To test whether SKF 38393 could ontogenetically sensitize dopamine (DA) D$\sb1$ receptors and whether this sensitization would be associated with biochemical changes, intact and neonatal 6-hydroxydopamine (6-OHDA)-lesioned rats (200 $\mu$g i.c.v.) were treated daily from birth with SKF 38393 (3.0 mg/kg i.p. x 28 days) or its vehicle. In DA D$\sb1$ neonatally sensitized 6-OHDA rats, enhanced locomotor responses were observed with the first SKF 38393 challenge dose (3.0 mg/kg i.p.) at 6 weeks. This response increased further with weekly SKF 38393 treatments. Enhanced stereotyped behaviors were seen in both lesioned and sensitized rats at 8 weeks. There was no change …

At this time there is no NIOSH approved means for bag sampling 1,3-butadiene. An attempt was made to use the application of cold (0°F) to inhibit the reactivity of the gas prior to analysis. The State of California Air Resources Board has an approved method for the gas chromatographic analysis of 1,3-butadiene from stationary sources. The method was utilized in the application of industrial hygiene monitoring of affected workers. The application of cold was found to be ineffective at inhibiting the highly reactive nature of 1,3-butadiene. Analyte losses were too great to classify the method as an accurate measure of …

Effects Of Portuguese Man-Of-War (Physalia Physalis) Venom On Isolated Rabbit Atria, Wayne R. Bonlie

The cardiac effects of Portuguese man-of-war venom were studied using isolated atria from rabbits. Venom added (7.1 µg protein/ml) to the bath solution stimulates heart rate 120% (P < 0.01) and increases contraction amplitude 308% (P < 0.001). Both effects are dependent on bath concentrations of venom and calcium. The increase in contraction amplitude is significantly attenuated by diltiazem (0.8 µM) and atenolol (10 µM). Atenolol, a β1-adrenergic antagonist, completely blocks the increase in heart rate. Dantrolene (10 µM) did not significantly block the venom's effects. Ryanodine binding increases in the presence of venom showing that the venom is not in competition for calcium channels on the sarcoplasmic reticulum. These results suggest that the venom acts on the sarcolemma to increase calcium permeability, thereby causing an increase in contraction amplitude and possibly causing β1-adrenergic stimulation.

In April, 1991 the Administrator of the U.S. Environmental Protection Agency (EPA) announced that the Agency would conduct a scientific reassessment of the risk of 2,3,7,8-tetrachlorodibenzo-p-dioxin (hereafter referred to as TCDD), and similar chemicals, to human health and the environment. Since 1985, EPA has classified TCDD, which it considers the most potent known animal carcinogen, as a probable human carcinogen. Sources of TCDD in the environment were subsequently regulated on the basis of animal cancer rates extrapolated to doses associated with human exposures. Recently, consensus has developed that the toxic effects of TCDD appear to be mediated by its binding …

The in vivo toxicokinetics and in vitro hepatic microsomal metabolism of (14C)aniline and (14C)4-chloroaniline in medaka (Oryzias latipes) were investigated to provide a basis upon which to interpret the toxicological responses of small aquarium fish to aniline derivatives. During static aqueous exposures of up to 320 min, parent equivalents failed to reach steady state and results from depuration studies clearly demonstrated biphasic elimination. Due to low elimination rates, 40 to 20% of absorbed aniline and 4-chloroaniline doses, respectively, remained within the fish through 330 min postexposure. A Non-Invasive Neurotoxicity Assay Using Larval Medaka, Steven P. Bradbury, Dave Featherstone, Charles D. Drewes, Joel R. Coats

We present a method for non-invasive electrophysiological analysis of rapid escape responses in intact, freely behaving larval medaka (Oryzias latipes) before and after short-term exposure to environmental toxicants. Recordings are obtained as a larval medaka swims in a small chamber of water above pairs of recording electrodes etched onto a printed circuit board. When the chamber is briefly vibrated by a sinusoidal pulse from a speaker attached to the grid, a stereotyped Mauthner cell-initiated escape response is evoked. Cell-mediated lysis of cultured tumor target cells by nonspecific cytotoxic cells (NCC) was examined in the oyster toadfish (Opsanus tau), an estuarine teleost. NCC activity was evaluated in cells taken from the head kidney, peripheral blood, spleen and peritoneal cavity. NCC activity was a property of plastic nonadherent cells which lacked phagocytic activity, indicating that in terms of their functional capacity, the cells which mediate nonspecific cytoxic immune responses in oyster toadfish do not appear to be monocytes or macrophages. However, light and electron microscopic examination of these cells revealed that morphologically, they resemble monocytes and macrophages. A new technique …
