Phototaxis and related visual behaviors can vary between species, and thus members of the genus Drosophila make an excellent study system to examine the evolution of vision. While some existing research points to these phototactic behaviors arising due to mating requirements or due to their species-specific ecology or environmental factors, there exists a lack of understanding as to why striking behavioral differences can exist between closely related species, or between members belonging to the same genus. Across phylogeny, integration of external factors, memory, and internal states of the organism dictate organismal behavior and mechanisms. The underlying genetic components can affect these behaviors such as in genomic changes arising from speciation. In this thesis, a new place preference assay was evaluated in the analysis and investigation of two species of Drosophila flies (D. melanogaster and D. simulans) to measure similarities and differences and their attraction to two different food substrates. With over 150,000 described species, flies constitute one of the most species-rich groups of animals on earth, and have managed to colonize almost every corner of it. Part of their success comes from their amazing flying skills, which are strongly tied to their visual capabilities. To navigate fast and accurately through their habitats, they must be able to process the inordinate amounts of visual information necessary to sort obstacles, avoid predators and remain on course. Species can arise as a result of reproductive barriers that prevent gene flow between diverging populations that force them to remain isolated from one another. Behavioural isolation is one of the earliest acting reproductive barriers determined by the evolution of mating preferences that prevent inter-specific matings. Several traits have been identified that contribute to behavioural isolation, but the genetic basis of interspecific female preference is yet to be determined. Cuticular hydrocarbons (CHCs) are produced by insects and primarily used to prevent desiccation. In Drosophila, certain compounds have secondary roles as infochemicals that may act during courtship to influence mate choice. Certain CHCs may stimulate courtship with heterospecifics or act to repel conspecifics. The CHC profile produced by an individual is the result of the interaction between its genetic background and the environment, though the genes that underlie species differences in CHC production and how the environment can modulate the abundance of individual compounds within a species is not well known.
