The number of daily bacterial infections is climbing and the CDC explains that this is due to the antibiotic-resistant threat in the United States. Finding a faster way of bacterial identification is necessary as it currently takes 1-4 days for a medical lab to culture and identify bacteria. Photoacoustic flow cytometry (PAFC) can be used as an alternative method resulting in swift identification within an hour (Edgar, 2019). Pseudomonas aeruginosa, cell line PA01, will be coated in up to a few hundred red dyed phages making it detectible by the photoacoustic flow cytometry system. Currently, 64.8% and 52.2% of Little Buffalo Creek and Sarver Run stream kilometers are impaired, respectively. Impairment sources to Little Buffalo Creek include agriculture, riparian deforestation, on-site wastewater, and urban runoff. Impairment sources to Sarver Run are unknown. Our goal was to understand sources of impairment through water quality analysis and fish surveys. Water samples were collected monthly and analyzed for cation and anion concentrations. Mass ratio analysis comparing Br/SO4 and Mg/Li, Ca/Mg and Ca/Sr, Mg/Na and SO4/Cl, and SO4/Cl and Mg/Li suggests that mine drainage and conventional oil and gas may be impacting water …

Characterizing a species’ dietary composition presents an avenue to understand many facets of its ecological niche and can provide essential information for the species’ long-term conservation. To date, the vast majority of diet studies have relied on direct identification of prey during foraging observations or from diet samples to characterize the dietary habits of birds. Streptomycetes are Gram-positive, soil-dwelling bacteria that possess a complex life cycle with the alternation of vegetative mycelium, aerial mycelium, and spores. Streptomyces. coelicolor spore maturation is a complex process that involves spore shape metamorphosis from cylindrical pre-spores into ellipsoid spores, but the details of this process have remained enigmatic. Previously, our lab identified a novel gene ssdA that might play a role in spore shape determination using a transposon-based insertion mutagenesis in S. coelicolor. With advancements in the DNA technology and methodology, more complete DNA profiles can be obtained. Extraction procedures and their reagents have been optimized for maximal DNA yield, but the cotton swab used and provided for sexual assault examinations has not advanced. Despite research suggesting the cotton swab’s absorbent nature and its inclination to retain cellular material, the field of forensic nursing has not yet implemented another swab.

Abandoned mine drainage (AMD) has resulted in worldwide devastation. Passive remediation systems (PRS) are a cost-effective option to treat AMD. While the geochemistry (abiotic) of these systems is well understood, little work has been done to characterize the microbial communities (biotic) that naturally form within these systems. The goal of this dissertation was to identify the effects that bacterial communities have on the efficiency of PRSs. In the US, over 1.1 billion pounds of pesticides are applied annually. Unfortunately, pesticides contaminate natural habitats affecting animal health. Chlorpyrifos (CPF) is an organophosphorous insecticide that contaminates surface waters. Few studies have assessed the impacts of CPF at low, ecologically realistic doses. I aimed to test the effects of low dose CPF exposures on biological parameters including body morphology, hormone levels, and brain development in Northern Leopard Frogs (Lithobates pipiens), a common vertebrate model. Celecoxib nanoemulsion (CXB-NE) has been developed as a macrophage targeted analgesics by Dr. Janjic and her team at Duquesne University, (Janjic et al, 2018; Liu et al, 2020; Saleem et al, 2019b; Vasudeva et al, 2014). The CXB-NE nanoemulsion carrying a Nonsteroidal Anti-inflammatory (NSAID) inhibitor of COX-2 activity result in a reduction in PGE2 expression in macrophages. Using CXB-NE in rats that have peripheral nerve injury constricting the sciatic nerve relieves hypersensitivity, a pain-like behavior. Bobcats (Lynx rufus) are an ecologically and genetically diverse species with a large contiguous range throughout North America. The species not only has a wide array of phenotypic variation compared to other mammals, but shows marked adaptability across ecozones with differing ecological influences. It is these various selective pressures in distinctive parts of the continent that have likely led to localized adaptations within the bobcat metapopulations. Populations of feral cats (Felis catus) have drawn public attention due to a variety of reasons, including the increasing population size and decreasing native fauna populations. Several communities have implemented the Trap-Neuter-Return (TNR) method, developed in Rome. This study examined whether cats that live in a certain social environment (colony cats) are more likely to be related than cats of the general population (non-colony cats). The degree of relatedness was to be determined separately for the populations. The alternative hypothesis for this study is that colony cats are more related than non-colony cats.

