dry Shampoo And Gel On Cocaine-Contaminated Hair, Marialejandra Faure Betancourt
Hair And Drugs: The Impact Of Dry Shampoo And Gel On Cocaine-Contaminated Hair, Marialejandra Faure Betancourt
Student Theses
Hair is a biological sample used in forensic toxicology to identify and quantify drug exposure. However, when interpreting analytical results, hair can be challenging due to the presence of external contamination. Previous studies have shown that cosmetic treatments remove drugs within the hair shaft decreasing their concentration. Nevertheless, not enough information is available to explain this phenomenon. This study focused to determine whether cosmetic products such as dry shampoo and gel influence the concentration of externally deposited cocaine (COC) and benzoylecgonine (BE) in different hair types/colors. Hair samples from fifteen COC-free volunteers of various hair colors/types were collected and divided …

Toxicity Of Different Insecticides To Sitophilus Oryzae, Tribolium Castaeneum And Tribolium Confusum Infesting Corn, Amina Twaibu
Graduate Theses and Dissertations
Corn is an important cereal crop cultivated across various countries, including the United States. Corn serves multiple purposes such as human and livestock food and raw material for industrial processes. Despite being an important crop, corn production faces challenges due to factors such as weather fluctuations and the presence of insect pests and diseases, both in the field and during storage. Among these challenges, insect infestation poses a major threat, as the insect damage not only reduces the quantity and quality of the grain but also renders it unfit for human and animal consumption, leading to a decrease in market …

Epitranscriptomic Regulation In Breast Cancer And Pcb-Induced Liver Disease., Belinda Petri
Electronic Theses and Dissertations
Post-transcriptional RNA modifications including N6-methyladenosine (m6A) regulate mRNA stability, splicing, and translation. My research examined m6A in two disease models: breast cancer (BCa) and non-alcoholic fatty liver disease (NAFLD). Acquired resistance to endocrine therapies (ET) develops in approximately 20% of BCa patients with estrogen receptor α positive (ER+) tumors following treatment. The mechanisms by which tumor cells evade ET are not completely understood. Using a cell line model, we investigated the role of an m6A reader protein, HNRNPA2B1 (A2B1) that is upregulated in ET-resistant ER+ BCa cells. Stable overexpression of A2B1 in ET-sensitive MCF-7 cells (MCF-7-A2B1), results in ET resistance, …
The Role Of E-Liquid Constituents In E-Cigarette-Induced Cardiac Arrhythmia And Autonomic Imbalance., Cory Kucera
The Role Of E-Liquid Constituents In E-Cigarette-Induced Cardiac Arrhythmia And Autonomic Imbalance., Cory Kucera
Electronic Theses and Dissertations
Introduction. Accumulating evidence indicates that exposure to electronic cigarettes (e-cigs) promotes sympathetic dominance and electrophysiologic instability in the heart, potentially culminating in arrhythmogenesis. E-liquids contain various formulations of nicotine and flavorings, but the effects of specific e-liquid constituents and their concentrations on e-cig-induced autonomic imbalance and electrical dysfunction are unresolved. To that end, we tested the hypothesis that e-cigs modify cardiac autonomic balance and ventricular arrhythmogenesis in mice dependent on e-liquid constituent type and concentration. Methods. Using a crossover design and a serial exposure regimen, ECG-telemetered male C57BL/6J mice underwent whole-body exposure to e-cig aerosols generated from e-liquids containing different …

