Determination Of The Presence Of Rickettsia Spp. And Borrelia Spp. Carried By Tortoise Ticks From Madagascar, Anna Phan

Ticks were removed from three species of Malagasy tortoises, Astrochelys yniphora, A. radiata, and Pyxis arachnoides (comprising two subspecies P. a. arachnoides and P. a. oblonga), between 2012 and 2015. The ticks were presumed to be from the genus Amblyomma. Ticks were morphologically identified and then checked molecularly to confirm their classification or identify any ticks that could not be morphologically identified. Molecular identification was done via end-point PCR that amplified tick cytochrome oxidase (CO1) and tick 12S rRNA genes. Cases of spotted fever group rickettsiosis are becoming more prevalent in the United States. In Virginia, there are three human-biting ticks which are largely responsible for the spread of rickettsial pathogens and the increase in disease cases. These species include Dermacentor variabilis, Amblyomma americanum, and Amblyomma maculatum; all of which are vectors of rickettsial agents to vertebrate hosts. These species are sympatric as adults and have the potential to share large and small mammal hosts. Increasing incidence of many tick-borne diseases have been linked to recent expansions of tick species distributions. Many tick species are expanding their ranges because of anthropogenic changes in the landscape, shifting climatic variables, and increasing populations of suitable host species and tick habitat. Few empirical studies have been performed, however, investigating the ecological mechanisms underlying these range expansions. Ticks are parasitic organisms that disperse across landscape by hitchhiking on hosts, but must then survive in the environment for long periods of time between bloodmeals. Vitellogenesis In Hyalomma Dromedarii (Acari: Ixodidae): A Model For Analysis Of Endocrine Regulation In Ixodid Ticks, Martin E. Schriefer

The egg yolk proteins, vitellins, and their hemolymph precursors, vitellogenins, were characterized in the ixodid tick Hyalomma dromedarii. Three vitellins were identified. The high molecular weight vitellins, VN A and VN B, were composed of seven homologous molecular weight subunit polypeptides, VN 1-7. VN A appears to be a dimeric form of VN B. VN C was composed of single polypeptide. Low levels of vitellogenin, were identified in western immunoblots utilizing antibody raised against purified vitellin. Adult protein and polypeptide profiles, including vitellin and vitellogenin, were characterized as a function of sex, tissue and reproductive development. Quantitative assessment of the …

Cholesterol oleate was demonstrated to be the cuticular contact sex pheromone of Dermacentor variabilis. This pheromone which has been termed the Mounting Sex Pheromone (MSP) was also demonstrated to be present on the surface of D. andersoni, Amblyomma maculatum and A. americanum. This contact sex pheromone enables males excited and attracted by 2,6-dichlorophenol to identify the female as a potential mating partner. The MSP is the second in the series of three sex pheromones guiding the hierarchy of behavioral responses which constitute tick courtship behavior.
