Autobiographical memory is central to one's sense of self and continuity from past to present. Despite this, there is little research on the neural correlates underlying individual subjective experience of autobiographical memory and how that is related to brain phenomena (i.e., activity, communication). The purpose of this study was to help minimize this gap. We recruited twenty healthy adult participants, who were asked to generate memory cues (1-3 word descriptions) for locations and objects from their early and recent life. A humor structure comprises two essential stages: the setup and the punch line. The punch line stage is to provide the incongruity resolution that creates amusement in humor. The current article aimed to look into how humor is amusing and how it differs between the sexes. Functional and effective connectivity analyses in cognitive and affective neuroscience have facilitated the implications of humor comprehension, appreciation, and laughter responses. The processing of incongruity-resolution humor revealed effective connectivity from the amygdala to the precuneus (amygdala → precuneus). Higher-order cognitive functions, such as working memory, attention, and decision making, depend strongly on the functional integrity of frontal and parietal cortices. However, the internal workings of the frontoparietal network (FPN) are not well understood. A major contributor to this knowledge gap is our limited understanding of the intrinsic functional organization of the FPN. In order to address this gap, we examine task-dependent reconfigurations of functional connectivity (FC) within the FPN. We analyzed fMRI task-state data from 924 individuals from the Human Connectome Project Young Adult study. Our results show that FC within the FPN is highly stable across time …

Background: Post-traumatic stress disorder (PTSD) is a debilitating disorder defined by the onset of intrusive, avoidant, negative cognitive or affective, and/or hyperarousal symptoms after witnessing or experiencing a traumatic event. Previous voxel-based morphometry studies have provided insight into structural brain alterations associated with PTSD with notable heterogeneity across these studies. Furthermore, how structural alterations may be associated with brain function, as measured by task-free and task-based functional connectivity, remains to be elucidated.

Anxiety-related disorders are some of the most pervasive mental health disorders affecting adult and youth populations. Despite growing evidence of the neurobiology associated with anxiety-related disorders, a consensus on the neurobiological mechanisms of anxiety-related disorders remains to be elucidated. We first provide background literature on the reasoning behind this dissertation in Chapter 1. Traumatic brain injury (TBI) patients are known to have altered functional connectivity (FC), which has cognitive and behavioral significance and bears clinical implications. Previous literature has discovered a hyperconnectivity response to TBI, most notably in the default mode network (DMN). However, the exact pattern of changes in resting FC during the first year of recovery is unknown. We used resting-state functional magnetic resonance imaging to investigate longitudinal connectivity patterns in the DMN of 28 moderate-to-severe TBI patients as compared to 33 demographically matched healthy controls (HC). Resting State Functional Magnetic Resonance Imaging (rs-fMRI) is a technique that is widely used for analyzing brain function using different approaches and methods. This study involves rs-fMRI analysis of Blood Oxygenation Level Dependent (BOLD) signals acquired from Alzheimer’s disease (AD) Patients and Healthy Controls (HC). Each subject in the study had both functional and anatomical images with at least one rs-fMRI scan with their Anatomical (T1) scans. Previous rs-fMRI studies have demonstrated that AD shows differences in Amplitude of Low Frequency (<0.1 Hz) Fluctuations (ALFF), and Regional Homogeneity (ReHo) measures according to HCs.

The function of sleep is a longstanding mystery of the brain. By contrast, the function of resting state networks (RSNs) is one of its most recent mysteries. The relationship between RSNs and neuronal activity has been unclear since RSNs were discovered during the advent of functional magnetic resonance imaging (fMRI). An influential idea in neuroscience is that action observation activates an observer’s sensory-motor system. This idea has recently been extended to motor learning; observing another individual undergoing motor learning can promote sensory-motor plasticity as well as behavioural changes in both the motor and somatosensory domains. While previous research has suggested a role for the motor system in motor learning by observing, this thesis presents a series of experiments testing the hypothesis that the somatosensory system is also involved in motor learning by observing.

Mesial temporal lobe epilepsy (TLE) is the most common form of focal epilepsy and is often resistant to medication. Recent studies have noted brain-wide disruptions to several neural networks in so-called “focal” epilepsy, notably TLE, leading to it being recognized as a network disease. We aimed to assess the integrity of functional networks while they were simultaneously activated in an ecologically valid manner, using an actively engaging, richly stimulating audio-visual film clip. The brain changes in many ways in the first year. It is not known which of these changes are most critical for the development of cognitive functions. According to the Interactive Specialization Theory, developments in behaviour result from changes in brain connectivity. We tested this using functional connectivity magnetic resonance imaging (fcMRI) of the motor system. fcMRI was acquired at three and nine months – two time-points between which motor behaviour develops enormously. Cognitive control is an executive process that has been associated with a distributed set of cortical regions. These distributed regions appear to cluster into distinct networks with dissociable functions. In this study, independent component analysis was used as a tool to investigate functional connectivity in event-related fMRI data. Extracted networks of interest were functionally characterized using a hybrid task that independently probed moment-to-moment adjustments in control, and stable task-set maintenance.