Neuroinflammatory And Metabolomic Temporal Dynamics From Inhaled Wildfire Smoke With Attenuation Via Pharmacological Intervention, David Scieszka, David Scieszka
Biomedical Sciences ETDs
Wildfire smoke is a global issue affecting nearly all humans on the surface of this Earth. Smoke dispersion from wildfires can span continents and cross oceans with amazing ease. Furthermore, total acres of land burned per year in the United States has roughly doubled since 1980, meaning this issue will persist for an indeterminate amount of time. Exposure to wildfire smoke is hazardous and promotes aging-associated events in the cardiopulmonary systems. Yet long- and short-term effects on the brain remain relatively unknown. From these, it is critically important to understand the neurological impacts from wildfire smoke exposure; to compare these …
Interspecies Differences In Organophosphate Anticholinesterase Inhibition Potency And Reactivation Using Novel Oximes, Katie Elizabeth Strickland
Interspecies Differences In Organophosphate Anticholinesterase Inhibition Potency And Reactivation Using Novel Oximes, Katie Elizabeth Strickland
Theses and Dissertations
Organophosphates are insecticides which result in acute adverse signs when exposed at toxic doses by animals and lead to death if left untreated. The current treatment for organophosphate toxicity includes atropine and the federally approved oxime 2-PAM. However, 2-PAM is not very effective at crossing the blood brain barrier which results in prolonged inactivation of acetylcholinesterase, which is the primary target of organophosphates, in the brain even after administration. The novel oximes, Oxime 15 and Oxime 20, are able to cross the blood brain barrier and reactivate the inhibited acetylcholinesterase. In this experiment with six animal species frequently used in …

Role Of Salinity In Perfluorooctane Sulfonate (Pfos) Bioconcentration Between Aquatic Species And The Differential Regulation Of Transporters In Fundulus Heteroclitus, Tyler Davis
All Theses
Perfluorooctane sulfonate (PFOS) is a persistent organic pollutant that is known for its surfactant properties and its resistance to degradation. The persistence and lipophilicity of PFOS in surface waters have led to its bioaccumulation in fish, which in turn leads to its accumulation and toxic effects in humans that consume fish. PFOS is also hypothesized to perturb reproduction and survival of low and mid-trophic level zooplankton species such as Daphnia magna that are food for multiple fish species. In this study, a 21-day D. magna chronic toxicity test was performed using 1,10, 100, 1000, and 5000 ppb of PFOS to …
Mechanisms Of Cigarette Smoke-Induced Mitochondrial Dysfunction In Striated Muscle And Aorta, Stephen T. Decker
Mechanisms Of Cigarette Smoke-Induced Mitochondrial Dysfunction In Striated Muscle And Aorta, Stephen T. Decker
Doctoral Dissertations
Cigarette Smoke is a significant cause of morbidity and mortality in the United States, accounting for over 480,000 annual deaths. Of these deaths, the most common cause of mortality in chronic smokers is cardiometabolic diseases. Likewise, a significant portion of smokers experience some form of cardiac, vascular, or metabolic dysfunction throughout their lifetime. More specifically, smoking is shown to induce mitochondrial dysfunction in these tissues, causing an increase in oxidative damage and poor overall health. However, despite the advances in the health outcomes related to cigarette smoke exposure, the mechanisms underlying mitochondrial dysfunction in striated muscle and the vasculature remain …

Undergraduate Theses, Professional Papers, and Capstone Artifacts
Mining has left massive environmental and physical scars across the landscape. Aquatic and riparian landscapes in particular have been significantly impacted by traditional mining practices. Waste products left over from hard-rock mining leech heavy metals onto the landscape and these metals spread from headwater streams to major waterways (Lottermoser 2010). Heavy metals have been shown to cause physiological stress and challenges to organisms depending on the metal and the concentration (Baos et al. 2019, Boyd & Rajakaruna 2013). While some mining-impaired areas have undergone restoration efforts, is it enough? Typical restoration methods replace the contaminated floodplain, but not the riverbed …

Characterizing The Effects Of Antiandrogens And Senolytics To Enhance The Therapeutic Response To Castration-Resistant Prostate Cancer, Justin M. Silverman
Theses and Dissertations
Prostate cancer is the most frequently diagnosed cancer in males and the second most common cause of cancer deaths. Androgen deprivation therapy, whether through surgical or chemical castration, is the mainstay for treatment of advanced prostate cancer; however, despite an initial response, most patients eventually develop a progressive PSA rise, and castration- sensitive prostate cancer gives rise to castration-resistant prostate cancer. The standard of care therapy includes the antiandrogens such as enzalutamide and abiraterone acetate as well as the microtubule poison, docetaxel, and various immunotherapies; however, while prostate cancer research is progressing, there continues to be a compelling need for …
