We investigated alternatives to whole blood for blood feeding of mosquitoes with a focus on improved stability and compatibility with mass rearing programs. In contrast to whole blood, an artificial blood diet of ATP-supplemented plasma was effective in maintaining mosquito populations and was compatible with storage for extended periods refrigerated, frozen, and as a lyophilized powder. The plasma ATP diet supported rearing of both Anopheles and Aedes mosquitoes. It was also effective in rearing Wolbachia-infected Aedes mosquitoes, suggesting compatibility with vector control efforts.

Multiple Functions Of Creb-Binding Protein During Postembryonic Development: Identification Of Target Genes, Amit Roy, Smitha George, Subba Reddy Palli
Entomology Faculty Publications
Background: Juvenile hormones (JH) and ecdysteroids control postembryonic development in insects. They serve as valuable targets for pest management. Hence, understanding the molecular mechanisms of their action is of crucial importance. CREB-binding protein (CBP) is a universal transcriptional co-regulator. It controls the expression of several genes including those from hormone signaling pathways through co-activation of many transcription factors. However, the role of CBP during postembryonic development in insects is not well understood. Therefore, we have studied the role of CBP in postembryonic development in Tribolium, a model coleopteran insect.
Results: CBP is ubiquitously expressed in the red flour beetle, …

Caterpillars are the soft bodied larvae of lepidopteran insects. They have evolved to occupy an extremely diverse range of natural environments and to locomote in complex three-dimensional structures without articulated joint or hydrostatic control. These animals make excellent bio-inspiration for the field of soft robotics because of their diversity and adaptability. In this paper, we present SquMA Bot, a caterpillar-inspired soft robot. The robot's body is primarily composed of a soft viscoelastic foam, and it is actuated using a motor-tendon system. SquMA Bot is able to mimic the inching gait of a caterpillar and can use its flexible body to …

Top-Down And Bottom-Up Approaches To Understanding The Fate Of The Federally Endangered Schaus’ Swallowtail Butterfly (Heraclides Aristodemus Ponceanus), Jaeson T. Clayborn
FIU Electronic Theses and Dissertations
The federally endangered Schaus’ swallowtail butterfly (Heraclides aristodemus ponceanus) populations have declined precipitously over the years. Despite tremendous efforts to augment the numbers of this butterfly through captive-bred releases and habitat enhancements, it remains imperiled and federally endangered. The dissertation’s objectives were: 1) to restore dry forest habitat in Biscayne National Park (BNP) for Schaus’ swallowtails; 2) to assess host plant (torchwood and wild lime) survivorship and growth in the restoration sites (BNP) and project dry forest habitat loss because of imminent sea level rise (SLR); 3) to quantify ant activity and record predator-prey interactions against Heraclides caterpillars …

Advanced Biotechnology Tools For Invasive Species Management, Invasive Species Advisory Committee
National Invasive Species Council
Increasingly, genetic tools are being used to detect and solve pressing environmental, social, and health-related challenges. It is clear that investments in technology innovation can be game changing, as advances in biotechnology may provide new methods to protect the nation’s resources from the negative impacts of invasive species. The current toolbox of management options is recognizably insufficient to deal with many of the high-impact species that have been introduced. However, “surrendering” to these species is generally not a viable option from ecological, health, economic, socio-cultural, or political perspectives. Cost-efficient solutions to these “grand invasive species challenges” need to be found. …

Enhancing Federal-Tribal Coordination Of Invasive Species, Blaine Parker, Chuck Bargeron, Sean Southey, Lori Buchanan, Miles Falck, Chris Fisher, Joe Maroney, Mervin Wright, Gintas Zavadkas
National Invasive Species Council
Invasive species are defined by the United States government to mean “with regard to a particular ecosystem, a non-native organism whose introduction causes or is likely to cause economic or environmental harm, or harm to human, animal, or plant health” (Executive Order [EO] 13751). The ecosystems to which invasive species are introduced or spread are not delimited by jurisdictional boundaries; they intersect with lands managed by federal, tribal, state, territorial, and county governments, as well as properties under private ownership. For this reason, effective coordination and cooperation across jurisdictions is of paramount importance in the prevention, eradication, and control of …

RNA interference (RNAi) based methods are being developed for pest management. A few products for control of coleopteran pests are expected to be commercialized soon. However, variability in RNAi efficiency among insects is preventing the widespread use of this technology. In this study, we conducted research to identify reasons for variability in RNAi efficiency among thirty-seven (37) insects belonging to five orders. Studies on double-stranded RNA (dsRNA) degradation by dsRNases and processing of labeled dsRNA to siRNA showed that both dsRNA degradation and processing are variable among insects belonging to different orders as well as among different insect species within …

Spatial Variation In Western Corn Rootworm (Coleoptera: Chrysomelidae) Susceptibility To Bacillus Thuringiensis Corn Events In Nebraska, Jordan D. Reinders
Department of Entomology: Dissertations, Theses, and Student Research
The western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte, is an economically important pest species of field corn (Zea mays L.) in the U.S. Corn Belt. Yield losses and control costs exceed greater than $1 billion annually. Over the past 10+ years, growers have adopted transgenic corn hybrids expressing rootworm-active Bacillus thuringiensis (Bt) proteins as a primary WCR management tactic. Field-evolved resistance to Cry3Bb1 and mCry3A proteins expressed in single-trait Bt hybrids has been confirmed in some areas of Nebraska. Growers have used different tactics as needed to prevent or mitigate resistance, resulting in a mosaic of selection pressure placed …
Horticulture For Pollinator Conservation, Carter M. Westerhold
Horticulture For Pollinator Conservation, Carter M. Westerhold
Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research
Pollinators worldwide are declining. Consequently, the agricultural and ecological services these insects provide are in danger of being lost. Land use intensification, via urbanization, has greatly influenced this decline in pollinators. Luckily, through targeted horticultural practices, stable populations of pollinators can be sustained within urban areas. The horticultural practices of planting diverse floral resources and managing pollinator habitat in urban areas can sustain these populations. Two studies were conducted with the intent to identify horticultural knowledge gaps that could be reduced to aid in pollinator conservation efforts. First, a study to compare Nebraska native and non-native ornamental plants was conducted. …

Tephritid pests, such as the Mexican fruit fly, Anastrepha ludens (Loew), represent a major threat to fruit production worldwide. In order to control these pests, sterile insect technique is used to suppress and eradicate wild populations. For this control method to be successful, hundreds of millions of flies must be produced weekly in mass rearing facilities. The large quantity of artificial diet and close proximity of flies at various life stages allows bacteria from family Enterobacteriaceae, Bacillaceae, Pseudomonadaceae, and others to multiply and spread more easily. In this study, bacteria with a possible pathogenic effect were isolated from Mexican fruit …
