The purpose of this research is to improve understanding of the neurodevelopmental effects of embryonic exposure to elevated inflammation and oxidative stress induced by the antipyretic drug acetaminophen (APAP). Our study was the first to examine the interactive effects of APAP and inflammation in zebrafish embryos and how the treatments affect brain development and larval behavior. Experimental groups of zebrafish larvae were exposed to lipopolysaccharide (LPS) to induce inflammation, APAP, or LPS + APAP and larval behavior was analyzed using Ethovision automated behavioral tracking software. Epigenetics is the study of molecular modification of a genome without changing its base pairs. The most studied type of epigenetic mechanism is DNA methylation, which is capable of turning a gene “on” or “off.” Epigenetic potential is the capacity to which an individual can have methylation on its genome. The more CpGs available, the greater the epigenetic potential. Within the past few decades, the presence of fluorine containing
organic molecules has increased significantly. Many of the
current industrial production methods are not cost-effective,
practical, or inherently safe. This work describes a new methodology
for the synthesis of trifluoromethyl ketones. Our new method involves
the use of benzoic acid and trifluoromethyl trimethylsilane (TMSCF3) as starting
materials along with diethylamino sulfur trifluoride (DAST) as a reagent
to obtain moderate to good yields of expected products in a short

Peptide nucleic acids (PNAs) are oligonucleotide analogues in which the sugar-phosphate backbone has been replaced by a pseudopeptide skeleton. Since PNAs use the natural nucleobases (Adenine, Thymine, Cytosine, Uracil, and Guanine) found in either DNA and/or RNA, they are able to hybridize according to Watson-Crick base-pairing to form duplexes. PNA is a promising therapeutic agent because they can function as antigene or antisense chemical agents. To further enhance their utility, we aim to incorporate a photoswitchable moiety using azobenzene. Here, we report the results of the synthesis and purification of a photoswitchable 11 mer PNA along with initial characterization efforts.

Geukensia demissa, the ribbed mussel, is a keystone species of Georgia’s coastline that is at risk of experiencing detrimental thermal stress due to climate change. G. demissa interacts positively with a species of salt marsh cordgrass, Spartina alterniflora. Mussels form aggregates beneath salt marsh cordgrass stems where they are shaded and less exposed to sun rays. However, some mussels end up in areas which lack cordgrass and are directly exposed to sun rays. Due to the accumulation of polymers in the environment, biodegradable alternatives should be used in place of commonly used polymers like poly(carboxylates). Poly(carboxylates) are water-soluble polymers (WSPs) that make up a variety of consumer products, such as detergents, descaling agents, and superabsorbent materials commonly found in diapers and feminine hygiene products. Epigenetic buffering, as an environmentally induced increase in variance of epigenetic states that increases phenotypic variation to buffer populations against decreased fitness, may be a factor that resolves the genetic paradox of introduced species. DNA methylation is a molecular mechanism that could facilitate epigenetic buffering by changing in response to environmental stress. Therefore, epigenetic buffering can be detected through increased variance in DNA methylation in novel or heterogeneous environments. Introduced house sparrows (Passer domesticus) have well-documented phenotypic changes with low genetic diversity, high epigenetic diversity, and high variance in DNA methylation that provide a characteristic signature of epigenetic buffering. …

Because initial entry of these infectious agents occurs while blood feeding, the immune response in the flea gut is considered to be the first line of defense against invading microbes. The intestinal microbiome of mammals plays a significant role in host health and response to environmental stimuli and can include both beneficial native bacteria as well as parasitic worms. In this study, I examined the intestinal cestode and bacterial communities of two closely related species of shrew, Sorex monticola and Sorex cinereus, over a six month period in 2016. Specimens were collected approximately every three weeks from May to October from the Sangre de Cristo Mountains in Cowles, New Mexico. However, the amount of carbon sequestered by wetlands is shrinking as droughts and human disturbance increases. Gold (I) coordinated N-heterocyclic carbenes containing appended redox cycling moieties were designed and analyzed for their ability to target the antioxidant network via multiple mechanisms in various human cancer cell models. Complexes containing either ferrocene or naphthoquinone redox cycling moieties were screened for inhibition of cell growth, inhibition of thioredoxin reductase (TrxR), and accentuation of exogenous ROS. The cell death pathway employed was determined by flow cytometry via detection of Annexin-V FITC. Furthermore, differential gene expression between treated cells and untreated cells revealed the induction of endoplasmic reticulum stress and unfolded protein response pathways.