Poison hemlock (Conium maculatum), the infamous plant responsible for the death of Socrates, is toxic to humans and animals. In cases where hemlock led to death, the cause of death is usually associated with respiratory arrest, muscle paralysis or complications of acute renal failure. While the entire plant is toxic, the most dangerous part is the seeds, which contain the highest alkaloid concentration. It contains eight piperidine alkaloids, with the most potent one being coniine. Coniine is a neurotoxin that disrupts the function of the peripheral nervous system, causing respiratory paralysis. The protein component of human semen largely consists of a group of proteins called the semenogelins. After ejaculation, they undergo protein-protein interactions to form the coagulation of seminal fluid for protection of spermatozoa prior to fertilization. Because of their abundance, the semenogelin proteins are an effective marker to detect human semen at crime scenes, even after substantial degradation. Neuropathic pain caused by peripheral nerve injury (PNI) leads to the activation and infiltration of microglial cells and to a neuroinflammatory-induced pain state. miRNAs and autophagy are two main factors and/or mechanisms which have the ability to alter the pain state. In this study, miR-195 was shown to be markedly increased after PNI and associated with the pain phenotype. In addition, inhibition of autophagy in vivo led to p62 accumulation, decreased production of LC3, and inhibition of ATG14.

The Hsp70 system is an essential component of chaperone activity in many organisms. Hsp70 functions include: protein folding, aggregation prevention, trafficking, and enzyme regulation. Hsp70’s ability to bind such a vast array of substrates suggests wide range of conformational plasticity. Autophagy is a potent intracellular degradation system and thus its activation requires exquisite regulation to maintain cellular homeostasis. Atg1, a serine-threonine protein kinase, is essential in both selective and non-selective autophagy. New findings suggest that in selective autophagy, Atg1 is activated at the vacuole by convergence of two independent recruitment pathways to prevent aberrant autophagy induction.

The 26S proteasome complex of the malaria parasite Plasmodium falciparum, which was previously unknown, was characterized by the authors through an affinity purification protocol that isolated functional 26S proteasome complexes. This allowed for the identification of subunit composition and PfUSP14, a proteasomeassociated deubiquitinase. Elucidating the diet of Neotropical migratory birds is essential to our understanding of their ecology and to their long-term conservation. Beyond broad taxonomic or morphological categories, however, the diet of Neotropical migrants is poorly documented. Using the molecular techniques of DNA barcoding and next-generation sequencing, we elucidated the diet of Neotropical migratory songbirds breeding in the riparian zones of headwater Appalachian streams. This approach resulted in a genus- or species-level description of diets that improved the current understanding of how songbirds utilize aquatic prey resources in riparian habitats. Humans (Homo sapiens), chimpanzees (Pan troglodytes), and gorillas (Gorilla gorilla) have diverse mating systems with varying levels of sperm competition. Several seminal plasma genes have been claimed to evolve under positive selection, while others are altered or lost. This study aims to identify biologically relevant differences among seminal plasma proteomes of primates in relation to mating systems and previous genomic studies. Seminal plasma from three individuals of each species were run in triplicate in shotgun liquid chromatography – tandem mass spectrometry (LC-MS/MS) and confirmed with Western blots. Millions of people in the United States are afflicted with chronic bladder pain syndromes including interstitial cystitis/bladder pain syndrome (IC/BPS) and chronic prostatitis and chronic pelvic pain syndrome (CP/CPPS). IC/BPS and CP/CPPS patients suffer from generalized pelvic pain, frequent urination with a decrease in urine volume, a constant urge to urinate, and psychological comorbidities in the absence of infection. The etiology is undetermined and treatments are limited. To better understand the mechanisms of IC/BPS and CP/CPPS, many animal models have been developed. Chronic neuropathic pain is a serious, worldwide health problem leading to life-long treatment and the possibility of significant disability. In this study, neuropathic pain was modeled using the chronic constriction injury (CCI) in rats. The CCI rats exhibited hypersensitivity (typical neuropathic pain symptom) to mechanical stimulation of the affected paw 11 days post-surgery, at a time when sham surgery animals did not.
