Ph+ ALL is a poor-prognosis leukemia subtype driven by the BCR-ABL1 oncogene, either the p190-or the p210-BCR/ABL isoform in a 70:30 ratio. Tyrosine Kinase inhibitors (TKIs) are the drugs of choice in the therapy of Ph+ ALL. In combination with standard chemotherapy, TKIs have markedly improved the outcome of Ph+ ALL, in particular if this treatment is followed by bone marrow transplantation. However, resistance to TKIs develops with high frequency, causing leukemia relapse that results in

ABSTRACT: Sorafenib, a kinase inhibitor, is among the approved drugs for the treatment of radioactive iodine resistant thyroid carcinoma, primary kidney and liver cancers. Reported targets of Sorafenib include VEGFR, Raf family, and PDGFR belonging to the general class of tyrosine kinases. Blocking growth signals in kidney and breast cancers underlie one of the mechanisms of Sorafenib antitumor effects’ leading to cell death. We hereby examine the effect of Sorafenib on human colon carcinoma cell-line HCT116. We also investigate the possible role of p53 in mediating this effect using mutant HCT116 p53-/- cells. Cultured wild and mutant cells are treated …

Cancer cells almost universally harbor constitutively active Phosphatidylinositol-3 Kinase (PI3K) Pathway ac-tivity via mutation of key signaling components and/or epigenetic mechanisms. Scores of PI3K Pathway in-hibitors are currently under investigation as putative chemotherapeutics. However, feedback and stem cell mechanisms induced by PI3K Pathway inhibition can lead to reduced treatment efficacy. To address therapeutic barriers, we examined whether JAKi would reduce stem gene expression in a setting of PI3K Pathway inhibition in order to improve treatment efficacy. We targeted the PI3K Pathway with NVP-BEZ235 (dual PI3K and mTOR inhibitor) in combination with the Janus Kinase inhibitor JAKi in glioblastoma (GBM) and …

Faculty Research 2021

OBJECTIVE: This study aimed to investigate the effects of multiwalled carbon nanotubes (MWNTs) co-delivering sorafenib (Sor) and epidermal growth factor receptor (EGFR) siRNA (MWNT/Sor/siRNA) on tumor growth in liver cancer (LC).

RESULTS: MWNT/Sor/siRNA was proved to possess increased Sor release, high siRNA stability, and enhanced cellular uptake. In addition, MWNT treatment has few effects on cell proliferation and apoptosis in HepG2 cells; however, MWNT/Sor/siRNA treatment significantly inhibited clone number and induced cell apoptosis, which shows a more favorable antitumor effect than MWNT/Sor and free Sor and free siRNA in HepG2 cells. Moreover MWNT/Sor/siRNA treatment has the most significant antitumor effect …

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was to investigate the effects of feed restriction on histopathological changes associated with white striping and wooden breast myopathies in pectoralis major muscle and meat quality characteristics in broiler chickens. In total, 600 male broiler chicks were randomly divided into five groups with 8 replicate pens (15 chicks/pen). Treatments consisted of control, treatment 1 (T1), treatment 2 (T2), treatment 3 (T3) and treatment 4 (T4). Control group was fed ad libitum throughout experimental period. T1 and T2 groups received feed as 80% and 70% of amount received by the control group from 11 to 24 …

The objective of this study was to investigate the effects of supplementation of beta-glucan to the basal diet of laying quails exposed to lead (Pb) toxicity. A total of 112 birds were randomly divided into 4 groups. The first group was fed a corn-soybean based basal diet without any supplementation (control). The second (Pb) and third (beta-glucan) groups were supplemented with 100 mg/kg of Pb (as Pb acetate) and 100 mg/kg of beta-glucan, respectively. The fourth group (Pb + beta-glucan) was fed a basal diet supplemented with 100 mg/kg of Pb and 100 mg/kg of beta-glucan. It was determined that …

Cellular Bioenergetics Regulates Cell Proliferation During Mammalian Regeneration, Sandeep Saxena Jan 2021

Cellular Bioenergetics Regulates Cell Proliferation During Mammalian Regeneration, Sandeep Saxena

Theses and Dissertations--Biology

Mammalian system consists of stress-sensing molecules that regulates their cellular response against damage, injury and oncogenic stress. During vertebrate regeneration, cells responding to injury re-enter the cell cycle and proliferate to form new tissue. Cell cycle re-entry or arrest is at least partly regulated by cellular senescence which negatively impacts the proliferative pool of cells during regeneration. What remains unclear is whether cells in regenerating systems possess an increased propensity to proliferate and are refractory to signals that induce senescence. My thesis work has focused on how fibroblasts from the ear pinna differentially regulate healing in highly regenerative mammals (e.g., …