In this experiment, the contribution of a standard production method for biofuels on their oxidative stability was investigated. Peroxide values were measured at different steps of the production process of Brassica carinata and peanut-based biofuels. The washing and drying steps in this production method showed significant increases in peroxide values for both biofuels and was identified as a major contributor of biofuel oxidation. Further analyses of the physical and thermal properties showed a more pronounced affect in the biofuel from Bassica carinata, and indicated an unusual composition much higher in saturated fatty acids much longer than those found in peanut. …

The barnacle Megabalanus coccopoma is a recent invader of the southeastern U.S.A. from the tropical eastern Pacific. In Georgia, M. coccopoma populations along the immediate coastline often suffer extensive mortality during the winter, but population rebuilding is common after these events suggesting that there may be nearby larval sources. I investigated the hypothesis that artificial structures (i.e., buoys, towers), occurring far enough offshore of Georgia for water temperatures to be moderated by the Gulf Stream, provide refuges for breeding adults of M. coccopoma and can serve as the larval source. Dermacentor is a recently evolved genus of hard ticks (Family Ixodiae) that includes 36 known species worldwide. Despite the importance of Dermacentor species as vectors of human and animal disease, the systematics of the genus remain largely unresolved. This study focuses on phylogenetic relationships of the eight North American Nearctic Dermacentor species: D. albipictus, D. variabilis, D. occidentalis, D. halli, D. parumapertus, D. hunteri, and D. andersoni, and the recently re-established species D. kamshadalus, as well as two of the Neotropical Dermacentor species D. nitens and D. dissimilis (both formerly Anocentor). Chemosensory behaviors are used by many mammalian species to assess chemical signals in the environment. These chemical signals may contain important information about reproductive state, identity, status, or location of conspecifics. Elephants are a long-lived species and males reproduce at a much later age than females, which provides a protracted developmental period for males. This study examined chemosensory behaviors in a population of African elephants living in Addo Elephant National Park, South Africa and demonstrated that patterns emerge as elephants develop and approach sexual maturity. Spontaneous alternating behavior (SAB) describes the tendency of an organism to spontaneously select the unfamiliar direction in a two choice situation. Paramecium is the only microscopic genus in which SAB has been studied. The two earlier studies regarding SAB in Paramecium have come to conclusive, but diametrically opposed results. Designing a single new experiment that incorporates the critical differences in the designs of both studies may help to clarify the results from these earlier studies, and provide an excellent opportunity to better understand the factors that influence SAB.

Spiroplasma bacteria (Mollicutes: Spiroplasmataceae) are characterized by motility, helical morphology and are most frequently found in insect guts and phloem tubes of plants. Traditionally, Spiroplasma have been classified by serology. Recent work has generated 16S rDNA sequences that generally correlate with the serological findings. Although the serology and the 16S rDNA sequence analysis clearly classifies strains to the group level, they do not distinguish between strains within the same group. The goal of this project was to investigate the utility of the 16S-23S rDNA intergenic spacer region sequence as a means to distinguish these closely related strains.

Female Maxwell's duikers (Cephalophus maxwellii) were studied at the Wildlife Survival Center on St. Catherine's Island, Georgia. The objectives were to quantify the effects of housing on activity budgets and progesterone cycling. Four animals were studied across two housing situations: housed individually versus housed in a grouped setting. Behavioral observations revealed no significant change in access to resources, rumination, or repetitive behaviors, once animals were housed together. Fecal samples were collected in order to determine levels of progesterone through radioimmunoassay (RIA). Fluctuations in progesterone levels revealed no clear estrous cycles. Spiroplasma bacteria isolates are classified or identified through a series of serological tests that normally consists of screening, one-way deformation tests, cloning, antisera production, and reciprocal deformation tests. Serological tests on the spiroplasmas are followed by molecular analysis. The standard molecular analysis used for spiroplasmas has been 16S rRNA sequencing. Despite the astounding diversity of flowering plants and of mobile organisms that serve as pollinators, sympatric co-flowering plants can potentially share pollinators with each other. This places two selection pressures on plants. First, plants compete for pollinators to visit their flowers and secondly, entice pollinators to be faithful (specialized) and visit only other members of their species.
