Brassinosteroids (BRs) have remarkable ability to increase stress tolerance in plants. Investigations to understand the molecular mechanisms underlying BR-mediated stress tolerance resulted in identification of genes belonging to the family calmodulin binding protein X (CBPX). The present study was focused on studying the role of CBPX1 and CBPX2 in BR mediated stress tolerance and functional characterization using a reverse genetic approach. The upregulation of CBPX1 and CBPX2 by BR and stress noted in publicly available AtGenexpress datasets and by qRT-PCR analysis strongly suggests that these are BR responsive genes and functional analysis of T-DNA insertion mutants showed salt stress related …

Identification Of Cyclophilin Gene Family In Soybean And Characterization Of Gmcyp1, Hemanta Raj Mainali Jul 2013

Identification Of Cyclophilin Gene Family In Soybean And Characterization Of Gmcyp1, Hemanta Raj Mainali

Electronic Thesis and Dissertation Repository

I identified members of the Cyclophilin (CYP) gene family in soybean (Glycine max) and characterized the GmCYP1, one of the members of soybean CYP. CYPs belong to the immunophilin superfamily with peptidyl-prolyl cis-trans isomerase (PPIase) activity. PPIase catalyzes the interconversion of the cis- and trans-rotamers of the peptidyl-prolyl amide bond of peptides. After extensive data mining, I identified 62 different CYP genes in soybean (GmCYP1 to GmCYP62), of which 8 are multi-domain proteins and 54 are single domain proteins. At least 25% of the GmCYP genes are expressed in soybean. GmCYP1 …

Brassinosteroid-Mediated Stress Tolerance In Arabidopsis Thaliana, Tawhidur Rahman Oct 2011

Brassinosteroid-Mediated Stress Tolerance In Arabidopsis Thaliana, Tawhidur Rahman

Brassinosteroids (BRs) are a group of steroidal plant hormones that are essential for proper plant development and also promote stress tolerance. Without BRs, plants are dwarfs and infertile. To understand the molecular mechanisms underlying BR-mediated stress tolerance, global gene expression analysis of untreated and 24-epibrassinolide (EBR)-treated Arabidopsis thaliana seedlings under non-stress and heat stress (HS) conditions was carried out. Microarray data analysis indicated that stress-related genes were predominant within the EBR up-regulated gene data set. Furthermore, several of these genes were abscisic acid (ABA) and jasmonic acid (JA) related. Measurements of endogenous hormones showed significant increases in the levels ABA …
